        Changes of phenolic-acids and vitamin E profiles on germinated rough rice (Oryza sativa L.) treated by high hydrostatic pressure

        This study was performed to investigate changes in the phenolic acid and vitamin E profiles of germinated rough rice following high hydrostatic pressure treatment (HPT). Rough rice was germinated at 37<SUP>o</SUP>C for two days and subjected to 0.1, 10, 30, 50, and 100MPa pressures for 24h. The total phenolic acid content increased from 85.37μg/g at 0.1MPa to 183.52μg/g at 100MPa. The highest gallic acid (4.29μg/g), catechin (9.55μg/g), p-coumaric acid (8.36μg/g), ferulic acid (14.99μg/g), salicylic acid (14.88μg/g), naringin (6.18μg/g), trans-cinnamic acid (45.23μg/g), and kaempferol (40.95μg/g) contents occurred in the sample treated at 100MPa after germination. The maximum vitamin E content of about 2.56 (BG) and 4.34mg/100g (AG) were achieved at 30MPa. These result suggest that a combination of HPT and germination are efficient method for enhancement of functionality in rough rice, and clarify the influence of HPT conditions on the vitamin E and phenolic acid in germination rough rice.

        New Evidence of Alleles (V199I and G52S) at the PRKAG3 (RN) Locus Affecting Pork Meat Quality

        The porcine PRKAG3 (RN) gene encodes the regulatory gamma subunit of adenosine monophosphate-activated protein kinase (AMPK), which is a good candidate gene affecting meat quality. In this study, the effects of two missense mutations A595G (Ile199Val) and G154A (Gly52Ser) in porcine PRKAG3 gene on meat quality traits were studied in M. Longissimus dorsi (LD), M. Semispinalis capitis (SC) and M. Biceps femoris (BF) from different populations of 326 pigs. The PRKAG3 alleles 199I, 199IV, 52S and 52G were identified with PCR-RFLPs and all genotypes - 199I/199I, 199I/199V, 199V/199V, 52S/52S, 52S/52G and 52G/52G - were found. The frequency of V allele was larger than that of I allele in all populations. I allele frequency was zero in Chinese Meishan pigs (population D) especially. G allele frequency was larger than that of S allele in all populations except Large White (population A). Both variations at the PRKAG3 locus significantly affected these meat quality traits. The pork meat quality has not previously been established in Meishan or crosses thereof. The results suggested that generally pH of LD, SC and BF was higher in Meishan pigs than that in other populations. Moreover, Meishan pigs showed higher water-holding capacity and intramuscular fat (IMF), lower water content and water loss percentage compared to other populations in terms of the two variations. The results present here supply new evidence that alleles V199I and G52S at the PRKAG3 locus affect pork meat quality and provide useful information on pork production.

      • Self-adaptive Si/reduced graphene oxide scrolls for high-performance Li-ion battery anodes

        <P>A Si/C composite with Si nanoparticles (nSi) uniformly dispersed in the interlayers of reduced graphene oxide scrolls (rGS) is successfully developed for high-performance Li-ion battery anodes. The rGS can deform reversibly with the repeated expansion/contraction of nSi to maintain contact between the nSi and the conductive rGS network, which can effectively buffer large volume changes and maintain continuous large-area core-shell electrical contact. Additionally, the continuous electrical network of rGS greatly enhances the electrical conductivity, and the open structures at the ends and sides of rGS provide paths for rapid diffusion of Li ions, thus enhancing the rate performance. By virtue of the rational design, the composite shows a high reversible specific capacity of 2030 mA h g(-1) at 0.2 A g(-1), high cycling stability of 1200 mA h g(-1) at 4 A g(-1) with 99.2% capacity retention after 200 cycles, and an excellent rate performance of 1000 mA h g(-1) even at 8 A g(-1).         MINERAL NUTRITION OF GRAZING SHEEP IN NORTHERN CHINA I. MACRO-MINERALS IN PASTURE, FEED SUPPLEMENTS AND SHEEP

