Associations Between Infiltrating Lymphocyte Subsets and Hepatocellular Carcinoma

Aims: We aimed to analyze the phenotype of tumor-infiltrating lymphocytes (TILs) and non-tumor infiltrating lymphocytes (NILs) in HCC and non-tumor tissues, and evaluate relationships between changes in these cells and the prognosis of HCC. Methods: Lymphocytes were isolated from HCC and corresponding non-tumor tissues and tested by flow cytometry. For comparison, clinical parameters were analyzed. Results: Compared with the non-tumor tissue, tumor tissue had a lower intensity of NK, NKT andCD8+T cell infiltration. TILs had higher intensity of CD4+CD25+Foxp3+regulatory T cell (Treg cells) infiltration compared with that in NILs. The prevalence of Treg cells was associated with fewer CD8 + T lymphocytes in the HCC immune microenvironment. The frequencies of NK cells and CD8+T cells in TILs of HCC patients with metastasis less than 12 months were lower than those without metastasis. However, the frequency of Treg cells was higher than those without metastasis. Conclusion: These results suggest that the frequencies of CD8+T, NK and NKT cells as well as Treg cells in the tumor tissue of HCC are significantly associated with patient survival, and could be applied as predictive indicators for HCC prognosis.

In a previous study, three larval cuticle protein genes were cloned from the mulberry longicorn beetle, Apriona germari (Comp. Biochem. Physiol. B 136, 803-811, 2003). In the present study, the genomic structures of these three larval cuticle protein genes (AgLCP9.2, AgLCP12.6 and AgLCP12.3) were elucidated. All three cuticle protein genes consist of one intron and two exons. Southern blot analysis of genomic DNA suggested that three cuticle protein genes are a single copy gene. In addition, a novel larval cuticle protein gene, AgLCP10.6, was cloned from A. germari in this study. The AgLCP10.6 cDNA contains an ORF of 300 nucleotides that are capable of encoding a 100-amino acid polypeptide with a predicted molecular mass of 10.6 kDa. The amino acid sequence deduced from the AgLCP10.6 cDNA contained a type-specific consensus sequence identifiable in other insect cuticle proteins and is most homologous to Drosophila melanogaster cuticle protein ACP65A (51 % protein sequence identity). Northern blot analysis revealed that AgLCP10.6 showed epidermis-specific expression.

Purpose: Intranasal dexmedetomidine is an effective sedative for premedication and is regularly used to reduce preoperative tension and anxiety in children. This study aimed to assess the effect of intranasally adjunctive dexmedetomidine on perioperativesedative and analgesic requirements in adults. Materials and Methods: Patients were randomly divided into four groups to receive preoperative administration of saline, intranasaldexmedetomidine 1 μg/kg and 2 μg/kg, and intravenous dexmedetomidine 1 μg/kg, respectively. Propofol and remifentanil were target-controlled infused to maintain intraoperative bispectral index at 45–55 and blood pressure at baseline value±20%. Sufentanil was administered to maintain postoperative visual analogue scale ≤3. Perioperative anesthetics requirements were compared using nonparametric tests. Results: Intranasal dexmedetomidine significantly attenuated propofol requirements for anesthesia induction and maintenance in a dose-dependent manner. Patients given intranasal dexmedetomidine 2 μg/kg required less remifentanil for anesthesia maintenance. The first postoperative request for sufentanil analgesia was delayed in patients given intranasal dexmedetomidine 2 μg/kg. The anesthetics-sparing effect of intranasal dexmedetomidine was significantly weaker than intravenous dexmedetomidine at the same dose of 1 μg/kg. The incidences of adverse events, including hemodynamic instability and delayed recovery, were comparablewith and without intranasal dexmedetomidine. Conclusion: Intranasal administration of dexmedetomidine can reduce perioperative anesthetic requirements, and a dose of dexmedetomidine 2 μg/kg produces a better effect in adults. The anesthetics-sparing effect of intranasal dexmedetomidine 1 μg/kg is less than that with the same intravenous dose of dexmedetomidine.
Wide input/output control strategy for multiphase series capacitor bidirectional DC-DC converters

The series capacitor converter is one kind of high-gain bidirectional DC-DC converter based on interleaved and switched capacitor techniques. For the n-phase series capacitor converter, n times voltage gain when compared with the traditional boost converter under a specific duty cycle, as well as automatic current sharing among phases can be achieved. The specific duty cycle is (n - 1)/n to 1 in the boost mode and 0 to 1/n in the buck mode, which means that it is not suitable for wide input and/or wide output applications. To solve this issue, an 180° interleaved switching strategy is proposed in this paper. The proposed strategy is more suitable for converters to widen the working duty cycle range. The working principle, voltage gain and current distribution/sharing characteristics with the duty cycle ranging from 0 to 1 are analyzed and calculated in greater detail. In addition, as one of the n-phase series capacitor converters, a small signal model of a three-phase series capacitor converter is built, and a dual closed-loop controller is studied and designed. Finally, an experimental prototype with an input voltage that varies from 10 to 30 V and an output voltage of 100 V is built and tested. Results obtained with the prototype verify the correctness of the proposed switching strategy.

