A cDNA encoding a cysteine protease inhibitor (CPI) was isolated from the cDNA library of clamworm Perinereis aibuhitensis Grube. The deduced amino acid sequence analysis showed that the protein had 51%, 48%, and 48% identity with Zgc: 153129 from Danio rerio, cystatin B from Theromyzon tessulatum, and the ChainA, stefin B tetramer from Homo sapiens, respectively. The gene was cloned into the intracellular expression vector pET-15b and expressed in Escherichia coli. The recombinant CPI (PA-CPI) was purified by affinity chromatography on Nicharged resin and ion-exchange chromatography on DEAE-Sepharose FF. The relative molecular mass of PACPI was 16 kDa as deduced by SDS-PAGE. Activity analysis showed that the recombinant protein could inhibit the proteolytic activity of papain. A constitutive and secretive expression vector was also constructed, and the cDNA encoding CPI was subcloned into the vector for extracellular expression. Western blotting analysis results showed that the PA-CPI was secreted into the medium. Bioassay demonstrated that E. coli DH5α harboring pUC18ompAcat-CPI showed a significant difference in mortality to the Asian longhorned beetle Anoplophora glabripennis compared with untransformed E. coli DH5α and control.

Patients at the same pathological stage of esophageal cancer (EC) that received the same surgical therapy by the same surgeon may have distinct prognoses. The current study aimed to explore the possibility of differentially-expressed microRNAs (miRNAs) underlying this phenomenon. Samples were collected from EC patients at the same tumor node metastasis (TNM) stage but with different prognoses. Paracancerous normal tissues were taken as controls. The specimens were histopathologically analyzed. Differentially-expressed miRNAs were analyzed using real-time quantitative reverse transcription polymerase chain reaction. Compared with patients with poor prognosis, those with good prognosis exhibited 88 two-fold or more than two-fold increased miRNA fragments and 4 half-decreased miRNAs. The most noticeably up-regulated miRNAs included hsa-miR-31, hsa-miR-196b, hsa-miR-652, hsa-miR-125a-5p, hsa-miR-146b, hsa-miR-200c, hsa-miR-23b, hsa-miR-29a, hsa-miR-186, hsa-miR-205, hsa-miR-376a, hsa-miR-410, hsa-miR-532-3p, and hsa-miR-598, whereas the most significantly-downregulated miRNAs were hsa-let-7e, hsa-miR-130b, and hsa-miR-103. EC patients at same TNM stage but with different prognoses show differentially-expressed miRNAs.
Analysis of Pressure Distribution of the Bolted Joints Based on the Finite Element Method

Bolted joints widely exist in the field of the national defense industry. However, its nonlinear stiffness degradation will occur under tangential load which may lead to the reliability reduction. According to Coulomb friction theory, the pressure distribution is the key in the tangential degradation of bolted joints. Therefore, this paper aims to analyze the pressure distribution of bolted joints. Firstly, we established a threaded bolt model and two simplified models. Secondly, the accuracy of the three models describing the contact area is verified, and the influence of preload and material properties on the contact radius is analyzed. Thirdly, we compared the pressure distribution of the three models, and results show that the smooth bolt model is more suitable for pressure analysis. Finally, the accuracy of several different functions to characterize the pressure distribution of bolted joints is analyzed, and results indicated that the Fernlund function is optimal. This paper provides a feasible simplified bolt model for finite element analysis, and the optimal pressure distribution function can be applied in the tangential stiffness degradation modeling.

This paper presents the virtual mass method to implement the prediction of total resistance for ice-going ship in floe ice region based on the combined method of CFD and DEM. Two ways of floe ice distribution are adopted for the analysis and comparison. The synthetic ice model test has been conducted to determine the optimal virtual mass coefficients for the two different floe ice distributions. Moreover, the further verification and prediction are developed in different ice conditions. The results show that, the fixed and random distributions in numerical method can simulate the interaction of ship and ice vividly, the trend of total resistance varying with the speed and ice concentration obtained by the numerical simulation is consistent with the model test. The random distribution of floe ice has higher similarity and better accuracy than fixed distribution.

Although a large number of AroA enzymes (EPSPS: 5-enopyruvylshikimate-3-phosphate synthase) have been identified, cloned, and tested for glyphosate resistance, only two AroA variants, derived from Agrobacterium tumefaciens strain CP4 and Zea mays, have been utilized to produce the commercial glyphosate-resistant crops. Here, we have used a PCR-based twostep DNA synthesis method to synthesize an aroA gene (aroAA. metalliredigens) from Alkaliphilus metalliredigens, encoding a new EPSPS. Furthermore, transgenic Arabidopsis with the new aroAA. metalliredigens gene was obtained to confirm the potential of the novel aroA gene in developing glyphosate-resistant crops.

