        Imaging Myocardial Ischemia and Reperfusion Injury via Cy5.5-Annexin V

        Aim The aim of this article is to present the results of an imaging study of myocardial apoptosis induced by ischemia/reperfusion injury. Methods Twenty nude mice were randomly divided into an experimental group (10 mice) and control group (10 mice). In the experimental group, myocardial apoptosis was induced by ligation of the left anterior descending coronary artery (LAD) for 30 min. This was followed by reperfusion for 90 min. In the control group, the heart was exposed for the same length of time as in the experimental group. Cy5. 5-annexin V (25 ${\mu}g$) was injected into both sets of mice after the onset of reperfusion. At 90 min post-injection, the mice were imaged. The region of interest (ROI) was obtained, and the fluorescence intensity of the ROI was quantified. The animals were sacrificed, and myocardial apoptosis was assayed by TUNEL assay. Results Fluorescence intensity in the ischemia/reperfusion hearts was significantly higher than that in the control group (P<0.05). In the TUNEL assay, more apoptotic cells were observed in the experimental group than in the control group, correlating with imaging results. Conclusion Fluorescence imaging of Cy5.5-annexin V in a mouse model of myocardial ischemia/reperfusion can be used in vivo as a noninvasive means of detecting ischemia/reperfusion-induced apoptotic cells in the heart.

        A fundamental trend of processor architecture evolving towards exaflops is fast increasing floating point performance (so-called "free" flops) accompanied by much slowly increasing memory and network bandwidth. In order to fully enjoy the "free" flops, a numerical algorithm of PDEs should request more flops per byte or increase arithmetic intensity. A meshfree/GFEM approximation can be the class of the algorithm. It is shown in a GFEM without extra dof that the kind of approximation takes advantages of the high performance of manycore GPUs by a high accuracy of approximation; the "expensive" method is found to be reversely hardware-efficient on the emerging architecture of manycore.

      • Retrospective Evaluation of Discrepancies between Radiological and Pathological Size of Hepatocellular Carcinoma Masses

        Background: The size of a hepatic neoplasm is critical for staging, prognosis and selection of appropriate treatment. Our study aimed to compare the radiological size of solid hepatocellular carcinoma (HCC) masses on magnetic resonance imaging (MRI) with the pathological size in a Chinese population, and to elucidate discrepancies. Materials and Methods: A total of 178 consecutive patients diagnosed with HCC who underwent curative hepatic resection after enhanced MRI between July 2010 and October 2013 were retrospectively identified and analyzed. Pathological data of the whole removed tumors wereassessed and differences between radiological and pathological tumor size were identified. All patients were restaged using a modified Tumor-Node-Metastasis (TNM) staging system postoperatively according to the maximum diameter alteration. The lesions were classified as hypo-staged, iso-staged or hyper-staged for qualitative assessment. In the quantitative analysis, the relative pre and postoperative tumor size contrast ratio ($%{\Delta}size$) was also computed according to size intervals. In addition, the relationship between radiological and pathological tumor diameter variation and histologic grade was analyzed. Results: Pathological examination showed 85 (47.8%) patients were overestimated, 82 (46.1%) patients underestimated, while accurate measurement by MRI was found in 11 (6.2%) patients. Among the total subjects, 14 (7.9%) patients were hypo-staged and 15 (8.4%) were hyper-staged post-operatively. Accuracy of MRI for calculation and characterized staging was related to the lesion size, ranging from 83.1% to 87.4% (<2cm to ${\geq}5cm$, p=0.328) and from 62.5% to 89.1% (cT1 to cT4, p=0.006), respectively. Overall, MRI misjudged pathological size by 6.0 mm (p=0.588 ), and the greatest difference was observed in tumors <2cm (3.6 mm, $%{\Delta}size=16.9%$, p=0.028). No statistically significant difference was observed for moderately differentiated HCC (5.5mm, p=0.781). However, for well differentiated and poorly differentiated cases, radiographic tumor maximum diameter was significantly larger than the pathological maximum diameter by 3.15 mm and underestimated by 4.51 mm, respectively (p=0.034 and 0.020). Conclusions: A preoperative HCC tumor size measurement using MRI can provide relatively acceptable accuracy but may give rise to discrepancy in tumors in a certain size range or histologic grade. In pathological well differentiated subjects, the pathological tumor size was significantly overestimated, but underestimated in poorly differentiated HCC. The difference between radiological and pathological tumor size was greatest for tumors <2 cm. For some HCC patients, the size difference may have implications for the decision of resection, transplantation, ablation, or arterially directed therapy, and should be considered in staging or selecting the appropriate treatment tactics.

