Case Report : Retroperitoneal Fibrosis in a Patient with Rheumatoid Arthritis

A 54-year-old male diagnosed with rheumatoid arthritis (RA) was effectively treated with methotrexate and adalimumab. He was admitted with fatigue and right lower back pain which had persisted for 1 month. An enhanced abdominal computed tomography scan showed an ill-defined mass with soft tissue attenuation surrounding the right common iliac artery involving the right middle portion of the ureter. Laparoscopic ureterolysis and biopsy were performed. Microscopic evaluation confirmed the presence of fibroblastic proliferation, with a pleomorphic inflammatory cell infiltrate consisting predominantly of lymphocytes, macrophages, and vascular endothelial cells, without granuloma or neoplastic cells. Therefore, our diagnosis was retroperitoneal fibrosis (RPF)-associated RA. Clinicians should consider the possibility of RPF in patients with RA who experience lower back pain, abdominal pain, or dysuria, and order suitable imaging studies. (J Rheum Dis 2015;22:391-394)
The Side-Chain Effects on Photovoltaic Properties of Copolymers Based on Phenothiazine and Thiophene

<P>In this study, the alternating conductive polymers based on phenothiazine and bithiophene unit, poly[(N-10-dodecyl-phenothiazin-3,7-ylene)-alt-(2,2’-bithiophen-5-yl)] (P1) and poly[(N-10- dodecyl-phenothiazin-3,7-ylene)-alt-(4,4’-didodecyl-2,2’-bithiophen-5-yl)] (P2), were synthesized by Suzuki coupling reaction in the presence of palladium catalyst. The structures of synthesized compounds were confirmed by 1H NMR. The optical, electrochemical properties of synthesized polymers were investigated by UV-Vis, cyclic voltammetry (CV). The optical band gap of 2.22 eV and 2.50 eV was obtained from absorption onset of UV-Vis spectrum. Then, the two devices using blends of the polymer, as a donor, and PC71BM, as an acceptor, were fabricated by spin-coating. The device with a P1:PC71BM (1:4, w/w) as an active layer exhibited the best performance with an open circuit voltage (VOC) of 0.68 V, short circuit current (JSC) of 3.5 mA/cm2, fill factor (FF) of 31.8 % and power conversion efficiency (PCE) of 0.74 %.</P>

<P>We compared a real time-nucleic acid sequence-based amplification (RTi-NASBA) with conventional NASBA, galactomannan enzyme immunosorbent assay (GM-EIA), and Mycology Study Group of the European Organization for Research and Treatment of Cancer (EORTC/MSG) criteria for the diagnosis of invasive aspergillosis (IA). From May 2004 to May 2005, blood samples (314 in total) were collected twice a week from 78 patients with hematologic diseases during neutropenic fever after chemotherapy or hematopoietic stem cell transplantation. Results were compared with each other on the basis of EORTC/ MSG criteria. The cutoff of conventional NASBA was set to be 3.5; GM 0.5; RTi-NASBA, 20% above the negative control. There were 22 patients with IA (7 probables and 15 possibles) and 56 patients with nonfungal infection. The Kappa statistic for RTi-NASBA versus conventional NASBA was 0.80 (0.66-0.82; <I>p</I><0.001) indicating that there was fairly good accordance between two tests. RTi-NASBA showed sensitivity 0.96, specificity 0.43, positive- and negative-predictive value 0.40 and 0.96, respectively. GM showed good specificity (0.98), while the sensitivity (0.45) was poor. When we use the combination of GM with either of two NASBAs, the sensitivity was improved up to 100%. In conclusion, RTi-NASBA could be a good alternative to the conventional one for the screening of IA.</P>

<P>It has been reported that the SW480 human colon cancer cell line consists of E-type and R-type cells. The long-term tumorigenic potential, invasive and metastatic properties of these subclones have not been characterized. E-type and R-type cells were subcloned using limiting dilution methods from parental SW480 cells. The cell growth rate was determined by MTT colorimetric assay, and colony forming efficiency was analyzed using Matrigel-coated plates. The activity of matrix metalloproteinase (MMP) and of urokinase plasminogen activator (uPA) was assessed by zymography. Invasive and locomotive ability was analyzed using transwell chambers. In situ apoptosis detection of these subclones was also performed. In vivo long-term tumorigenicity and nodal metastasis were evaluated using nude mice. E-type cells produced spontaneously regressive tumors in spite of invasion and lymph node metastasis. In contrast, R-type cells revealed progressively growing tumors without invasion or metastasis. E-type cells exhibited increased apoptosis and invasive and motile ability, as well as strong MMP-9 and -2 activity. Although phorbol 12-myristate 13-acetate treatment induced MMP-9 activity in E-type cells, it had no effect on R-type cells. These findings suggest that E- and R-type cells may have different biological properties in terms of colon cancer progression, regression, invasion and nodal metastasis, and might serve as a useful model for these studies.</P>

