대한약전 제 4개정이 공포되어 2년이 경과하여 의약품의 이용에 많은 역할을 하고 있다. 대한약전 제 4개정에 수재된 의약품 중에서 생약 이 차지하는 비율은 순수생약이 18%, 항생물질을 포함하여 천연약물 유래의 성분 및 처방을 포함시키면 39%예 달하고 있으며 제 3개정에 비하여 한방에서 이용되는 생약이 많이 추가되었다. 이와같이 생약이 차지하는 비중이 높아진 것은 가까운 장래 실시 예정인 한방의료 보험등 천연약물에 대한 높은 관심에 부응하는 것이라 하겠다. 연자는 생약을 교육하는 입장에서 대한약전 제 4개정에 수재된 생약에 대해서 그 내용면에서 몇가지 고찰하고자 한다.
시호 Saponin의 면역 분석법에 관한 연구. : Ⅰ. Saikosaponin a의 면역원의 합성과 면역

For the immunoassay of saikosaponin a, one of the saponins from Bupleurum falcatum L. (Umbelliferae), immunogen was synthesized and immunization is in progress. For the immunogen, saikosaponin a was treated with succinic anhydride to get hemisuccinate of saikosaponin a and this hemisuccinate was reacted with BSA using CDI method to get sikosaponin a-BSA conjugate. From the results of quantitative analysis of this conjugate using phenol-H₂S0₄ method and diene transforming method, 3.8∼4.5 mole of saikosaponin a were found to be conjugated per mole of BSA.

These studies were attempted to investigate the therapeutic effect of Aster yomena extract on the liver damage induced by carbon tetrachloride (CCl₄) in rats. Male rats (SD) were treated with acute concentration of CCl₄, which was 1 ㎖/㎏ of CCl₄ via s.c injection for 3 days or 0.25, 0.5 ㎖/㎏ of CCl₄ via i.p injection. MeOH Ex. administration to rats (500 ㎎/㎏, p.o, twice a day) did not significantly decrease the increment in serum transaminase activities, but is`s frations (Frs), EtOAc and BuOH Fr., administration to rats (300 ㎎/㎏, p.o) significantly (p<0.05) decreased the enzyme activities (GOP, GPT) and liver enlargment. From histopathological observations, we could find that it`s administration prevented the development of fatty liver, necrosis and cytoplasmic vacuolations induced by CCl₄, and it gave a good agreement with the serological data. Futhermore, in doseresponse test, EtOAc Fr. administration at a dose of 400 ㎎/㎏ exhibited the most significant (p<0.01) activity, but it is net dose-dependent. This hepatoprotective effect was observed after 2 days from the last injection of CCl₄. From these results, we could suggest that Aster yomena contains a certain constituent in EtOAc or/and BuOH Fr. related a hepatoprotective activity for CCl₄-induced hepatotoxicity and we are investigating it`s active compound.

In order to survey possibility to produce saikosaponin from in vitro hairy root culture, culture of callus, adventitous root, and hairy root of Bupleurum falcatum L. were estabilished, and quantitative and qualitative aspects in saikosaponin extracted from thses were compared with those of cultivated root. Callus grew well in MS medium containing 0.9 ㎛ 2,4-D. In contrast, both of adventitous root and hairy root grew well in hormone-free MS medium. However, hairy root showed more rapid growth with extensive lateral root branches, characteristics of lower content of water and softer than in adventitous root. Among the selected lines of adventitous root and hairy root were observed difference in the growth rate. Mannopine, one of opine synthesized in the transformed tissue with Agrobacterium rhizogenes A_4 were detected in the extract of hairy root lines. Pattern and content of crude saponin from adventitous and hairy root showed no difference, but somewhat difference from those of cultivated root. However, in callus, distinct production-aspect of saponin was not observed.