Epidermal growth factor receptor (EGFR) is considered to be one of the key driver genes in non-small cell lung cancer (NSCLC). Several clinical trials have shown great promise of EGFR tyrosine kinase inhibitors (TKIs) in the first-line treatment of NSCLC. Many advances have been made in the understanding of EGFR signal transduction network and the interaction between EGFR and tumor microenvironment in mediating cancer survival and development. The concomitant targeted therapy and radiation is a new strategy in the treatment of NSCLC. A number of preclinical studies have demonstrated synergistic anti-tumor activity in the combination of EGFR inhibitors and radiotherapy in vitro and in vivo. In the present review, we discuss the rationale of the combination of EGFR inhibitors and radiotherapy in the treatment of NSCLC.
Influence of processing parameters on warpage according to the Taguchi experiment

An automotive triangle trim was chosen as a research model, and the influence of processing parameters was determined with the Taguchiexperimental method. An L27 (313) orthogonal array was constructed to investigate the significance of each factor (mold temperature,melt temperature, injection time, V/P switchover, packing pressure, and packing time). The computer-aided engineering softwareMoldflow was employed to determine the extent of warpage. The influence of processing parameters was analyzed by obtaining the signal-to-noise ratio and by conducting a range analysis. Furthermore, the processing parameters for minimizing warpage were optimized. Subsequently, the optimization packing condition for warpage minimization based on the filling conditions.

Background and purpose Self-rated health (SRH) is a consistent and strong predictor of all-cause and cardiovascular mortality in various populations. However, the associations between SRH measures and risk of first-ever or recurrent stroke were rarely explored. We thus aim to prospectively investigate the associations between SRH measures and risk of total and subtypes of stroke in Chinese population. Methods A total of 494,113 participants from the China Kadoorie Biobank without prior heart diseases or cancer (486,541 without stroke and 7,572 with stroke) were followed from baseline (2004 to 2008) until December 31, 2013. General and age-comparative SRH were obtained from baseline questionnaires. First-ever stroke or recurrent events were ascertained through linkage to disease registry system and health insurance data. Results We identified 27,662 first-ever stroke and 2,909 recurrent events during an average of 7.0 years of follow-up. Compared with excellent general SRH, the hazard ratios (HRs) and 95% confidence intervals (CIs) for first-ever stroke associated with good, fair, and poor general SRH were 1.04 (1.00 to 1.08), 1.19 (1.15 to 1.23), and 1.49 (1.42 to 1.56) in the multivariate model, respectively. Compared with better age-comparative SRH, the HRs (95% CIs) of same and worse age-comparative SRH were 1.13 (1.10 to 1.17) and 1.51 (1.45 to 1.58), respectively. The relations of SRH measures with ischemic stroke, hemorrhagic stroke, and recurrent stroke were similar to that with total first-ever stroke. However, the magnitude of associations was much stronger for fatal stroke than for non-fatal stroke. Conclusions This large-scale prospective cohort suggests that self-perceived health status is associated with incident stroke, regardless of stroke subtype.

In this work, a novel machine tool thermal error modeling method based on dynamic temperature gradient is proposed, and a thermal error compensation method based on unfalsified control is developed. The dynamic temperature gradient is used to optimize the locations of temperature measuring points on the machine tool. Real-time compensation for the thermal error can be achieved using the developed compensation method by establishing the correlations between dynamic temperature gradient and thermal error in the machine tool. Different from traditional model-based methods, the developed compensation approach does not rely on an accurate model of the thermal error but instead uses online input/output data to adaptively select the best controller at any moment, thereby improving thermal error prediction accuracy and robustness. The effectiveness of the developed thermal error compensation method is demonstrated on a turning center, where the spindle thermal error is compensated during the manufacturing of 120 inner bore parts and 120 shaft parts. After compensation using the proposed approach, thermal errors are reduced from 27 µm to 9 µm for the inner bore parts and from 31 µm to 11 µm for the shaft parts, respectively.