The microemulsion formulation (hereafter formulation) of curcuma oil and its acaricidal efficacy against Tetranychus cinnabarinus Boisduval (Acari: Tetranychidae) were optimized in the laboratory to evaluate their spray effectiveness of oviposition inhibition and repellence. Ethovision XT6 was used to analyse the effects of the sublethal concentrations (LC 20 ) of curcuma oil and the formulation on the behaviors of T. cinnabarinus. The results showed that Tween-80 was the best surfactant, Isopropanol was the best co-surfactant and K m = 2:1 was the best condition for the formulation. The prepared microemulsions are stable under conditions of centrifugation and incubation for extended periods. The results showed that the effect of the spray bioassays of the formulation against T. cinnabarinus continuously increased during the experiment, but for curcuma oil almost no longer increase observed when the exposure time went beyond 24 h. Moreover, compared with curcuma oil (LC 50 = 0.716%), the spray bioassay of the formulation (LC 50 = 0.035%) was stronger against T. cinnabarinus. The repellency of the formulation to T. cinnabarinus was stronger with increasing exposure time, but that of curcuma oil declined after 12 h of exposure. The mobile distance of T. cinnabarinus treated with the formulation continuously declined during the experiment but that due to the curcuma oil almost no longer declined when the treatment time reached 12 h. The maximum mobile frequency of T. cinnabarinus treated by curcuma oil and the formulation was recorded at 6 h and 12 h, respectively. Thus, the formulation is a promising candidate as a botanical acaricide of green vegetables.

The phase separationphenomena or cloud points (CP) behaviorof nonionic octylphenol polyoxyethylene etherwith oxyethylene glycol ether unit of 15 (OP-15) were investigated in aqueous micellar solutions containinghigh concentrations of inorganic salts including NaCl, KCl, MgCl2, CaCl2, BaCl2, AlCl3, FeCl3, Na2SO4and Na2SiO3. The change in CP induced by salts and its mechanism are discussed theoretically. In aqueous solutionscontaining high concentration of salt, for thefirst time the linear relationship between CP and saltconcentrationiswellestablishedtocharacterizethedepressionofCP.Basedonpseudophaseseparationmodel,thermodynamic parameters are estimated and adopted to further describe the phase separation process andthe interactions between OP-15 molecules or the OP-15/water interactions. Thermodynamic parametersincluding standard Gibbs energy change (DG0CP), enthalpy change (DH0CP) and entropy change (DS0CP)indicate that the additions of different salts result in the change in entropy-driven nonspontaneousprocess of phase separation. These data, which are from the compensation temperatures and the changeof molar heat capacity, also prove that different ions induce the change in the interactions betweensurfactant molecules or the surfactant–water interaction and thus affect the depression of CP. Thesefindings help with understanding the effect of inorganic salts, especially the high concentration of salt, onCP of nonionic surfactant and then broadening the application of nonionic surfactant based on CP such asthe extraction of biomaterials in biological samples derived from animals or plants, etc.
Involvement of MicroRNA-198 Overexpression in the Poor Prognosis of Esophageal Cancer

Objective: This study aimed to investigate whether the miR-198 expression level is related to clinicopathological factors and prognosis of esophageal cancer. Methods: MicroRNA was extracted from esophageal cancer patients who underwent surgery for assessment using the Taqman@ MicroRNA assay. The correlation between miR-198 expression and clinicopathological features was analyzed, and the significance of miR-198 as a prognostic factor and its relationship with survival was determined. Results: MicroRNA-198 (miR-198) expression was higher in patients with poor prognosis than those with good prognosis (P<0.05). Kaplan-Meier analysis results showed that the miR-198 expression level had a significant correlation with survival time (P=0.030) and that patients with a higher expression of miR-198 had a shorter survival time. Cox multi-factor model analysis showed that patient prognosis (P=0.014), tumor length (P=0.040) and expression (P=0.012), and survival time had a significant correlation; the corresponding risks were 7.268, 1.246, and 3.524, respectively. Conclusion: miR-198 overexpression is involved in the poor prognosis of esophageal cancer and can be used as a biomarker for selection of cases requiring especial attention.
