전계효과 트랜지스터(FETs)를 이용한 전하 검출형 DNA 센서에서 Debye length에 따른 검출 감도

전계효과 트랜지스터(FETs)를 이용한 전하 검출형 DNA센서는 DNA가 가지고 있는 음전하를 중성화 시키는 양이온의 영향은 매우 중요하다. 본 논문에서는 양이온 농도에 의존하는 Debye length에 관한 연구를 통해 DNA 검출감도를 평가하였다. Debye length는 낮은 농도의 NaCl 용액에서 긴 거리를 유지하며, Debye length가 높은 용액에서 DNA가 가지고 있은 음전하는 게이트 채널에 보다 많은 영향을 미친다. 용액내 NaCl농도가 1 mM인 버퍼 용액에서 상보적 DNA의 hybridization에 의한 전계효과 트랜지스터의 게이전압은 21 ㎷ 시프트 했으며, NaCl 농도가 10 mM인 버퍼 용액에서는 7.2 ㎷, NaCl농도가 100mM인 버퍼 용액에서는 전계효과 트랜지스터의 게이트 전압이 5.1 ㎷ 각각 시프트 하였다. 이러한 결과를 바탕으로 전계효과 트랜지스터를 이용한 전하 검출형 DNA센서의 검출 감도는 Debye length에 의존하는 것을 규명하였다. The effects of cations are very important in field-effect transistors (FETs) type DNA sensors detecting the intrinsic negative charge between single-stranded DNA and double-stranded DNA without labeling, because the intrinsic negative charge of DNA is neutralized by cations in electrolyte solution. We consider the Debye length, which depends on the concentration of cations in solution, to detect DNA hybridization based on the intrinsic negative charge of DNA. The Debye length is longer in buffer solution with a lower concentration of NaCl and the intrinsic negative charge of DNA is more effective on the channel surface in longer Debye length solution. The shifts in the gate voltage by DNA hybridization with complementary target DNA are 21 ㎷ in 1 mM NaCl buffer solution, 7.2 ㎷ in 10 mM NaCl buffer solution, and 5.1 ㎷ in 100 mM NaCl buffer solution. The sensitivity of FETs to detect DNA hybridization based on charge detection without labeling depends on the Debye length.
다이아몬드 FETs에서 전기적 바이어스 방법을 이용한 단일염기 다형성(SNPs) 검출

돌연변이 및 유전병의 원인이 되고 있는 유전자 단일염기 다형성(single nucleotide polymorphisms; SNPs) 검출은 조기진단, 치료 및 제약등 바이오관련 분야에서 매우 중요하다. SNPs 검출을 위한 방법은 다양하게 제시되고 있으나 상보적 DNA와 SNPs의 에너지 차이가 미세하여 SNPs 검출에는 많은 어려움이 존재한다. 본 논문에서는 SNPs를 검출하기 위하여 전하 검출형 전계효과 트랜지스터(field-effect transistors; FETs)를 이용하여 DNA가 가지고 있는 음전하 측정 방법으로 SNPs를 검출하였다. 상보적 DNA와 SNPs의 미세한 에너지 차이를 구분하기 위하여 타게트 DNA hybridization공정에서 드레인-소스 전극에 -0.3 V의 음전압을 인가하였다. 음전압 인가에 따라 DNA 자체 음전하와 센서 표면의 음전압의 전기적 반발력에 의해 센서에 검출되는 타게트 DNA hybridization 신호 크기는 감소하였으나 상보적 DNA와 SNPs의 신호 차는 1.7 ㎷에서 8.7 ㎷로 5배 이상 증가하여 검출되었다. The detection of single nucleotide polymorphisms (SNPs) caused of mutant or genetic diseases is important to diagnosis and medicine. There are many methods have been proposed to detect SNPs. However the detection of SNPs is difficulty, because the difference of energy between complementary DNA (cDMA) and SNPs is very small. In this work, we detect the SNPs using field-effect transistors (FETs) which based on the detection of negative charge of DNA. We bias ?0.3 V on the drain-source electrode at the target DNA hybridization process. The efficiency of hybridization and the amplitude of signal decrease by repulsive force between negative charge of DNA and negative bias on the electrode. However, the sensitivity of SNPs increases about 5 times from 1.7 ㎷ to 8.7 ㎷.

