        Assembly of the Superconducting Tokamak EAST

        EAST is a new machine, Experimental Advanced Superconducting Tokamak, being built in CASIPP. Steady-state operation is a main mission for this machine and the superconducting magnets were all poloidal field and toroidal field systems, constituting an engineering mission to establish the technology basis of chosen to be full superconducting Tokamak to investigate a future fusion device. The parameters of EAST are: the main radius is 1.7 m and the minor radius is 0.4 m, with a maximum toroidal magnetic field of 3.5 Tesla at the plasma center. The plasma current is 1 MA, the pulse length is 1000 s, elongation is about 1.6 . 2.0, and triangularity is about 0.4 . 0.6. The EAST device has a large number of components to be assembled at the main building since June 18, 2003. These include: support structure, cryostat, thermal shields, vacuum vessel, toroidal field magnets, poloidal field magnets, current leads and joints, cooling tubes and insulators, first wall and other auxiliary systems. In this device the required assembly should maintain gaps and insulation blocks between toroidal field magnets, thermal shields and vacuum vessel. It was required to meet the tolerances for accurate positioning of the components in the radial, toroidal, poloidal and vertical directions, to ensure that the machine satisfied the requirements and to minimize subsequent corrective operations. This paper presents a brief description of the device in terms of design, fabrication, testing and assembly.

        High expression of ring finger protein 115 contributes to the progression of colorectal cancer via WNT/β-catenin pathway

        Background Colorectal cancer (CRC) is one of the most common malignancies worldwide, with the second highest incidence among men and the third incidence among women. Objective To uncover the expression of RNF115 in human colorectal cancer (CRC) tissues and cell lines, investigate its effects on CRC progression, and further explore the mechanisms. Results TCGA database showed the high transcript per million (TPM) in human CRC tissues and its relationship with poor prognosis. RNF115 was highly expressed in CRC tissues and cell lines. RNF115 contributed to the growth of CRC cells via mediating cell cycle and colony formation. RNF115 knockdown also stimulated the apoptosis of CRC cells by increasing Bax and cleaved caspase-3 expression levels. Mechanistically, RNF115 affected the proliferation and apoptosis via targeting WNT/β-catenin pathway in CRC cells. Conclusion RNF115 was highly expressed in human CRC tissues and cell lines and correlated with poor prognosis. RNF115 contributed to the growth of CRC cells via mediating cell cycle and colony formation, and also induced the apoptosis of CRC cells via targeting WNT/β-catenin pathway. RNF115 could serve as a target for CRC treatment.

        Self-Tuning Position Control for the Linear Long-Stroke, Compound Switched Reluctance Conveyance Machine

        This paper proposes a long-stroke linear switched reluctance machine (LSRM) with a primary and a secondary translator for industrial conveyance applications. The secondary one can translate according to the primary one so that linear compound motions can be achieved. Considering the fact that either one translator imposes a time-variant, nonlinear disturbance onto the other, the selftuning position controllers are implemented for the proposed linear compound machine and experimental results demonstrate that the absolute steady-state error values can fall into 0.03 mm and 0.05 mm for the secondary and primary translator, respectively. A composite absolute precision of less than 0.6 mm can be achieved under the proposed control strategy.

        Intracellular Localization and Sustained Prodrug Cell Killing Activity of TAT-HSVTK Fusion Protein in Hepatocelullar Carcinoma Cells

        Gene therapy with nonviral vectors using the suicide gene/prodrug activating system of herpes simplex virus type-1 thymidine kinase (HSV1-TK)/ganciclovir (GCV) is inefficient in killing malignant tumor cells due to two major factors: (a) an unsatisfactory bystander effect; (b) short-lived expression of the protein. To study the capacity of the protein transduction domain (PTD) of HIV-1 TAT protein to enhance HSV1-TK/GCV cancer gene therapy, we constructed three fusion proteins TAT-TK, TK-TAT and TK. TATTK retained as much enzyme activity as TK, whereas that of TK-TAT was much lower. TAT-TK can enter HepG2 cells and much of it is translocated to the nucleus. The transduced HepG2 cells are killed by exogenously added GCV and have bystander effects on untransduced HepG2 cells. Most importantly, the introduced recombinant protein is stable and remains functional for several days at least, probably because nuclear localization protects it from the cytoplasmic degradation machinery and provides access to the nuclear transcription machinery. Our results indicate that TAT fusion proteins traffic intercellularly and have enhanced stability and prodrug cell killing activity. We conclude that TAT has potential for enhancing enzyme prodrug treatment of liver cancers.

        Microphysical Structures of an Extreme Rainfall Event Over the Coastal Metropolitan City of Guangzhou, China: Observation Analysis with Polarimetric Radar

        A record-breaking nocturnal rainfall event (543 mm in 16-h) under weak synoptic forcing occurred in the metropolitan city of Guangzhou, China, during 6–7 May 2017. The evolution and microphysical structures of this torrential rainfall event are investigated using S-band polarimetric radar datasets. The torrential rainfall concentrated in two cores: one over Huadu District (HD) in which the storms were initiated between urban areas and mountains at mid-night, and the other over Huangpu and Zengcheng District (ZC) which was characterized by locally triggered storms merging with the storms from HD. The two heavy precipitation regions show some similarities, including strong reflectivity factor for horizontal polarizations (ZH) magnitude, low centroid cumulonimbus structures, and column shape of differential reflectivity (ZDR). But obvious differences can also be viewed between them. Compared to HD, ZC has higher precipitation intensity, longer precipitation duration, and larger accumulated rainfall. Besides, ZC also has a relatively lower ZDR value of ~ 0.2 dB and a higher specific differential phase (KDP) of approximately ~ 0.35° km− 1, which indicates the larger population of medium-sized rain droplet and higher water content in ZC. The radar-retrieved drop size distributions (DSDs) (i.e., mass-weighted diameter, logarithmic normalized intercept, and liquid water content) show that small size particles and high particle number concentration are more obvious in the storm over ZC. Combined with the retrieved DSDs, the merger process brings more medium-sized raindrops to ZC, and increases the possibility of raindrop growth via the accretion of cloud water by rain, which leads to enhancement of precipitation. In addition, strong KDP may be a good indicator of intensity for extreme precipitation.

        Understanding soybean (Glycine max) domestication and improvement at a genetic level is important to inform future efforts to further improve a crop that provides the world's main source of oilseed. We detect 230 selective sweeps and 162 selected copy number variants by analysis of 302 resequenced wild, landrace and improved soybean accessions at >11× depth. A genome-wide association study using these new sequences reveals associations between 10 selected regions and 9 domestication or improvement traits, and identifies 13 previously uncharacterized loci for agronomic traits including oil content, plant height and pubescence form. Combined with previous quantitative trait loci (QTL) information, we find that, of the 230 selected regions, 96 correlate with reported oil QTLs and 21 contain fatty acid biosynthesis genes. Moreover, we observe that some traits and loci are associated with geographical regions, which shows that soybean populations are structured geographically. This study provides resources for genomics-enabled improvements in soybean breeding.
