        Characterization of a New Bacteriocin, Carocin D, from Pectobacterium carotovorum subsp. carotovorum Pcc21

        <B>ABSTRACT</B><P>Two different bacteriocins, carotovoricin and carocin S1, had been found in <I>Pectobacterium carotovorum</I> subsp. <I>carotovorum</I>, which causes soft-rot disease in diverse plants. Previously, we reported that the particular strain Pcc21, producing only one high-molecular-weight bacteriocin, carried a new antibacterial activity against the indicator strain Pcc3. Here, we report that this new antibacterial activity is due to a new bacteriocin produced by strain Pcc21 and named carocin D. Carocin D is encoded by the <I>caro</I>DK gene located in the genomic DNA together with the <I>caro</I>DI gene, which seems to encode an immunity protein. N-terminal amino acid sequences of purified carocin D were determined by Edman degradation. In comparison with the primary translation product of <I>caro</I>DK, it was found that 8 amino acids are missing at the N terminus. This finding proved that carocin D is synthesized as a precursor peptide and that 8 amino acids are removed from its N terminus during maturation. Carocin D has two putative translocation domains; the N-terminal and C-terminal domains are homologous to those of <I>Escherichia coli</I> colicin E3 and <I>Pseudomonas aeruginosa</I> S-type pyocin, respectively. When <I>caro</I>DK and <I>caro</I>DI genes were transformed into carocin D-sensitive bacteria such as Pcc3, the bacteria became resistant to this bacteriocin. Carocin D has one putative DNase domain at the extreme C terminus and showed DNase activity <I>in vitro</I>. This bacteriocin had slight tolerance to heat but not to proteases. The <I>caro</I>DK gene was present in only 5 of 54 strains of <I>P. carotovorum</I> subsp. <I>carotovorum</I>. These results indicate that carocin D is a third bacteriocin found in <I>P. carotovorum</I> subsp. <I>carotovorum</I>, and this bacteriocin can be readily expressed in carocin D-sensitive nonpathogenic bacteria, which may have high potential as a biological control agent in the field.</P>

        Genus-Specific Distribution and Pathovar-Specific Variation of the Glycinecin R Gene Homologs in Xanthomonas genomes

        Xanthomonas axonopodis pv. glycines produces bacteriocins called glycinecin, and two glycinecin genes, glyA and glyR, were reported previously. In this paper, we describe genomic distribution and variation of the glyR gene revealed by extensive Southern hybridization analysis. In contrast to the glyA gene present only in X. axonopodis pv. glycines, the glyR gene was found to be distributed widely in all the pathovars of Xanthomas genus. It was also found that the glyR gene is a multigene family while the glyA is a single copy gene. Moreover, the copy number and the variation of the glyR multigene are unique to each pathovar of Xanthomonas. The uniqueness can be easily detected by the patterns resulted from Southern hybridization using the genomic digests. Thus, we suggest the glyR gene can serve as a useful genus-specific and pathovarspecific DNA marker for Xanthomonas. One of the glyR homologs was further isolated from X. axonopodis pv. glycines, and analyzed to be functional with strong inhibitory activity against several members of Xanthomonas.

        영상정보를 이용한 HMD용 실시간 아이트랙커 시스템

        본 논문은 영상정보를 이용하여 사용자의 눈의 움직임을 통해 응시점을 추적하는 실시간 아이트랙커 시스템 개발에 대한 연구이다. 개발된 시스템은 광학기반의 동공추적 기법을 이용하여 사용자의 눈의 움직임을 추적한다. 광학기반의 방법은 사용자의 눈에 아무런 장애도 일으키지 않고 눈의 위치를 매우 정확하게 측정 할 수 있다는 장점을 가진다. 동공영상을 획득하기 위해 적외선 카메라를 사용하며, 획득한 영상으로부터 정확한 동공영역을 추출하기 위해 적외선 LED를 사용한다. 실시간 영상처리가 가능하게 하기위해 칼만필터를 적용한 동공추적 알고리즘을 개발하고 DSP(Digital Signal Processing) 시스템을 사용하여 동공영상을 획득한다. 실시간 아이트랙커 시스템을 통하여 실시간으로 사용자의 동공움직임을 추적하고 사용자가 바라보는 배경영상에 사용자의 응시점을 나타낸다. In this paper, development and tests of a real-time eye-tracker system are discussed. The tracker system tracks a user′s gaze point through movement of eyes by means of vision-based pupil detection. The vision-based method has an advantage of detecting the exact positions of user′s eyes. An infrared camera and a LED are used to acquire a user's pupil image and to extract pupil region, which was hard to extract with software only, from the obtained image, respectively. We develop a pupil-tracking algorithm with Kalman filter and grab the pupil images by using DSP(Digital Signal Processing) system for real-time image processing technique. The real-time eye-tracker system tracks the movements of user′s pupils to project their gaze point onto a background image.