최근 환경, 의료분야에서 실시가 검출 및 인체 삽입형 pH 센서에 대한 요구가 증가하고 있다. 이에 본 연구에서는 생체적합성이 우수한 그래핀을 이용하여 실시간 pH 검출이 가능한 센서를 개발하였다. Polyethylene terephthalate(PET) 기판에 전사된 그래핀 표면에 이온 용액속에서 동작하는 전계효과 트랜지스터(solution-gated field-effect transistors; SGFETs)를 제작하였으며 이를 이용하여 이온 용액의 pH를 검출하였다. 제작한 트랜지스터의 게이트 채널 길이는 500 ㎛, 게이트 채널 폭은 8 ㎜이다. 이온 용액속에서 트랜지스터 동작특성 및 pH 감도를 평가하기 위하여 드레인-소스 전압(VDS)에 따른 드레인-소스 전류(IDS) 및 게이트-소스 전압(VGS)에 따른 드레인-소스 전류(IDS)를 측정하였다. PET기판에 전사된 그래핀 위에 제작한 그래핀 SGFETs의 전류-전압 특성은 이온 용액내에서 매우 안정적으로 동작하였으며 그래핀 SGFETs의 Dirac point는 이온 용액의 pH값이 증가함에 따라 양의 방향으로 19.32 ㎷/pH씩 증가하였다. Recently, the detection of pH with real-time and in vivo has been focal pointed in the environmental or medical fields. In this work, we developed the pH sensor using graphene sheet. Graphene has high biocompatibility. We fabricated flexible solution-gated field-effect transistors (SGFETs) on graphene sheet transferred on the polyethylene terephthalate (PET) substrate to detect pH in electrolyte solution. The gate length was 500 ㎛ and the gate width was 8 ㎜. We evaluated the current-voltage (I-V) transfer characteristics of graphene SGFETs in pH solution. The drain-source current (IDS) and the gate-source voltage (VGS) curves of graphene SGFETs were depended on pH value. The Dirac point of graphene SGFETs linearly shifted to the positive direction about 19.32 ㎷/pH depending on the pH value in electrolyte solution.
Characterization of partially functionalized diamond for detecting single mismatched DNA

Here we report a partially aminated micropattern via direct functionalization and examine eleven different solution-phase probe DNAs hybridizing with the same target DNA on both hydrogen and oxygen terminated diamond. The hybridization intensities determined by epifluorescence microscopy were compared and are influenced strongly by the negatively charged surface and mismatched position of its sequence with immobilized probe DNA.

Polyethylene terephthalate(PET) 기판에 전사된 그래핀 표면에 이온 용액속에서 동작하는 전계 효과 트랜지스터(graphene solution-gated field-effect transistor; G-SGFET)를 제작하였으며 이를 이용하여 배양된 세포 수를 개수하였다. 배양된 세포수를 개수하기 위하여 G-SGFET 게이트 채널 표면에 세포를 도포하고 온도 37℃, 5% CO₂ 조건의 인큐베이터 안에서 G-SGFET의 게이트 채널 표면에 세포 부착을 유도하였다. 세포 부착 여부에 따른 전류-전압 특성 변화를 관찰하기 위해 세포 부착 후 G-SGFET의 게이트-소스 전압(VGS)에 따른 드레인-소스 전류(IDS) 특성과 게이트 채널 표면에 세포 부착 전 IDS-VGS 특성과 비교하였다. 게이트 채널 표면에 세포 부착에 따라 G-SGFET의 Dirac point 전압(VDirac)은 부착 되는 세포 수에 따라 0.29 mV/cell 씩 왼쪽으로 이동하였다. We fabricated solution-gated field-effect transistor on the graphene sheet (G-SGFET) transferred on the polyethylene terephthalate (PET) substrate to detect cell count in electrolyte solution. The cells were dropped on the gate channel surface of G-SGFET, and cell adhesion was induced in an incubator at 37℃ and 5% CO₂ condition. In order to observe the change in current-voltage characteristics according to cell adhesion, the drain-source current (IDS) and gate-source voltage (VGS) of the G-SGFET was measured and compared with the characteristics before cell adhesion to the gate channel surface. The Dirac point voltage (VDirac) of G-SGFET shifted to the left direction (0.29 mV/cell) due to cell adhesion on the gate channel surface.
