        Three-dimensional Printed Mg-Doped β-TCP Bone Tissue Engineering Scaffolds: Effects of Magnesium Ion Concentration on Osteogenesis and Angiogenesis In Vitro

        BACKGROUND: Three-dimensional (3D) printed bone tissue engineering scaffolds have been widely used in research and clinical applications. b-TCP is a biomaterial commonly used in bone tissue engineering to treat bone defects, and its multifunctionality can be achieved by co-doping different metal ions. Magnesium doping in biomaterials has been shown to alter physicochemical properties of cells and enhance osteogenesis. METHODS: A series of Mg-doped TCP scaffolds were manufactured by using cryogenic 3D printing technology and sintering. The characteristics of the porous scaffolds, such as microstructure, chemical composition, mechanical properties, apparent porosity, etc., were examined. To further study the role of magnesium ions in simultaneously inducing osteogenesis and angiogenesis, human bone marrow mesenchymal stem cells (hBMSCs) and human umblical vein endothelial cells (HUVECs) were cultured in scaffold extracts to investigate cell proliferation, viability, and expression of osteogenic and angiogenic genes. RESULTS: The results showed that Mg-doped TCP scaffolds have the advantages of precise design, interconnected porous structure, and similar compressive strength to natural cancellous bone. hBMSCs and HUVECs exhibit high proliferation rate, cell morphology and viability in a certain amount of Mg2?. In addition, this concentration of magnesium can also increase the expression levels of osteogenic and angiogenic biomarkers. CONCLUSION: A certain concentration of magnesium ions plays an important role in new bone regeneration and reconstruction. It can be used as a simple and effective method to enhance the osteogenesis and angiogenesis of bioceramic scaffolds, and support the development of biomaterials and bone tissue engineering scaffolds.

        Spontaneous transposition of HzSINE1 into CYP321A2 is undetectable in the field populations of Helicoverpa zea

        The enrichment of transposable elements (TEs) within allelochemical- and insecticide-metabolizing P450 alleles in Helicoverpa zea enables these P450s to gain TE-introduced adaptive variations otherwise not readily available to cope with the ever-changing and diverse xenobiotic stress factors in varying cropping systems. The critical role of each TE-inserted P450 allele depends on whether the inserted P450 allele is more adaptive than its TE-free counterpart or not. Previous study has reported a HzSINE1-inserted CYP321A2 allele in a laboratory strain of H. zea reared with xenobiotic-free artificial diets. Here we show that the HzSINE1-inserted CYP321A2 allele transcribes into two HzSINE1 sequence-containing mutant mRNA isoforms of different length that encode an identical C terminus-truncated and heme-binding region deleted non-functional P450. Nonetheless, HzSINE1 insertion does not disrupt the regulatory functional aspect of CYP321A2 since this allele is constitutively expressed and highly inducible by the allelochemicals xanthotoxin, quercetin and chlorogenic acid. Furthermore, while the HzSINE1-inserted CYP321A2 allele is fixed in the laboratory strain, the insertion is purged in the bifenthrin-resistant strain and the Georgia field population of H. zea. To sum up, the HzSINE1-inserted CYP321A2 allele represents an allelochemical-inducible non-functional P450 allele that is selected against in the field populations frequently encountering toxic plant allelochemicals and synthetic insecticides. However, such an insertion can reach fixation under the xenobiotic-free laboratory rearing conditions most likely due to random genetic drift across multiple generations.

        Outcomes of epidural steroids following percutaneous transforaminal endoscopic discectomy: a meta-analysis and systematic review

        Background: Percutaneous transforaminal endoscopic discectomy (PTED) has been widely used in the treatment of lumbar degenerative diseases. Epidural injection of steroids can reduce the incidence and duration of postoperative pain in a short period of time. Although steroids are widely believed to reduce the effect of surgical trauma, the observation indicators are not uniform, especially the long-term effects, so the problem remains controversial. Therefore, the purpose of this paper was to evaluate the efficacy of epidural steroids following PTED. Methods: We searched PubMed, Embase, and the Cochrane Database from 1980 to June 2021 to identify randomized and non-randomized controlled trials comparing epidural steroids and saline alone following PTED. The primary outcomes included postoperative pain at least 6 months as assessed using a visual analogue scale (VAS) and the Oswestry Disability Index (ODI). The secondary outcomes included length of hospital stay and the time of return to work. Results: A total of 451 patients were included in three randomized and two nonrandomized controlled trials. The primary outcomes, including VAS and ODI scores, did not differ significantly between epidural steroids following PTED and saline alone. There were no significant intergroup differences in length of hospital stay. Epidural steroids were shown to be superior in terms of the time to return to work (P < 0.001). Conclusions: Intraoperative epidural steroids did not provide significant benefits, leg pain control, improvement in ODI scores, and length of stay in the hospital, but it can enable the patient to return to work faster.
