        Cadmium stress inhibits the growth of primary roots by interfering auxin homeostasis in Sorghum bicolor seedlings

        Phytotoxic effects of cadmium (Cd), a heavy metalpollutant, on plants have been extensively examined. Auxinplays vital roles in many aspects of plant development. Theassociation between root growth and auxin signaling in CdstressedSorghum bicolor was analyzed in our study. Rootelongation, shoot length and the maximal photochemicalefficiency (Fv/Fm) in S. bicolor seedlings were dramaticallyreduced after Cd stress treatment. Cd was found to bepredominantly confined in the meristematic zone using a Cdstainingmethod. Cd stress remarkably influenced the cellcycle progression at the root tip as shown by EdU (ethynyldeoxyuridine) assay. The content of IAA was markedlydiminished in the roots of Cd-stressed S. bicolor, which wasalong with the increase of IAA oxidase activity. Auxintransport inhibitors, 1-naphthoxyacetic acid (1-NOA) or 1-naphthylphthalamic acid (NPA), greatly reduced plant toleranceto Cd stress, whereas exogenous application of 1-naphthaleneacetic acid (NAA) improved Cd tolerance in S. bicolor seedlings. Cd stress altered the transcript level ofsome putative auxin biosynthetic genes. In addition, NAAinterfered with the homeostasis of Cd-induced reactiveoxygen species (ROS). These results revealed that Cd stressdisturbed the growth of S. bicolor seedlings by affecting thehomeostasis of auxin and ROS.

        Efficacy of Lauromacrogol Injection for Ablation of Benign Predominantly Cystic Thyroid Nodules and Related Factors: A Prospective Study

        Objective: To prospectively evaluate the efficacy of lauromacrogol injection for ablation (LIA) of benign predominantly cystic thyroid nodules and its related factors. Materials and Methods: A total of 142 benign predominantly cystic thyroid nodules (median volume, 12.5 mL; range, 0.4– 156 mL) in 137 patients (male:female sex ratio, 36:101; mean age ± standard deviation [SD], 49 ± 13 years) were treated with LIA after being confirmed as benign via cytology. The volume reduction rate (VRR) of the nodules and cosmetic score were evaluated during follow-up at 1, 3, and 6 months after treatment and every 6 months thereafter. A VRR of ≥ 50% at the 12-month follow-up was considered to indicate effective treatment. The associations between the clinical factors and nodular ultrasound features, including the initial nodule volume, proportion of solid components, vascularity grade and ineffective treatment (VRR of < 50% at the 12-month follow-up), and regrowth were analyzed. Results: All patients completed follow-up for at least 12 months. The average ± SD follow-up period was 32 ± 11 months (range, 12–54 months). The effective treatment rate was 73.2% (104/142), while the regrowth rate was 12.0% (17/142) at the last follow-up. Grade 2–3 intranodular vascularity in the solid components of the nodules was the only independent factor associated with ineffective treatment, with an odds ratio (reference category, grade 0–1) of 3.054 (95% confidence interval, 1.148–8.127) (p = 0.025). Conclusion: LIA is an effective treatment for predominantly cystic thyroid nodules. Grade 2–3 intranodular vascularity in the solid components of nodules is the only independent risk factor for ineffective LIA.

        Isolation and Identification of Lipopeptide Antibiotics from Paenibacillus elgii B69 with Inhibitory Activity Against Methicillin-Resistant Staphylococcus aureus

        Two lipopeptide antibiotics, pelgipeptins C and D, were isolated from Paenibacillus elgii B69 strain. The molecular masses of the two compounds were both determined to be 1,086 Da. Mass-spectrometry, amino acid analysis and NMR spectroscopy indicated that pelgipeptin C was the same compound as BMY-28160, while pelgipeptin D was identified as a new antibiotic of the polypeptin family. These two peptides were active against all the tested microorganisms, including antibiotic-resistant pathogenic bacterial strains such as methicillin-resistant Staphylococcus aureus (MRSA). Time-kill assays demonstrated that pelgipeptin D exhibited rapid and effective bactericidal action against MRSA at 4×MIC. Based on acute toxicity test, the intraperitoneal LD50 value of pelgipeptin D was slightly higher than that of the structurally related antimicrobial agent polymyxin B. Pelgipeptins are highly potent antibacterial and antifungal agents, particularly against MRSA, and warrant further investigation as possible therapeutic agents for bacteria infections resistant to currently available antibiotics.
