The present study was undertaken to investigate effects of DMSO on the responses of dorsal horn cells to electrical or mechanical stimulation and also on the generation and conduction of neuronal impulses respectively at the receptors and sensory sxons. The responses of wide dynamic range (WDR) cells to electrical or mechanical stimulation were recorded before and after application of varying concentrations (15-100%) of DMSO onto the spinal cord, the sciatic nerve axons proximal to the stimulating electrode or the receptive field (RF). Effects of DMSO on the compound action potential recorded from the sciatic nerve were also studied. The responses of WDR cells to pinch of RF or to stimulation of the afferent C-fibers were more strongly inhibited by DMSO than the responses to brush or to the A-fiber stimulation. When applied to afferent neuronal axons and onto the spinal cord, DMSO induced a preferential conduction block of the C-fiber component of the compound action potential. These findings suggest that DMSO interferes with the mechanoelectric transducing mechanism at the receptor level and also causes conduction block of the afferent fibers and the spinal neurons as well.
