        IL-33 down-regulates CLDN1 expression through the ERK/STAT3 pathway in keratinocytes

        <P><B>Abstract</B></P> <P><B>Background</B></P> <P>Tight junctions (TJs) have important roles in skin barrier function. The TJ protein claudin-1 (CLDN1) is decreased in atopic dermatitis (AD). However, little is known about the mechanism of CLDN1 down-expression.</P> <P><B>Objective</B></P> <P>To elucidate the effect of IL-33 on CLDN1 expression in keratinocytes.</P> <P><B>Methods</B></P> <P>Normal human epidermal keratinocytes (NHEKs) and human skin equivalent models (HSEMs) were cultured <I>in vitro</I> in the presence of IL-33. Production of CLDN1, signal transducer and activator of transcription 3 (STAT3) and Mitogen-activated protein kinases (MAPK) expression were measured by real-time PCR, western blot and immunofluorescence assay. MAPK inhibitors and small interfering RNA were used to confirm the signal pathway of STAT3 and CLDN1. Barrier function was measured by transepithelial electric resistance (TEER) and FITC-dextran flux assays. Electrophoretic Mobility Shift Assay was used to detect STAT3 transcriptional activity.</P> <P><B>Results</B></P> <P>Levels of CLDN1 expression were reduced in the epidermis of AD-model mice overexpressing IL-33. IL-33 down-regulated the expression of CLDN1 mRNA and protein in NHEKs and HSEMs. IL-33 attenuated transepithelial electric resistance and induced FITC-dextran flux in NHEKs. The IL-33 suppressed CLDN1 expression was regulated by an extracellular signal-regulated kinase (ERK) and signal transducer and activator of transcription 3 (STAT3). STAT3 suppressed CLDN1 expression by direct binding to the promoters.</P> <P><B>Conclusion</B></P> <P>IL-33 may down-regulate CLDN1 expression through the ERK/STAT3 pathway in keratinocytes.</P> <P><B>Highlights</B></P> <P> <UL> <LI> IL-33 may be key regulator of CLDN1 downregulation in keratinocytes. </LI> <LI> The expression of CLDN1 is regulated by ERK/STAT3 pathway in IL-33 treated keratinocytes. </LI> <LI> IL-33 reduced CLDN1 expression is regulated by direct binding of STAT3 to CLDN1 promoter region. </LI> </UL> </P>

        Quassinoids isolated from <i>Brucea javanica</i> inhibit pepper mottle virus in pepper

        <P><B>Abstract</B></P> <P>A green fluorescent protein (GFP)-tagged pepper mottle virus (PepMoV) based leaf-disc method and systemic host method were developed to identify antiviral agents. Preliminary experiments using a PepMoV-GFP based leaf-disc method led to the isolation of five quassinoids, including brusatol (<B>1</B>), bruceantin (<B>2</B>), brucein A (<B>3</B>), bruceantinol (<B>4</B>), and brucein B (<B>5</B>), from the CH<SUB>3</SUB>OH extract of <I>Brucea javanica</I>. All isolated compounds exhibited inactivation effects in systemic host plants, and compounds <B>3</B> and <B>4</B> were potent, with a minimum inhibitory concentration of 10 <I>μ</I>M.         가우시안 혼합모델을 이용한 공항 접근 패턴 추출 및 패턴 별 과이탈 확률 분석