        This study determined the macro-mineral levels in plants and sheep, at different times during the year, at three farms in northern China. Samples of plants, animal tissues and faeces were collected at 5 to 8 times during the year from each site. They were analysed for calcium, sodium, phosphorus, magnesium and potassium. Sodium concentrations in plants were below those recommended for optimum animal production at all sites for all or part of the year (0.01-1.66 g/kg DM). Low concentrations of sodium in faeces were measured and signs of sodium deficiency (soil ingestion) were observed on one farm. There were seasonal trends in other mineral levels in plants and animals. Plants were lowest in potassium (2.3-13.4 g/kg DM), magnesium (1.28-4.82 g/kg DM) and phosphorus (0.24-1.62 g/kg DM) in winter and spring. However, high levels of these elements were supplied in the feed supplements used at this time of the year. During the periods of rapid pasture growth, in summer and autumn, supplements of feed and salt are often not provide even though pasture concentrations of phosphorus and sodium are low. It may be at these times that sheep will be most susceptible to deficiencies of these elements.

        Investigation of PCR-RFLPs within Major Histocompatibility Complex B-G Genes Using Two Restriction Enzymes in Eight Breeds of Chinese Indigenous Chickens

        New polymorphism of major histocompatibility complex B-G genes was investigated by amplification and digestion of a 401bp fragment including intron 1 and exon 2 using polymerase chain reaction-restriction fragment length polymorphism (PCRRFLP) technique with two restriction enzymes of Msp I and Tas I in eight breeds of Chinese indigenous chickens and one exotic breed. In the fragment region of the gene, three novel single nucleotide polymorphisms (SNPs) were detected at the two restriction sites. We found the transition of two nucleotides of A294G and T295C occurred at Tas I restriction site, and consequently led to a nonsynonymous substitution of asparagine into serine at position 54 within the deduced amino acid sequence of immunoglobulin variableregion- like domain encoded by the exon 2 of B-G gene. It was observed at rare frequency that a single mutation of A294G occurring at the site, also caused an identical substitution of amino acid, asparagine 54-to-serine, to that we described previously. And the transversion of G319C at Msp I site led to a non-synonymous substitution, glutamine 62-to-histidine. The new alleles and allele frequencies identified by the PCR-RFLP method with the two enzymes were characterized, of which the allele A and B frequencies at Msp I and Tas I loci were given disequilibrium distribution either in the eight Chinese local breeds or in the exotic breed. By comparison, allele A at Msp I locus tended to be dominant, while, the allele B at Tas I locus tended to be dominant in all of the breeds analyzed. In Tibetan chickens, the preliminary association analysis revealed that no significant difference was observed between the different genotypes identified at the Msp I and Tas I loci and the laying performance traits, respectively.

        True Digestibility of Phosphorus in Different Resources of Feed Ingredients in Growing Pigs

        To determine the true digestible phosphorus (TDP) requirement of growing pigs, two experiments were designed with the experimental diets containing five true digestible P levels (0.16%, 0.20%, 0.23%, 0.26% and 0.39%) and the ratio of total calcium to true digestible P (TDP) kept at 2:1. In Experiment 1, five barrows (Duroc${\times}$Landrace${\times}$Yorkshire) with an average initial body weight of 27.9 kg were used in a $5{\times}5$ Latin-square design to evaluate the effect of different dietary P levels on the digestibility and output of P and nitrogen. In Experiment 2, sixty healthy growing pigs (Duroc${\times}$Landrace${\times}$Yorkshire) with an average body weight (BW) of 21.4 kg were assigned randomly to one of the five dietary treatments (12 pigs/diet), and were used to determine the true digestible phosphorus (TDP) requirement of growing pigs on the basis of growth performance and serum biochemical indices. The results indicated that the true digestibility of P increased (p<0.05) linearly with increasing dietary TDP level below 0.26%. The true P digestibility was highest (56.6%) when dietary TDP was 0.34%. Expressed as g/kg dry matter intake (DMI), fecal P output increased (p<0.05) linearly with increasing P input. On the basis of g/kg fecal dry matter (DM), fecal P output was lowest for Diet 4 and highest (p<0.05) for Diet 5. The apparent digestibility of crude protein (CP) did not differ (p>0.05) among the five diets, with the average nitrogen output of 12.14 g/d and nitrogen retention of 66% to 74% (p>0.05), which suggested that there was no interaction between dietary P and CP protein levels. During the 28-d experimental period of Experiment 2, the average daily gain (ADG) of pigs was affected by dietary TDP levels as described by Eq. (1): $y=-809,532x^4+788,079x^3-276,250x^2+42,114x-1,759$; ($R^2=0.99$; p<0.01; y = ADG, g/d; x = dietary TDP, %), F/G for pigs by Eq. (2): $y=3,651.1x^4-3,480.4x^3+1,183.8x^2-172.5x+10.9$ ($R^2=0.99$; p<0.01; y = F/G; x = dietary TDP, %), and Total P concentrations in serum by Eq. (3): $y=-3,311.7x^4+3,342.7x^3-1,224.6x^2+195.6x-8.7$ (R2 = 0.99; p<0.01; y = total serum P concentration and x = dietary TDP, %). The highest ADG (782 g/d), the lowest F/G (1.07) and the highest total serum P concentration (3.1 mmol/L) were obtained when dietary TDP level was 0.34%. Collectively, these results indicate that the optimal TDP requirement of growing pigs is 0.34% of the diet at a total Ca to TDP ratio of 2:1.

