        Selective modulation of the glucocorticoid receptor can distinguish between transrepression of NF-κB and AP-1

        <P>Glucocorticoids (GCs) block inflammation via interference of the liganded glucocorticoid receptor (GR) with the activity of pro-inflammatory transcription factors NF-κB and AP-1, a mechanism known as transrepression. This mechanism is believed to involve the activity of GR monomers. Here, we explored how the GR monomer-favoring Compound A (CpdA) affects AP-1 activation and activity. Our results demonstrate that non-steroidal CpdA, unlike classic steroidal GCs, blocks NF-κB- but not AP-1-driven gene expression. CpdA rather sustains AP-1-driven gene expression, a result which could mechanistically be explained by the failure of CpdA to block upstream JNK kinase activation and concomitantly also phosphorylation of c-Jun. In concordance and in contrast to DEX, CpdA maintained the expression of the activated AP-1 target gene <I>c</I>-<I>jun</I>, as well as the production of the c-Jun protein. As for the underlying mechanism, GR is a necessary intermediate in the CpdA-mediated gene expression of AP-1-regulated genes, but seems to be superfluous to CpdA-mediated JNK phosphorylation prolongation. The latter phenomenon concurs with the inability of CpdA to stimulate DUSP1 gene expression. ChIP analysis demonstrates that DEX-activated GR, but not CpdA-activated GR, is recruited to AP-1-driven promoters. Furthermore, in mice we observed that CpdA instigates a strong enhancement of TNF-induced AP-1-driven gene expression. Finally, we demonstrate that this phenomenon coincides with an increased sensitivity towards TNF lethality, and implicate again a role for JNK2. In conclusion, our data support the hypothesis that a ligand-induced differential conformation of GR yields a different transcription factor cross-talk profile.</P><P><B>Electronic supplementary material</B></P><P>The online version of this article (doi:10.1007/s00018-013-1367-4) contains supplementary material, which is available to authorized users.</P>

        Towards a System Theoretical Understanding of the Parasport Context

        Contextual factors are fundamental but often overlooked ele- ments of parasport that can introduce various complexities within the parasport system. The purpose of this paper is to identify and categorize contextual factors that influence the parasport system and to examine the extent to which these factors influence the development of para-athletes’ pathways. Drawing from a system theoretical perspective, this research uses data gathered from 32 semi-structured interviews with stakeholders of the Brazilian Paralympic sports system. Thematic coding resulted in the identi- fication of nine main themes that constitute the contextual fac- tors of the Brazilian parasport system. The findings confirm that insofar as parasport is embedded in the complex cultural and national environment, it is not possible to understand parasport and the para-athlete trajectories without understanding the coun- try’s particular context with respect to disability. This paper thus provides insights toward better understanding the relationship between the diversity within the parasport systems and the socio- cultural context of a country.

        The Development of a Scale to Measure Shared Leadership in Youth Sport

        Shared leadership is described as the dynamic process of distrib- uting leadership influence across multiple team members. It is viewed as an effective form of management to deal with increased complexity in youth team sport. However, literature on youth leadership development and reliable and valid measures to assess shared leadership in youth sport is still lacking. This paper seeks to make a contribution to the youth leadership literature by developing an instrument to measure shared leadership among young football players. A total of 746 U17 players from 62 differ- ent youth football teams in two samples in The Netherlands were surveyed. The Youth Athlete Shared Leadership (YASL) in football instrument comprises three factors: ‘steering’, ‘coaching’ and ‘intervening’. This research contributes to the common under- standing of youth leadership development in general and youth team leadership in particular. The practical contribution is a tool for coaches to develop shared leadership behavior in youth foot- ball.
