Abstract

A contracted doubly N‐confused dioxohexaphyrin derivative served as a dinucleating metal ligand for unsymmetrical coordination. The complexation of two palladium(II) cations led to the formation of π‐radical species that were persistent in atmospheric air in the presence of moisture. Effective delocalization of an unpaired electron over the hexaphyrin backbone could contribute to the distinct chemical stability.

We present high-resolution long-slit spectroscopy of a Balmer-dominated shock in the northeastern limb of the Cygnus Loop with the Subaru high dispersion spectrograph. By setting the slit angle along the shock normal, we investigate variations of the flux and profile of the H alpha line from preshock to postshock regions with a spatial resolution of similar to 4 x 10(15) cm. The Ha line profile can be represented by a narrow (28.9 +/- 0.7 km s(-1)) Gaussian in a diffuse region ahead of the shock, i.e., a photoionization precursor, and narrow (33.1 +/- 0.2 km s(-1)) plus broad (130-230 km s(-1)) Gaussians at the shock itself. We find that the width of the narrow component abruptly increases up to 33.1 +/- 0.2 km s(-1), or 38.8 +/- 0.4 km s(-1) if we eliminate projected emission originating from the photoionization precursor, in an unresolved thin layer (less than or similar to 4 x 10(15) cm at a distance of 540 pc) at the shock. We show that the sudden broadening can be best explained by heating via damping of Alfven waves in a thin cosmic-ray (CR) precursor, although other possibilities are not fully ruled out. The thickness of the CR precursor in the Cygnus Loop (a soft gamma-ray emitter) is an order of magnitude thinner than that in Tycho's Knot g (a hard gamma-ray emitter), which may be caused by the different energy distribution of accelerated particles between the two sources. In this context, systematic studies might reveal a positive correlation between the thickness of the CR precursor and the hardness of the CR energy distribution.

Similarity of Intracellular Signaling Toward Apoptosis Following UVB and UVC Irradiation

UV irradiation activates various intracellular signaling pathways causing cell death in a DNA damage-dependent and an independent manner. As DNA photoproducts, major forms of DNA damage, are maximally formed by UV light at 260-nm, short wavelength UV (UVC) is more harmful than middle wavelength UV (UVB). However, the differences or similarities in responses of DNA damage-independent intracellular signaling molecules to UVB and UVC are not elucidated. We examined activation of signaling molecules towards apoptosis in normal human fibroblastic cells after irradiation with UVB or UVC at a dose generating the equal amount of DNA photoproducts. Both UVB and UVC induced transient phosphorylation of ERK and sustained phosphorylation of p38. Phosphorylation of p53 at Ser15 and at Ser392 residues were also observed, which were inhibited by a phosphoinositide 3-kinase inhibitor, wortmannin. In contrast, an antioxidant N-acetyl-cysteine and a p38 inhibitor SB203580 suppressed only Ser392 phosphorylation, suggesting that UV-induced oxidative stress and p38 activation were involved in the phosphorylation of this site. The apoptic signals such as mitochondrial cytochrome C release and annexin V binding were then observed. Overall, no difference was found in chronological responses of p53, MAPK, and apoptosis between UVB-irradiated and UVC-irradiated cells. These results suggested that DNA damage-independent intracellular signaling molecules similarly responded to UVB and UVC when the equal level of DNA photoproducts were generated.

Nocturnal heart rate variability (HRV) is thought to reflect healthy recov-ery function of the autonomic nervous system. Although exercise is recommended for health promotion, exercise itself decreases HRV. We studied acute effect of daytime exercise on nocturnal HRV in 5 healthy adults (age, 22–40 years; 2 female subjects) without regular exercise habit. Using a treadmill, they performed 30-min walking at 4 km/hr and 30-min running at 9 km/hr from 11 a.m. on different days at an interval of 2 weeks. On these days and a day without exercise (control), Holter electrocardiograms were recorded from 9 a.m. for 24 hr. The amplitudes of low-frequency (LF, 0.04–0.15 Hz) and high-frequency (HF, 0.15–0.45 Hz) components of HRV were measured continuously by complex de-modulation and were averaged over periods of 11:00–11:30 a.m., 3 hr af-ter going to bed, and time in bed at night. Exercise intensities of the walking and running were at 10% to 44% and 55% to 67% of heart rate reserve, respectively. During exercise, heart rate increased and LF and HF amplitudes decreased with exercise intensity. Nocturnal heart rate and LF and HF amplitude, however, showed no consistent changes with exercise intensity and their averages on the days of walking and run-ning did not differ significantly from those of the control day. In conclu-sion, 30-min walking and running exercises performed in the morning had no significant acute effects on nocturnal heart rate or HRV.

An endocellulase-free multienzyme complex was produced by a thermophilic anaerobic bacterium, Thermoanaerobacterium thermosaccharolyticum strain NOI-1, when grown on xylan. The temperature and pH optima for growth were 60℃ and 6.0, respectively. The bacterial cells were found to adhere to insoluble xylan and Avicel. A scanning electron microscopy analysis showed the adhesion of xylan to the cells. An endocellulase-free multienzyme complex was isolated from the crude enzyme of strain NOI-1 by affinity purification on cellulose and Sephacryl S-300 gel filtration. The molecular mass of the multienzyme complex was estimated to be about 1,200 kDa. The multienzyme complex showed one protein on native PAGE, one xylanase on a native zymogram, 21 proteins on SDS-PAGE, and 5 xylanases on a SDS zymogram. The multienzyme complex consisted of xylanase, β-xylosidase, α-L-arabinofuranosidase, β-glucosidase, and cellobiohydrolase. The multienzyme complex was effective in hydrolyzing xylan and corn hulls. This is the first report of an endocellulase-free multienzyme complex produced by a thermophilic anaerobic bacterium, T. thermosaccharolyticum strain NOI-1.