      • Cyclic AMP concentrations in dendritic cells induce and regulate Th2 immunity and allergic asthma

        <P><B>Significance</B></P><P>Allergic asthma is characterized by Th2 type inflammation, leading to airway hyperresponsiveness and remodeling. However, the mechanisms by which DC promote Th2 differentiation remain unclear. Herein we demonstrate that low cAMP levels in DC induce Th2-biased responses in vitro and in vivo. Furthermore, mice with conditional deletion of <I>Gnas</I> in DC (<I>Gnas</I><SUP>ΔCD11c</SUP> mice) develop spontaneous bronchial asthma that shares multiple similarities with human asthma. In contrast, increasing cAMP levels inhibit these responses. Thus, regulators of cAMP levels in DC such as G-protein-coupled receptors are non-pattern recognition receptors that play a significant role in CD4 T cell differentiation.</P><P>The inductive role of dendritic cells (DC) in Th2 differentiation has not been fully defined. We addressed this gap in knowledge by focusing on signaling events mediated by the heterotrimeric GTP binding proteins Gαs, and Gαi, which respectively stimulate and inhibit the activation of adenylyl cyclases and the synthesis of cAMP. We show here that deletion of <I>Gnas</I>, the gene that encodes Gαs in mouse CD11c<SUP>+</SUP> cells (<I>Gnas</I><SUP>ΔCD11c</SUP> mice), and the accompanying decrease in cAMP provoke Th2 polarization and yields a prominent allergic phenotype, whereas increases in cAMP inhibit these responses. The effects of cAMP on DC can be demonstrated in vitro and in vivo and are mediated via PKA. Certain gene products made by <I>Gnas</I><SUP>ΔCD11c</SUP> DC affect the Th2 bias. These findings imply that G protein-coupled receptors, the physiological regulators of Gαs and Gαi activation and cAMP formation, act via PKA to regulate Th bias in DC and in turn, Th2-mediated immunopathologies.</P>

        Effect of L-Carnitine and Source of Dietary Fat on Growth Performance and Serum Biochemical Parameters of Piglets Weaned at 35 Days of Age

        The effects of carnitine in diets with or without added fat (5% lard or soybean oil) were evaluated in 72 Large White ${\times}$ Landrace ${\times}$ Duroc pigs weaned at 35 days of age. Pigs were fed a 1.30% lysine corn-soybean basal diet+15% dried whey+4% fish meal with carnitine at 0 or 50 mg/kg and either 0% added fat, 5% soybean oil or 5% lard for 6 weeks in a $2{\times}3$ factorial trial (6 treatments, 3 pens per treatment, 4 pigs per pen). Addition of carnitine increased average daily gain (ADG) and average daily feed intake (ADFI) in the second two weeks of the six-week trial and overall, but had no significant effect on feed per gain (F/G). Lard alone depressed ADG (p<0.05) in the last two weeks of the trial and overall, but the ADG for pigs fed lard+carnitine was similar to the control. Lard reduced feed intake in the first two weeks of the trial (p<0.05). Carnitine reduced the percentage of pigs with poor (ADG<375 g/d) growth (15 vs 40%; p<0.05). The greater uniformity of growth was most evident in low-weaning-weight pigs in the second period (16 vs 62%, p<0.005). Addition of fat did not produce any positive effect on uniformity and had no interaction with carnitine on uniformity. Carnitine addition increased serum total carnitione and short-chain acyl-carnitine levels (p<0.05), but did not modify free carnitine levels. Serum carnitine levels were lower at weaning than at 14, 28, or 39 days after weaning (p<0.05). Carnitine increased serum protein levels on day 14 (p<0.05). Addition of fat in the form of soybean oil or lard did not improve piglet growth performance. Addition of 50 mg/kg of carnitine to the diet of weanling pigs enhanced postweaning performance.

