        Study on the Impact of Positioning Errors on the Process Performance of Robotic Bonnet Polishing

        High-precision optical components are widely used in major fields such as strong lasers and astronomy, but the production cycle of components is greatly constrained by the difficulty of machining optical components and the high price of special polishing machines, so it is important to develop a high-efficiency and low-cost fast polishing system for optical components. By combining bonnet polishing technology with industrial robotics, we have developed an industrial robotics bonnet polishing system for optical components, and the effect of robot positioning errors on pad trimming and on the polishing of the entire surface is also analyzed. To verify the processing capability of the robotic bonnet polishing device, polishing pad dressing experiments and optical component surface correction polishing experiments were carried out. After the pad was dressed, the runout value was reduced from 182 to 23 μm with a convergence ratio of 7.9. After polishing the optical component twice, the PV and RMS values on the surface of the component decreased significantly, from 38.05 λ and 9.98 λ to 8.65 λ and 1.38 λ (λ = 632.8) respectively in the middle area of the component, with a convergence ratio of 4.4 and 7.2, proving that the robotic bonnet polishing system can be applied to the polishing process of optical components.

        CO2-responsive smart wormlike micelles based on monomer and “pseudo” gemini surfactant

        CO2-responsive wormlike micelles based on monomer erucic acid 3-(N,N-dimethylamino)propylamide (EADP) with or without sodium salicylate (NaSal) in aqueous solution were studied and compared with “pseudo” gemini system. The “pseudo” gemini system were formed by a mixture of EADP and maleic acid (MA) with molar ratio between 2:1–1:1. Different morphological transformations of microstructures in response to EADP concentrations, additives and CO2 stimuli were further investigated in these systems, the wormlike micelle or vesicles can be obtained. The mechanism of these transitions were further investigated by rheology, cryo-TEM and DFT calculation, the results showed that the mechanism behind this transition involved reversible reaction of CO2 with EADP. The mechanism in this research may give further evidence to the self-assembly of CO2-responsive surfactants.

        Biological Characteristics of Recombinant Arthrobotrys oligospora Chitinase AO-801

        Chitinase AO-801 is a hydrolase secreted by Arthrobotrys oligospora during nematode feeding, while its role remained elusive. This study analyzed the molecular characteristics of recombinant chitinase of Arthrobotrys oligospora (reAO-801). AO-801 belongs to the typical glycoside hydrolase 18 family with conserved chitinase sequence and tertiary structure of (α/β)<sub>8</sub> triose-phosphate isomerase (TIM) barrel. The molecular weight of reAO-801 was 42 kDa. reAO-801 effectively degraded colloidal and powdered chitin, egg lysate, and stage I larval lysate of Caenorhabditis elegans. The activity of reAO-801 reached its peak at 40℃ and pH values between 4-7. Enzyme activity was inhibited by Zn2<sup>+</sup>, Ca2<sup>+</sup>, and Fe3<sup>+</sup>, whereas Mg2<sup>+</sup> and K<sup>+</sup> potentiated its activity. In addition, urea, sodium dodecyl sulfate, and 2-mercaptoethanol significantly inhibited enzyme activity. reAO-801 showed complete nematicidal activity against C. elegans stage I larvae. reAO-801 broke down the C. elegans egg shells, causing them to die or die prematurely by hatching the eggs. It also invoked degradation of Haemonchus contortus eggs, resulting in apparent changes in the morphological structure. This study demonstrated the cytotoxic effect of reAO-801, which laid the foundation for further dissecting the mechanism of nematode infestation by A. oligospora.

        Low METTL3 level in midgut of the Bombyx mori inhibit the proliferation of nucleopolyhedrovirus

        N6-methyladeosine (m 6 A) plays an important role in virus infection and replication. Bombyx mori nuclear polyhedrosis is caused by Bombyx mori nucleopolyhedrovirus (BmNPV) infection. Expression levels of m 6 Amodification-related genes after the infection of BmNPV were detected at first. Then, expression levels of BmNPV nucleocapsid protein gene VP39 and envelope fusion protein gene GP64 after knockdown of METTL3in vitro were quantified to identify the effect of m 6 A modification on BmNPV. BmNPV firstly infects the larval midgut in case of oral infection. Subsequently, to clarify the relationship between m 6 A modification and resistance of the silkworm to BmNPV, we detected the expression levels of m 6 A-modification-related genes invivo before and after infection of BmNPV. The results indicated that low METTL3 level hindered the proliferation of BmNPV to some extent, and silkworm strain with low METTL3 level showed stronger resistance against BmNPV. This study will accumulate new experimental data for elucidating the resistance mechanism of silkworm against BmNPV.
