        Histone deacetylase 6 interference protects mice against experimental strokeinduced brain injury via activating Nrf2/HO-1 pathway

        Cerebral stroke is a fatal disease with increasing incidence. The study was to investigate the role and mechanism of Histone deacetylase 6 (HDAC6) on experimental stroke-induced brain injury. The recombinant shRNA-HDAC6 or scramble shRNA lentivirus was transfected to ICR mice. Then, the ischemia/reperfusion injury (I/RI) mice were constructed using middle cerebral artery occlusion (MCAO) method. Brain TTC staining was used to determine infarct areas. Serum levels of oxidative stress-related factors were detected by enzyme linked immunosorbnent assay (ELISA). Realtime-qPCR (RT-qPCR) and Western blot were used to respectively detect mRNA and protein levels. HDAC6 was up-regulated in brain I/RI mice (MCAO group), and down-regulated again in MCAO mice transfected with shRNA-HDAC6 (MCAO + shRNA group). The infarct area of the MCAO mice was increased, neurological scores were higher, and serum protein levels of 3-NT, 4- HNE and 8-OHdG were higher. HDAC6 interference attenuated above effects. Though protein levels of Nrf2 and HO-1 in cytoplasm increased slightly in MCAO group, they increased significantly by HDAC6 interference. The protein levels of Nrf2 in cytoblast decreased significantly in MCAO group, and increased markedly by HDAC6 interference. HDAC6 interference protected mice against experimental stroke-induced brain injury via Nrf2/HO-1 pathway.

        Determination of benzo[a]pyrene in camellia oil via vortexassisted extraction using the UPLC-FLD method

        In this paper, vortex-assisted extraction using the ultraperformance liquid chromatography analysis method was performed to determine benzo[a]pyrene in camellia oil. Optimum results were obtained when 0.5 g of oil sample was used followed by vortex-assisted extraction for 10 min with 25 mL of acetonitrile. Chromatographic separation was performed on an Agilent ZORBAX Eclipse Plus C18 column (2.1mm×100mm, particle size 1.8 μm). The optimum mobile phase comprised 70% acetone and 30% water. The detection limit of benzo[a]pyrene was 0.2 μg/kg. The recoveries were in the range of 81.0–97.0%. The proposed method was simple and fast, and it provided high throughput in the determination of benzo[a]pyrene in an oil matrix sample.

      • Effect of control route on the unstart/restart characteristics of an over-under TBCC inlet

        Numerical simulations have been conducted to study the unstart/restart characteristics of an over-under turbine-based combined-cycle propulsion system (TBCC) inlet during the inlet transition phase. A dual-solution area exists according to the Kantrowitz theory, in which the inlet states may be different even with the same input parameters. The entire transition process was divided into five stages and the unstart/restart hysteresis loop for each stage was also obtained. These loops construct a hysteresis surface which separates the operating space of the engine into three parts: in which a) inlet can maintain a started state; b) inlet keeps an unstarted state; c) inlet state depends on its initial state. During the transition, the operation of the engine follows a certain order with different backpressures and splitter angles, namely control route, which may result in disparate inlet states. Nine control routes with different backpressures and transition stages were designed to illuminate the route-dependent behavior of the inlet. The control routes operating towards the unstart boundary can make the inlet transit from a started state into an unstarted one. But operating backward the same route cannot make the inlet restart, additional effort should be made.

        Isolation, Identification, and Potential Probiotic Characterization of One Lactococcus from Kefir Grain

        “Probiotic” refers to viable microorganisms that promote or support a beneficial balance of the autochthonous microbial population of the gut. In the present study, we isolated and identified one potential probiotic of Lactococcus lactis WH-C1 from Tibetan Kefir grain. We assessed its probiotic and fermentation properties. WH-C1 showed good tolerance in the condition of high bile salt concentration and acidity. Moreover, WH-C1 could remove cholesterol from the culture medium at a percentage of 31.23% during 24 h and it had high capability of exopolysaccharide production. As part of starter culture it gave the better quality yogurt with good mouth feeling and a great viscosity. The solid-phase microextraction-gas chromatography-mass spectrometry analysis showed that fermented milk had 25 kinds of flavor compounds including acids, esters, alcohols, carbonyls, diacetyl, etc. In conclusion, our results showed that WH-C1 is a very promising strain which can be used in various fields.

        Clinical significance and prognostic value of C-reactive protein/albumin ratio in gastric cancer

        Purpose: This study was aimed to evaluate the clinical significance and prognostic value of CRP/albumin ratio (CAR) in patients with gastric cancer. Methods: The data of 205 gastric cancer patients who underwent surgery was analyzed retrospectively. The association of CAR with the clinical features and prognostic value in gastric cancer was analyzed. The data of this study was combined with previous studies to further determine the prognostic value of CAR in patients with gastric cancer using a metaanalysis method. Results: Cox analysis revealed that preoperative CAR was an independent prognosis indicator in patients with gastric cancer. High expression of CAR indicated a shorter survival time than in those with lower expression. CAR has a higher prognostic value in the 1-, 3-, and 5-year overall survival in patients with gastric cancer. CAR showed significant difference regarding the gastric cancer patients’ age, M stage, and clinical stage. The discriminate value of CAR in M stage of gastric cancer was high (area under the curve, 0.809). A meta-analysis combining previous data and our data showed that preoperative CAR demonstrated a significant association with the overall survival of patients with gastric cancer. Conclusion: This study demonstrated that preoperative CAR could serve as an important prognostic indicator in patients with gastric cancer.