      • A novel imidazopyridine derivative, HS-106, induces apoptosis of breast cancer cells and represses angiogenesis by targeting the PI3K/mTOR pathway

        Abnormal activation of the phosphatidylinositol 3-kinase (PI3K)/Akt pathway is an essential step for the formation and growth of tumors in humans. HS-106 is an imidazopyridine derivative that inhibits the kinase activity of PI3K by binding to the ATP-binding cleft. We found that this compound suppressed breast cancer cell proliferation and induced apoptosis by specifically inhibiting the activity of target proteins in the PI3K/Akt/mTOR signaling pathway. Cell cycle analysis revealed that treatment with HS-106 resulted in cell cycle arrest at the G<SUB>2</SUB>/M phase due to up-regulation of p-cdc25 and down-regulation of cyclin B1. Also, HS-106 induced apoptosis by increasing the levels of cleaved caspase-3 and cleaved PARP. In addition, chromatin condensation and apoptotic bodies were detected in HS-106-treated breast cancer cells. Furthermore, HS-106 decreased the expression of hypoxia-inducible factor 1α (HIF-1α), and inhibited tube formation and migration of human umbilical vein endothelial cells (HUVECs) in vitro and blood vessel formation in an in vivo Matrigel plug assay. These results show that HS-106 may be an effective novel therapeutic candidate in clinical trials as a potential treatment for human breast cancers or other advanced malignancies with aberrant PI3K/Akt/mTOR signaling.

        Growth Performance, Meat Quality and Fatty Acid Metabolism Response of Growing Meat Rabbits to Dietary Linoleic Acid

        An experiment was conducted to determine the effects of different amounts of dietary linoleic acid (LA) on growth performance, serum biochemical traits, meat quality, fatty acids composition of muscle and liver, acetyl-CoA carboxylase (ACC) and carnitine palmitoyl transferase 1 (CPT 1) mRNA expression in the liver of 9 wks old to 13 wks old growing meat rabbits. One hundred and fifty 9 wks old meat rabbits were allocated to individual cages and randomly divided into five groups. Animals in each group were fed with a diet with the following LA addition concentrations: 0, 3, 6, 9 and 12 g/kg diet (as-fed basis) and LA concentrations were 0.84, 1.21, 1.34, 1.61 and 1.80% in the diet, respectively. The results showed as follows: the dietary LA levels significantly affected muscle color of LL included $a^*$ and $b^*$ of experimental rabbits (p<0.05). The linear effect of LA on serum high density lipoprotein cholesterol was obtained (p = 0.0119). The saturated fatty acids (SFAs) and monounsaturated fatty acids (MUFAs) contents of LL decreased and the polyunsaturated fatty acids (PUFAs) content of LL increased with dietary LA increase (p<0.0001). The PUFA n-6 content and PUFA n-3 content in the LL was significantly affected by the dietary LA levels (p<0.01, p<0.05). The MUFAs content in the liver decreased and the PUFAs contents in the liver increased with dietary LA increase (p<0.0001). The PUFA n-6 content and the PUFA n-6/n-3 ratio in the liver increased and PUFA n-3 content in the liver decreased with dietary LA increase (p<0.01). The linear effect of LA on CPT 1 mRNA expression in the liver was obtained (p = 0.0081). In summary, dietary LA addition had significant effects on liver and muscle fatty acid composition (increased PUFAs) of 9 wks old to 13 wks old growing meat rabbits, but had little effects on growth performance, meat physical traits and mRNA expression of liver relative enzyme of experimental rabbits.
