        Proteomic Analysis of Shigella Virulence Effectors Secreted under Different Conditions

        A series of novel effector molecules secreted by the type three secretion system (T3SS) of Shigella spp. have been reported in recent years. In this study, a proteomic approach was applied to study T3SS effectors systematically. First, proteins secreted by the S. flexneri wildtype strain after Congo Red induction were separated and identified using two-dimensional electrophoresis to display the relative abundance of all kinds of early effectors for the first time. Then, a gene deletion mutant of known virulence repressor (OspD1) and a gene overexpressed mutant of two known virulence activators (MxiE and IpgC) were constructed and analyzed to discover potential late effectors. Furthermore, the supernatant proteins of gene deletion mutants of two known translocators (IpaB and IpaD), which would constantly secrete effectors, were also analyzed. Among all of the secreted proteins identified in our study, IpaH1.4, IpaH_5, and IpaH_7 have not been reported before. These proteomics data of the secreted effectors will be valuable to understand the pathogenesis of S. flexneri.

        Research on the Carbonation Resistance and Carbonation Depth Prediction Model of Fly Ash- and Slag-Based Geopolymer Concrete

        To study the effect of different slag content and different curing methods on the carbonation resistance of fly ash- and slag-based geopolymer concrete (FA&SL-GPC), a series of experiments were carried out with slag content (accounting for 10%, 30% and 50% of cementitious materials) and curing methods (standard curing and high temperature curing) as variables. The results show that increasing the slag content is helpful for improving the carbonation resistance properties, microstructure, and carbonation resistance components of FA&SL-GPC substrate. Adopting a standard curing method and prolonging the curing age are more favorable to the carbonation resistance of FA&SL-GPC, and the longer the curing time is, the better. Finally, the carbonation depth prediction model suitable for this experiment is deduced and verified. From the research results, it can be seen that using a high slag content mix and curing under standard conditions is beneficial for the carbonization resistance of GPC structural components.

      • Controlled synthesis of nanoplate, nanoprism and nanopyramid-shaped CdSe decorated on porous TiO<sub>2</sub> photocatalysts for visible-light-driven hydrogen evolution

        <P><B>Abstract</B></P> <P>Herein, we report a successful synthesis of porous TiO<SUB>2</SUB> monoliths decorated with unique nanoplate, nanoprism, and nanopyramid-shaped CdSe particles through a mild selenylation of CdO embedded inside porous TiO<SUB>2</SUB> monoliths via a hydrothermal method in a very controlled manner. Compared with pure TiO<SUB>2</SUB>, as-synthesized CdSe/TiO<SUB>2</SUB> photocatalyst not only enhances light absorption but also leads to a highly efficient charge-carrier separation. Particularly, the nanoplate-shaped 7% CdSe/TiO<SUB>2</SUB> photocatalyst (molar percentages of CdSe to TiO<SUB>2</SUB> is 7:100) exhibits an exceptional hydrogen evolution rate up to 3650 μmol h<SUP>−1</SUP> g<SUP>−1</SUP> without resorting to any noble-metal co-catalysts under visible-light irradiation owing to synergistic effects envisaged by a rational material design. Our results may provide a useful strategy to develop a highly-efficient visible-light-driven hydrogen production system via water splitting.</P>

      • A stable and highly efficient visible-light-driven hydrogen evolution porous CdS/WO<sub>3</sub>/TiO<sub>2</sub> photocatalysts

        <P><B>Abstract</B></P> <P>It is well known that both catalytic efficiency and stability are the two important parameters of photocatalysts for visible-light-driven hydrogen production reactions. However, light-driven hydrogen evolution based applications still suffer from sluggish reaction kinetics due to the lack of high-performance photocatalysts. In this paper, we successfully synthesized a ternary porous CdS/WO<SUB>3</SUB>/TiO<SUB>2</SUB> photocatalyst with high efficiency and stability via two-stage approach. The as-prepared samples are characterized by XRD, FESEM, EDS, TEM, XPS, and UV–Vis, respectively, which illustrated that the CdS and WO<SUB>3</SUB> moieties are in-situ formed inside the porous TiO<SUB>2</SUB>. Particularly, the photocatalytic hydrogen (H<SUB>2</SUB>) evolution rate of such ternary 8% CdS/WO<SUB>3</SUB>/TiO<SUB>2</SUB> (molar ration of CdS:WO<SUB>3</SUB>:TiO<SUB>2</SUB> = 8:8:100) photocatalyst ranges up to 2106 μmol h<SUP>−1</SUP> g<SUP>−1</SUP> under visible-light irradiation, which is higher than that of pure TiO<SUB>2</SUB> and other binary (CdS/TiO<SUB>2</SUB> and WO<SUB>3</SUB>/TiO<SUB>2</SUB>) porous photocatalysts. The superior H<SUB>2</SUB> evolution efficiency can be attributed to the coexistence of CdS and WO<SUB>3</SUB> in porous TiO<SUB>2</SUB> which can promote the interfacial charge transfer and separation as well as extend the light absorption up to the visible range.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Highly efficient and stable photocatalytic activity of CdS/WO<SUB>3</SUB>/TiO<SUB>2</SUB> photocatalysts were synthesized by a two-step method. </LI> <LI> CdS and WO<SUB>3</SUB> inlaid on porous TiO<SUB>2</SUB> can extend the light absorption and enhance photogenerated electron-hole pairs separation. </LI> <LI> The porous structure can provide more reaction active sites and improve photoproduced-electrons and holes transport speed. </LI> </UL> </P> <P><B>Graphical Abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Epigenetic regulation of key gene of PCK1 by enhancer and super-enhancer in the pathogenesis of fatty liver hemorrhagic syndrome

        Objective: Rare study of the non-coding and regulatory regions of the genome limits our ability to decode the mechanisms of fatty liver hemorrhage syndrome (FLHS) in chickens.Methods: Herein, we constructed the high-fat diet-induced FLHS chicken model to investigate the genome-wide active enhancers and transcriptome by H3K27ac target chromatin immunoprecipitation sequencing (ChIP-seq) and RNA sequencing (RNA-Seq) profiles of normal and FLHS liver tissues. Concurrently, an integrative analysis combining ChIP-seq with RNA-Seq and a comparative analysis with chicken FLHS, rat non-alcoholic fatty liver disease (NAFLD) and human NAFLD at the transcriptome level revealed the enhancer and super enhancer target genes and conservative genes involved in metabolic processes.Results: In total, 56 and 199 peak-genes were identified in upregulated peak-genes positively regulated by H3K27ac (Cor (peak-gene correlation) ≥0.5 and log2(FoldChange) ≥1) (PP) and downregulated peak-genes positively regulated by H3K27ac (Cor (peak-gene correlation) ≥0.5 and log2(FoldChange)≤–1) (PN), respectively; then we screened key regulatory targets mainly distributing in lipid metabolism (PCK1, APOA4, APOA1, INHBE) and apoptosis (KIT, NTRK2) together with MAPK and PPAR signaling pathway in FLHS. Intriguingly, PCK1 was also significantly covered in up-regulated super-enhancers (SEs), which further implied the vital role of PCK1 during the development of FLHS.Conclusion: Together, our studies have identified potential therapeutic biomarkers of PCK1 and elucidated novel insights into the pathogenesis of FLHS, especially for the epigenetic perspective.