      • Molecular networks underlying ovarian aging for clinical applications

        Due to modern trends with postponing child-bearing and getting worse living environment in women, an ovarian aging increased pregnancy failure and other complications with menopause or premature ovarian failure. Although several theories have been suggested such as mitochondrial malfunction, DNA damage/repair/methylation, caloric restriction, studies regarding ovarian aging-related molecular mechanisms for development of therapeutic methods are insufficient so far. Our objective is to determine molecular pathways of ovarian aging that result in pregnancy failure and other complications in women health to develop treatment strategies. This study is consisted of two parts: in Phase I stage, we analyzed distinct gene expression profile between young and aged mouse ovaries, and in Phase II stage several preferentially expressed genes in both ovaries were selected and analyzed their physiological functions and involved molecular networks related to ovarian aging for development of diagnostic markers and therapeutic methods. Ovaries from 10 week and 11 month-old FVB/NJ female mice with synchronized estrus cycle were collected for this study. A half of each ovary was used for RNA preparation and the other half for histological analysis. Using the Illumina HiSeq 2000 System, preferentially expressed genes were identified. Functional annotation database-based gene-set enrichment analyses and Pathway Studio® were employed to evaluate aging-related molecular networks. These findings were confirmed through qRT-PCR and immunohistochemistry. To validate RNA-Seq data, we examined expression patterns of marker genes (Amh, Bmp15 and Nobox) that were wellknown to be decreased in ovarian aging process. In young or aged ovary, preferentially expressed 876 genes were identified and extracellular matrix (ECM; p<0.001) and chromatin/nucleosome-related (p<0.001) protein-coded genes have the majority in these genes by GOTERM analysis. Amongthem, we selected several candidate genes and confirmed their expression profiles by qRT-PCR and immunohistochemistry followed by molecular network analysis. Regarding molecular interactions in these genes, PathwayStudio® was employed to predict aging-involved molecular networks in mouse ovary. Here we report a couple of candidate molecular networks and medicines (chemicals) for targeting these preferentially expressed genes/proteins. Further analyses are scheduled to produce transgenic animal models and with human ovarian tissues/cell lines.

        강의안 작성에 대한 예비 한국어 교원들의 인식 연구 -한국어교육실습 수강생들을 대상으로-

        The purpose of this study is to analyze the difficulties that prospective Korean language teachers taking Korean language teaching practice experience when writing lesson plans. To this end, in this study a survey was conducted on students who took Korean language teaching practice at a Cyber University in Seoul to examine the perceptions of prospective Korean language teachers on the preparation of the lesson plan. As a result of the survey analysis, prospective teachers found it difficult to prepare the use and warm-up stages in lesson construction, and perceived that the follow-up and presentation stages, which were relatively formal, were easier than other stages. Based on the responses of prospective teachers and the feedback received from their supervisors, it was found that prospective teachers had difficulty understanding the characteristics of each lesson stage, organizing the contents of each lesson stage, and using teacher talks according to the learner’s level. In addition, it was found that some difficulties could be resolved through observing Korean classes. Also, it was confirmed that practicing repeatedly for being aware of the lesson plan as well as writing the lesson plan plays a major role in successfully conducting the mock class. Based on these analysis results, this study suggested how to use teacher talks and prepare each lesson stage related to the preparation of lesson plans for prospective Korean language teachers.

        온라인 강의 참관을 통한 한국어교육 전공생들의 현장 경험 인식 연구

        본 연구에서는 ‘COVID-19’로 인해 대면 강의 참관이 물리적으로 어려운 상황에서 한국어교육 전공자들이 온라인 동영상 강의 참관을 통해서 강의 참관의 목표를 달성할 수 있는지 살펴보고자 하였다. 이를 위해 본 연구에서는 서울 소재 사이버대학에서 한국어교육 실습 교과목 내 온라인 동영상 강의 참관을 통해 강의 참관을 한 수강생들을 대상으로 설문조사를 실시하여 온라인 동영상 강의 참관에 대한 한국어교육 전공자들의 인식을 살펴보았다. 설문조사 분석 결과, 수강생들은 동영상 참관 프로그램에 대해 비교적 긍정적인 인식을 가지고 있음을 알 수 있었다. 또한, 강의 참관 만족도는 수업 몰입도와 프로그램의 질과 강한 상관관계를 보였고, 프로그램의 질과 수업 몰입도, 인터페이스와 프로그램의 질 사이에서도 중간 강도의 상관관계를 보였다. 강의 참관 만족도에 어떤 요소가 영향을 끼치는지를 확인하기 위해 실시한 회귀분석 결과를 살펴보면 수업 몰입도와 프로그램의 질이 참관 수업 만족도에 유의미한 영향을 끼치고 있었다. 본고에서는 온라인으로 실시된 동영상 강의 참관에 대한 학습자의 인식을 조사하고, 이를 바탕으로 동영상 강의 참관의 개선점에 대하여 논하였다. 또, 각 요소들의 관계를 조사함으로써 강의 참관 활용과 개선을 위한 활용점을 찾고자 하였다. In this study, we tried to examine whether Korean language education majors can achieve the goal of class observation by observing VOD of face-to-face classes in a situation where it is physically difficult to observe face-to-face lectures due to 'COVID-19'. To this end, in this study, a survey was conducted on students who observed VOD in Korean language education practice courses at Cyber University in Seoul to examine the perceptions of Korean language education majors about observing VOD of face-to-face classes. As a result of the survey analysis, it was found that the students had a relatively positive perception of the online VOD observation. In addition, the observed class satisfaction showed a strong correlation with class immersion and program quality, and showed a moderate correlation between program quality and class immersion, and interface and program quality. The results of the regression analysis conducted to check which factors affect the satisfaction of the class observation program showed that the class immersion and the quality of the program had a significant effect on the satisfaction of the program. In this paper, we investigated learners' perceptions of online VOD observations, and based on this, we discussed the improvement points of the program. In addition, by examining the relationship between each element, it was attempted to find useful points for the utilization and improvement of the class observation.

        Influence of Trust and Delivery System on Customer Satisfaction and Company Performance on Social Commerce Sites

        Many consumers are skeptical or suspicious of the functional mechanisms of social commerce, its valuable processes and fast delivery, and the quality of many products that are offered online. This paper discusses the role of consumer trust as a foundation for the diffusion and acceptance of social commerce. First, trust is seen as a distinct but potentially coexisting mechanism for reducing the uncertainty and complexity of transactions in social commerce markets. Second, this paper provides actual proof of whether or not quality of service can be attributed to customer satisfaction and company performance on social commerce sites.
