        Müller glial responses compensate for degenerating photoreceptors in retinitis pigmentosa

        Photoreceptor degeneration caused by genetic defects leads to retinitis pigmentosa, a rare disease typically diagnosed in adolescents and young adults. In most cases, rod loss occurs first, followed by cone loss as well as altered function in cells connected to photoreceptors directly or indirectly. There remains a gap in our understanding of retinal cellular responses to photoreceptor abnormalities. Here, we utilized single-cell transcriptomics to investigate cellular responses in each major retinal cell type in retinitis pigmentosa model (P23H) mice vs. wild-type littermate mice. We found a significant decrease in the expression of genes associated with phototransduction, the inner/outer segment, photoreceptor cell cilium, and photoreceptor development in both rod and cone clusters, in line with the structural changes seen with immunohistochemistry. Accompanying this loss was a significant decrease in the expression of genes involved in metabolic pathways and energy production in both rods and cones. We found that in the Müller glia/astrocyte cluster, there was a significant increase in gene expression in pathways involving photoreceptor maintenance, while concomitant decreases were observed in rods and cones. Additionally, the expression of genes involved in mitochondrial localization and transport was increased in the Müller glia/astrocyte cluster. The Müller glial compensatory increase in the expression of genes downregulated in photoreceptors suggests that Müller glia adapt their transcriptome to support photoreceptors and could be thought of as general therapeutic targets to protect against retinal degeneration.

      • The EEG Analysis Method for Obtaining the Feeling

        The electroencephalogram (EEG), the brain activity of neurons in the brain, is used to record brain activity for many purposes. It also changes depends on mental conditions because mental conditions are generated in the brain. In this study, we try to estimate the mental conditions by observing the EEG patterns. The EEG data caused by mental changes is obtained as giving the subjects animal assisted therapy. The method for estimation is AdaBoosting as the neural network. We set the input vector as EEG changes and output vector is set as mental changes. Subjects are 25 people: 19 males and 6 females (age: 21-24 years old). Previous study reveals the correlation between the EEG and the activeness, and between the EEG and the tiredness. Therefore, in this paper, we try to estimate these mental parameters by the EEG at the first step.

        In this paper, we propose an extraction method of ride comfort according to the difference of the tire by electroencephalogram (EEG). Recently, the sensory evaluation is used for development of the product. However, the sensory evaluation by a general user who doesn’t have a clear standard is unstable. Moreover, ride comfort depends on the sensibility and the preference of the individual. It is considered that these psychological changes influence the brain that rules center of sensation. Therefore, we pay attention to the EEG. We use the EEG that it is possible to measure it simply in the brain function measurement technique as an objective evaluation. In this study, the feature of the EEG during the driving is extracted by the factor analysis (FA). Moreover, we investigate the correlation of the subjective evaluation and the EEG features. From the result, the EEG features and subjective evaluation features has correlation. Thus, the effectiveness of the proposed method as an objective evaluation method was shown.

        Rehabilitation Nutrition and Exercise Therapy for Sarcopenia

        Sarcopenia is an age-related loss of skeletal muscle associated with adverse outcomes such as falls, fractures, disability, and increased mortality in older people and hospitalized patients. About half of older male nursing home residents have sarcopenia. The diagnostic criteria by the European Working Group on Sarcopenia in Older People (EWGSOP) and the Asian Working Group for Sarcopenia (AWGS) have led to increased interest in sarcopenia. Exercise and nutritional management are crucial for the prevention and treatment of sarcopenia. Nutritional therapy for sarcopenia that includes 20 g of whey protein and 800 IU of vitamin D twice a day improves lower limb strength. Exercise therapy for sarcopenia, such as resistance training and 6 months of home exercises, improves muscle strength and physical function. Combination therapy that includes both nutritional and exercise therapy improves gait speed and knee extension strength more than either exercise alone or nutrition therapy alone. Excessive bedrest and mismanagement of nutrition in medical facilities can lead to iatrogenic sarcopenia. Iatrogenic sarcopenia is sarcopenia caused by the activities of health care workers in health care facilities. Appropriate nutritional management and exercise programs through rehabilitation nutrition are important for prevention and treatment of iatrogenic sarcopenia. Nutritional and exercise therapy should be started very early after admission and adjusted to the level of inflammation and disease status. Repeated assessment, diagnosis, goal setting, interventions, and monitoring using the rehabilitation nutrition care process is important to maximize treatment effectiveness and improve patients’ functional recovery and quality of life.