        항공기 착륙 시에는 정해진 절차에 따라 접근이 이루어진 다음, 활주로 중심선과 정렬하여 착륙하게 된다. 하지만 공항의 상황, 주변 항공기의 상황, 또는 관제사의 지시 등에 따라 빈번한 레이더 벡터링이 일어나기 때문에, 교통 흐름을 파악하거나, 비행 안전성을 파악하기 위해서는 항공기의 접근 패턴을 인지할 필요가 있다. 또한 최종 접근 시 활주로 중심선과 정렬하는 과정에서 과이탈이 발생하는 경우가 있는 데, 이는 이후 불안정 접근 등과 같이 보다 위험한 상황을 초래할 수 있다. 본 논문에서는 클러스터링 기법을 이용하여 접근 구간에서의 항공기 궤적들의 패턴을 추출하였다. Gaussian Mixture Model(GMM)을 이용하여 김해공항 접근 항공기 궤적에 대한 클러스터링을 진행하였으며, 2019년 1년간 김해공항으로 착륙한 항공기의 데이터를 이용하였다. 클러스터 별 centroid 값을 이용하여, 총 86개의 접근 궤적 패턴을 추출하였다. 그 후 각 클러스터 내 항공기 중 최종 접근시 과이탈하는 항공기를 탐지하여 확률 분포를 계산하였다. When an aircraft is landing, it is expected that the aircraft will follow a specified approach procedure and then land at the airport. However, depending on the airport situation, neighbouring aircraft or the instructions of the air traffic controller, there can be a deviation from the specified approach. Detecting aircraft approach patterns is necessary for traffic flow and flight safety, and this paper suggests clustering techniques to identify aircraft patterns in the approach segment. The Gaussian Mixture Model (GMM), one of the machine learning techniques, is used to cluster the trajectories of aircraft, and ADS-B data from aircraft landing at the Gimhae airport in 2019 are used. The aircraft trajectories are clustered on the plane, and a total of 86 approach trajectory patterns are extracted using the centroid value of each cluster. Considering the correlation between the approach procedure pattern and overshoots, the distribution of overshoots is calculated.

        Open versus closed treatment for extracapsular fracture of the mandibular condyle

        Objectives: Selection of treatment methods for mandibular condylar fractures remains controversial. In this study, we investigated treatment methods for condylar fractures to determine the indications for open or closed reduction. Patients and Methods: Patients >12 years of age treated for mandibular condylar fractures with a follow-up period of ≥3 months were included in this study. The medical records of enrolled patients were reviewed for sex, age, fracture site, treatment method (open or closed reconstruction), postop-erative intermaxillary fixation period, operation time, and complications. Radiological analysis of fracture fragment displacement and changes in ramal height difference was performed using computed tomography and panoramic radiography. Results: A total of 198 patients was investigated, 48.0% (n=95) of whom underwent closed reduction and 52.0% (n=103) underwent open reduction. There was no significant correlation between reduction method and patient sex, age, or follow-up period. No statistically significant difference between the incidence of complications and treatment method was observed. None of the patients underwent open reduction of condylar head fracture. Binary logistic regression analysis showed that open reduction was significantly more frequent in patients with subcondylar fracture compared to in those with a fracture in the condylar head area. There was no statistically significant correlation between the groups and fracture fragment displacement. However, there was a significant difference between the treatment groups in amount of change in ramal height difference between the fractured and the non-fractured sides during treatment. Conclusion: No significant clinical differences were found between the open and closed reduction methods in patients with mandibular condylar fractures. According to fracture site, closed reduction was preferred for condyle head fractures. There was no significant relationship between fracture fragment displacement and treatment method.

        <i>Xylodon flaviporus</i>-Derived Drimane Sesquiterpenoids That Inhibit Osteoclast Differentiation

        <P>The presence of excessive osteoclasts is a major factor in skeletal diseases. The present study aimed to discover osteoclast differentiation inhibitors from the basidiomycete <I>Xylodon flaviporus</I>. Seven new drimane sesquiterpenoids (<B>1</B>-<B>7</B>) and 7-ketoisodrimenin-5-ene (<B>8</B>) were obtained and characterized by various spectroscopic methods. The isolated compounds were evaluated for their inhibitory effects against receptor activator of nuclear factor-kappa-B ligand-induced osteoclastogenesis in mouse bone marrow macrophages. Compounds <B>1</B>, <B>3</B>, and <B>6</B> showed potent activities with IC<SUB>50</SUB> values of 1.6, 0.9, and 2.1 μM, respectively, while <B>4</B>, <B>5</B>, and <B>7</B> exhibited relatively weak activities with IC<SUB>50</SUB> values of 10.7, 10.1, and 8.5 μM, respectively.</P> [FIG OMISSION]</BR>