<P><B>Abstract</B></P><P>Emerging evidence suggests that not only β-amyloid but also other amyloid precursor protein (APP) fragments, such as the β-C-terminal fragment (βCTF), might be involved in Alzheimer's disease (AD). Treatment of neurons with okadaic acid (OA), a protein phosphatase-2A inhibitor, has been used to induce tau phosphorylation and neuronal death to create a research model of AD. In this study, we analyzed axonopathy and APP regulation in cultured rat neurons treated with OA. After OA treatment, the neurons presented with axonal swellings filled with vesicles, microtubule fragments, and transport molecules such as kinesin and synapsin-I. Western blotting showed that intracellular APP levels were increased and immunocytochemistry using antibodies against the APP C-terminus showed that APP accumulated in the axonal swellings. This APP C-terminus immunoreactivity disappeared when neurons were cotreated with a β-secretase inhibitor, but not with α- or γ-secretase inhibitors, indicating that the accumulation was primarily composed of APP-βCTF. These findings provide the first evidence that APP-βCTF can accumulate in the axons of OA-treated neurons, and may suggest that APP-βCTF is involved in the pathogenesis of AD.</P>

<P>OBJECTIVES: This study tested whether sodium para-amino salicylic dihydrate, an antibacterial drug for tuberculosis, could block manganese-induced apoptosis in SK-N-MC neurons. METHODS: Cell viability, Hoechst staining, dichlorofluorescin diacetate analysis for reactive oxygen species measurement, and immunoblotting were performed. KEY FINDINGS: In vitro, manganese chloride significantly decreased the viability of SK-N-MC cells, accompanied by apoptotic features such as changes in nuclear morphology. Sodium para-amino salicylic dihydrate inhibited these apoptotic characteristics through reducing intracellular reactive oxygen species generation, protecting mitochondrial membrane potential and caspase-3 activation. CONCLUSIONS: Sodium para-amino salicylic dihydrate inhibits manganese-induced apoptosis in neurons and may reduce manganese-mediated neurodegeneration.</P>

Vimentin is a growth-related gene and often expressed when epithelial cells are stimulated to proliferate by growth factors. In cancer, vimentin expression is associated with a dedifferentiated malignant phenotype, increased motility, invasive ability and poor prognosis. We studied the regulation of vimentin mRNA and multistep invasion processes following treatment of 12-O-tetradecanoylphorbol 13-acetate (TPA) and all-trans-retinoic acid (RA) in Hep 3B hepatocellular carcinoma cells. TPA showed marked induction of vimentin mRNA, while RA decreased the mRNA level. TPA or RA did not affect cell proliferation, cell-matrix prolein adhesion, and matrix metalloproteinases and urokinase plasminogen activator activities. In vitro invasion ability was significantly increased or decreased with TPA or RA treatment, paralleled to the in vitro motile activity, respectively. These findings suggest that TPA and RA could modulate the invasive potential of Hep 3B cells by altering cellular motility related to differential regulation of vimentin mRNA.
Unraveling Oxygen Transfer at the Graphene Oxide–ZnO Nanorod Interface

<P>ZnO nanorods (NRs) were grown on graphene oxide (GO) with hydrothermal method and their structural, optical, and electrical properties were compared with those of ZnO NRs grown on quartz without GO. The enhancement of the crystallinity and the reduction of the oxygen vacancies of ZnO NRs on GO (ZnO/GO) were observed. TEM/EDS observation demonstrated the increase of oxygen concentration in ZnO NR by growing on GO due to the diffusion from the GO during solution synthesis process. From the conductivity measurement of a ZnO NR, however, the conductivity of ZnO NR on GO turned out to be much lower than that of ZnO NR on quartz. This confirms that diffused oxygen from GO fills the oxygen vacancies inside ZnO NRs, and that it leads to a decrease of the conductivity that is caused by the reduction of the charge-carrier concentration in ZnO NRs.
