      • 두류의 항산화활성 검정

        시판되고 있는 두류 13종류의 70% 에탄을 추출물을 제조하여, 이들의 항산화 활성을 환원력, 지질과산화 억제 활성, superoxide radical 소저활성, hydroxyl radical 소거활성, mitomycin C로 유도된 DNA의 산화적 손상에 대한 억제활성을 지표로 조사하였다. 환원력은 예팥, 속피리, 유월콩 및 적두가 높았다. 지질과산화 억제능을 조사한 결과, linoleic acid 자동산화계를 이용한 실험계에서 모든 시료가 억제활성을 보인 반면, 토끼 적혈구막 지질과 산화에 대한 억제활성은 쥐눈이콩과 적두, 속피리에서만 관찰되었다. Superoxide radical 소거활성은 예팥, 나물콩, 적두, 쥐눈이콩에서 높게 나타났으며, hydroxyl radical 소거활성은 속피리, 청태, 예팥과 제비콩에서 높았다. Mitomycin C로 유도된 DNA의 산화적 손상에 대한 억제활성을 조사한 결과, 모든 품종들이 DNA의 산화적 손상을 억제할 수 있었으나, 특히 예팥, 쥐눈이콩, 나물콩, 녹두, 적두의 억제활성이 우수하였다. 이상의 실험결과에서 예팥, 적두, 쥐눈이콩, 속피리가 환원력, 지질과산화 억제, superoxide radical 및 hydroxyl radical 소거활성, DNA의 산화적 손상에 대한 억제활성 둥 항산화활성이 우수한 품종임을 알 수 있었다. Seventy percent ethanolic extracts from commercially available 13 legumes were made to investigated their antioxidative activities by determining the reducing power, inhibitory effect on lipid peroxidation, scavenging activity against both superoxide radical and hydroxyl radical, together with inhibitory activity toward mitomycin C-induced oxidative damage of DNA. High level of reducing powers were detected in Yepat, Sokpiri, Yuweol-bean and Jeokdu. Inhibitory effects on lipid peroxidation were found ubiquitously in all extracts examined when employing the linoleic acid autoxidation system, whereas, only 3 legumes, Yepat, Namul-bean and Jeenuni-bean, were revealed marked inhibition in rabbit erythrocyte-ghost membrane lipid peroxidation system. Yepat, Namul-bean, Jeokdu and Jeenuni-bean showed great scavenging activities on superoxide radical, on the other hand, high level of hydroxyl radical scavenging activities were demonstrated in Sokpiri, Chungtae, Yepat and Jebi-bean. Ubiquitous inhibitory effects on mitomycin C-induced oxidative damage on DNA were found in all extracts tested, Among them, however, Yepat, Jeenuni-bean, Namul-bean, Nokdu and Jeokdu showed the higher level of inhibition. Taken together, we could assign Yepat, Jeokdu, Jeenuni-bean and Sokpiri, for the legume species highly functional on overall antioxidative activity.

      • 발아 거대배아미의 당화력 및 전분입자의 이화학적 특성

        메품종의 거대배아미인 화청거대배아미, 남풍거대배아미, 그리고 찰품종인 화청찰거대배아미, 신선찰거대배아미 등 4품종의 거대배아미 품종의 볍씨를 각각 $27^{\circ}C$에서 3일간 발아시켜, 발아에 따른 전분 가수분해 효소의 활성 및 전분입자의 이화학적 특성을 각각 비교하였다. 3일간 발아시킨 벼의 ${\alpha}-amylase$의 활성은 맥아에 비해서 활성이 높게 나타났으며, 특히 화청거대배아미 및 신선찰거대배아미는 맥아보다 약 2배 정도의 활성을 나타내고 있었다. 이에 비해서 ${\beta}-amylase$의 경우는 일반품종보다는 거대배아미품종의 활성이 높기는 하지만 비교군인 맥아에 비해서 상당히 낮은 활성을 나타내고 있었다. 전분분자 중 아밀로오스 분자 유래의 긴 포도당 사슬의 양은 발아와 더불어 메품종 거대배아미에서는 줄어들고 있었으며, 찰품종에서는 증가하고 있었다. 아밀로펙틴 분자 유래의 포도당 사슬길이 분포는 찰벼와 메벼 품종에 관계없이 발아와 더불어 중합도가 14부터 60가지 비율은 증가하고 있었으며, 중합도 130이상 또는 13이하의 비율은 감소하고 있었다. Glucoamylase에 의한 가수분해도는 거대배아미메품종의 경우에는 발아와 더불어 현저히 낮아지고 있었고, 찰품종의 경우에는 오히려 증가하고 있었다. 그리고 호화개시온도, 호화종료 및 호화엔탈피는 감소하고 있었다. Rice seeds of 4 cultivars including Whachung-giant embryonic rice and Nampung-giant embryonic rice, as a group of the non-waxy rice cultivars, and Shinsunchal-giant embryonic rice and Whachungchal-giant embryonic rice, as that of the waxy rice cultivars, were germinated at $27^{\circ}C$ for 3 days to compare the changes in some physicochemical properties of the starch granules and the starch-hydrolysing enzyme activities during germination, respectively. ${\alpha}-Amylase$ activity of rices germinated for 3 days found to be higher than that of malt. Especially, Whachung-giant embryonic rice and Shinsunchal-giant embryonic rice were greater in activity than other rice cultivars and possessed the activities double that of malt. In contrast, ${\beta}-amylase$ of germinated rice found to be considerably less active than malt, although the giant embryonic rice group showed prevalent activity as compared o the normal rice group. With the starch granules, the amount of long glucose chains from amylose molecules were reduced in the non-waxy type giant embryonic rices, while the chain length increase was found in the waxy type giant embryonic rices. For the distribution profile of the glucose chain length from amylopectin molecules, we could observed that the chain length with DP (degree of polymerization) ranged 33 to 66 and 14 to 32 increased with the decreasing rate of that above 67 and below 13 regardless of starch waxiness. With non-waxy type of giant embryonic rices, susceptibility for glucoamylase were found to reduce along with germination, however, increase in susceptibility was observed with waxy rice types. In addition, we found the reduction in both initiation and termination temperature, and enthalpy for gelatinization.