        Complete chromosome and plasmid sequences of Staphylococcus aureus strain JDFM SA01, isolated from a milk filter in Korean dairy farm

        Staphylococcus aureus is a significant pathogen that can source a variety of illness worldwide. In this announcement, we report here the complete genome sequence of S. aureus strain JDFM SA01, isolated from a milk filter collected from Korean dairy farm. The final complete genome assembly consists of one circular chromosome (2,748,925 bp) with an overall GC content of 32.9% and one circular plasmid sequence (24,655 bp) with a GC content of 28.7%.

        <P>Indium tri(isopropoxide)-catalyzed Meerwein–Ponndorf–Verley reduction of aliphatic and aromatic aldehydes in 2-propanol gave selectively the corresponding primary alcohols in good to excellent yields at room temperature. A wide range of functional groups including alkene, ether, ketone, ester, nitrile, and nitro were tolerated under the optimum reaction conditions.         배우자와의 나이차이와 건강상태 및 의료이용의 관련성

        연구목적 : 본 연구는 배우자와의 나이차이와 건강상태 및 의료이용의 관련성을 파악하고자 하였다. 연구방법 : 자료는 한국의료패널 연간데이터 중 2012년 데이터를 활용하였으며, 배우자자료가 있는 40세 이상의 응답자 6,910명을 분석대상으로 선정하였다. 배우자와의 나이차이에 따라 건강상태(주관적 건강상태, 동년배 비교 건강상태, 불안/우울)와 의료이용(외래이용횟수, 재원일수)의 관련성을 파악하였다. 연구결과 : 남자(여자)는 배우자보다 평균 3.7세(3.4세) 많았으며(적었으며), 남자와 여자 모두 3세 차이가 가장 많은 빈도(최빈값)를 차지하였으며, 배우자보다 1세~5세 많은 남자(1세~5세 적은 여자)의 경우가 각각 59% 수준이었다. 남자의 경우, 배우자와의 나이차이는 주관적 건강상태 및 동년배 비교 건강상태와 모두 약한 음의 상관성이 있었으며, 불안/우울, 외래방문수, 재원일수 등과는 약한 양의 상관성이 있었다. 여자의 경우, 배우자와의 나이차이는 자신의 주관적 건강수준 및 동년배 비교 건강상태와 약한 양의 상관성이 있었으나, 불안/우울과는 음의 상관성이 있었으나, 유의하지 않았다. 결론 : 배우자와의 나이차이에 대한 기술통계, 그리고 배우자의 나이차이와 건강상태 및 의료이용의 상관성은 건강을 고려한 배우자 선택과 보건의료정책에 활용성이 기대된다. 그러나, 연령대, 생활습관, 배우자의 지지, 혼인기간, 혼인상태 등을 고려한 추가연구를 통하여 보다 유용한 의미를 도출하여야 할 것이다. Objective: The purpose of this study was to investigate the relationships of age gap between partners with health status and utilization of health service. Methods: We used the data of year 2012 in the annual Korea Health Panel administered by joint supervision of Korea Institute for Health and Social Affairs and National Health Insurance Corporation. We analyzed data for 6,910 individuals that had a spouse and were above 40 years old. Results: Man(woman)`s average age gap with partner was 3.7(-3.4) years, and mode of man`s(woman`s) age gap with partner was 3(-3) years. For men, age gap between partners had weak positive correlations with frequency of outpatient service utilization, anxiety and depressive symptoms, and length of hospital stay, and weak negative correlations with self-rated health status. For women, age gap between partners had weak positive correlations with self-rated health status, and weak negative correlations with frequency of outpatient service use. Conclusion: The relationships of age gap between partners with health status and health service utilization have implications to culture of marriage and health policy on family life. Further research on the mediating factors with the relationships are needed.