      • 거대배 돌연변이 계통벼 쌀 배유 전분의 이화학적 특성

        화청찰벼, 화청벼, 그리고 이들의 거대배아미 품종인 화청찰거대배아미, 화청거대배아미 등 4품종의 쌀로부터 전분을 분리하여 몇몇 이화학적 특성을 비교하였다. 거대배아 품종으로 유기되면서 아밀로오스의 함량은 감소하고 있었으며, 아밀로오스 분획의 포도당 사슬의 길이도 짧아지고 있었다. 아밀로펙틴의 미세구조는 거대배아 품종으로 유기됨에 따라 B chain의 사슬길이가 약간씩 증가하는 경향이 있고, A chain의 양은 감소하고 있었으며, 아밀로펙틴을 구성하는 포도당의 평균 사슬길이는 약간씩 증가하고 있었다. X선 회절도상에서는 4품종 모두 전형적인 A type이었으며, glucoamylase에 의한 가수분해도는 찰품종의 경우에는 거대배아미로 유기되면서 가수분해도가 낮아지는데 비해서 메품종의 경우에는 반대의 결과를 얻었다. 호화종료온도는 거대배아미 품종으로 유기됨에 따라 낮아지고 있었고, 호화 엔탈피는 거대배아미 품종으로 유기됨에 따라 증가하고 있었다. Starches from 4 rice cultivars, including Whachungchalbyeo and Whachungbyeo, together with two correspondiug conversion mutants with giant embryonic phenotype, Whachungchal-giant embryonic rice and Whachung-giant embryonic rice, were isolated to compare its physico-chemical properties. The amylose content and the length of glucose chains in the fractionated amylose was reduced in response to conversion of the rice cultivars for developing giant embryonic phenotype. For the fine structure of amylopectin, conversion to giant embyonic rice cultivars accompanied with slight increase in the length of B chain plus decrease in the amount of A chain, and also a slight increase in average glucose chains length of amylopectin fractions could be observed. X-ray diffractogram revealed 4 cultivars to be the representative A types. We could also obtain the results that the hydrolysis rate by glucoamylase in the waxy rice cultivars decreased in response to conversion to the giant embryonic rice cultivars, while the rate increased in the non-waxy rice cultivas. Termination temperature for gelatinization was observed to decrease in response to conversion into the giant embryonic rice cultivars, however, the enthalpy for gelatinization was found to increase with the same conversion process.

        Effects of Solid-State Fermented Rice on Lipid Metabolism and Antioxidant Status in High-Cholesterol-Fed Rats

        We investigated the effect of solid-state fermented rice cultured with Basidiomycota (sangwhang) and Monascusruberon lipid metabolism and antioxidant activity. Forty 4-week-old male Sprague-Dawley rats were fed high cholesterol di-ets in which carbohydrate sources in the treatment groups consisted of non-fermented rice and sangwhang or M. ruberriceat 80% and 20%, respectively, for 5 weeks. Supplementation with sangwhang and M. ruberrice had no effect on growth andfood intakes in high-cholesterol-fed rats. The plasma triglyceride concentration was not significantly different among theM. ruber rice resulted in lower plasma and hepatic cholesterol concentrations and atherogenicindex compared to the control group, while the plasma high-density lipoprotein-cholesterol concentration was elevated. In ad-dition, fermented rice cultured with M. ruber-supplemented animals had greater bile acid excretion. The M. rubergroups hadsignificantly lower plasma and hepatic levels of thiobarbituric acid-reactive substances than the control group. Moreover, he-patic antioxidant enzyme activities, including catalase and superoxide dismutase, were significantly higher in the M. ruberM. ruberrice, was very effective for improving the lipid metabolism and re-ducing oxidative stress by up-regulating the hepatic antioxidant enzymes in high-cholesterol-fed rats.

        유색미 쌀겨 추출물의 화학적 변이원 mitomycin C 에 대한 변이원성 억제기각

        Inhibitory mechanism of colored rice bran against cellular genotoxicity induced by chemical mutagen was studied using organic solvent extracts from a colored rice cultivar termed as Suwon415, and the mutagen, mitomycin C. Inhibitory effects of 70% ethanol extact and chloroform fraction from rice bran of Suwon415 were higher than those from Chuchung used as control. However, antioxidative activities of each fraction from Suwon415 were slightly lower than those from Chuchung, suggesting the involvement of a different inhibitory mechanism not related to antioxidation pathway. Using E. coli as the indicator cell, inhibitory mechanism of rice bran extract from colored rice against mutagenicity induced by mitomycin C was investigated to reveal the possibility that it acts in a desmutagenic manner. Further investigation to quantify the free mitomycin C in reaction mixture following incubation with rice bran extract demonstrated that rice bran extract might inhibit the cellular genotoxicity of mitomycin C by direct adsorption of the mutagen.