        Establishment of a novel myocarditis mouse model based on cyclosporine A

        Background: Myocarditis is a myocardial injury that can easily cause adolescent death. Traditional research models of animal invasion with viral components, lipopolysaccharide (LPS) or porcine myocardial myosin, among others, have the shortcomings of potential biological safety hazards and high animal mortality. Objective: To explore the construction of a novel myocarditis model with cyclosporine A and the potential genes and pathways associated with it. Methods: BALB/c mice were used in this study, and cyclosporin A and LPS were injected into the peritoneal cavity of mice. The successful establishment of the model was assessed by detecting serum myocardial injury markers and inflammatory factors levels, HE, IHC staining, and RT-qPCR methods. Key genes were obtained using the GSE35182 dataset from the GEO database and validated with the RT-qPCR method. Results: We found that a large number of inflammatory cells infiltrated the myocardium of mice in each group of Cyclosporin A constructed model, while the expression of inflammatory factor indicators was increased, and this model has the characteristics of high degree of local inflammation in myocardial tissue, low mortality, and safe and non-toxic treatment. Using GSE35182 data, we selected 18 Hub genes and validated Hub genes in myocardial tissue with RT-qPCR and found that multiple signaling pathways such as Toll-likereceptor signaling pathway(TLRs), Rap1 signal pathway(Rap1), and Chemokine signaling pathway may be involved in the development of myocarditis. Conclusion: Cyclosporin A can construct a new myocarditis model, and TLRs, Chemokines and Rap1 signaling pathways may be the core pathways of myocarditis.

        Temperature effect on seismic behavior of transmission tower-line system equipped with SMA-TMD

        Transmission tower-line system is one of most critical lifeline systems to cities. However, it is found that the transmission tower-line system is prone to be damaged by earthquakes in past decades. To mitigate seismic demands, this study introduces a tuned-mass damper (TMD) using superelastic shape memory alloy (SMA) spring for the system. In addition, considering the dynamic characteristics of both tower-line system and SMA are affected by temperature change. Particular attention is paid on the effect of temperature variation on seismic behavior. In doing so, the SMA-TMD is installed into the system, and its properties are optimized through parametric analyses. The considered temperature range is from -40 to 40°C. The seismic control effect of using SMA-TMD is investigated under the considered temperatures. Interested seismic performance indices include peak displacement and peak acceleration at the tower top and the height-wise deformation. Parametric analyses on seismic intensity and frequency ratio were carried out as well. This study indicates that the nonlinear behavior of SMA-TMD is critical to the control effect, and proper tuning before application is advisable. Seismic demand mitigation is always achieved in this wide temperature range, and the control effect is increased at high temperatures.

        Novel blood-based hypomethylation of SH3BP5 is associated with very early-stage lung adenocarcinoma

        Background: Early detection is essential to improve the survival of lung cancer (LC). The quantitative measurement of specific DNA methylation changes in the peripheral blood could provide an efficient strategy for the detection of early cancer. Objective: We applied a candidate approach and assess the association between blood-based SH3BP5 methylation and the risk of lung adenocarcinoma (LUAD) in a case-control cohort. Methods: The methylation level of four CpG sites in the promoter of SH3BP5 gene was quantitatively determined by mass spectrometry in 171 very early-stage LUAD patients (93.6% LUAD at stage I) and 190 age and gender-matched controls. The logistic regression and non-parametric tests were used for the statistical analyses. Results: We observed a significant association between decreased methylation of SH3BP5_CpG_4 in the peripheral blood and increased risk of LUAD (odds ratio (OR) per-10% methylation = 1.51, P = 0.006, FDR = 0.024), and even for the LUAD at stage I (OR per-10% methylation = 1.53, P = 0.006, FDR = 0.024). Moreover, the lower quartile of SH3BP5_CpG_4 methylation was correlated with increased risk for LUAD with a P trend of 0.011. Further investigation disclosed that the hypomethylation of SH3BP5_CpG_4 was mostly associated with LUAD in younger subjects (OR per-10% methylation = 2.02, P = 0.010, age < 55 years old) and probably could be enhanced by advance stage. Conclusion: Our study revealed an association between blood-based SH3BP5 hypomethylation and very early-stage LUAD, which provides a novel support for the blood-based methylation signatures as a potential marker for the evaluation of cancer risk.

        Seismic response control of transmission tower-line system using SMA-based TMD

        This study proposes a new shape memory alloy-tuned mass damper (SMA-TMD) and investigates the effectiveness of this damper in reducing and controlling the vibrations of a transmission tower-line system under various seismic excitations. Based on a practical transmission line system and considering the geometric nonlinearity of this system, the finite element (FE) software ANSYS is used to create an FE model of the transmission tower-line system and simulate the proposed SMA-TMD. Additionally, the parameters of the SMA springs are optimized. The effectiveness of a conventional TMD and the proposed SMA-TMD in reducing and controlling the vibrations of the transmission tower-line system under seismic excitations is investigated. Moreover, the effects of the ground motion intensity and frequency ratio on the reduction ratio (η) of the SMA-TMD are studied. The vibration reduction effect of the SMA-TMD under various seismic excitations is superior to that of the conventional TMD. Changes in the ground motion intensity and frequency ratio have a significant impact on the η of the SMA-TMD. As the ground motion intensity and frequency ratio increase, the η values of the SMA-TMD first increase and then decrease. Studying the vibration reduction effects of the SMA-TMD can provide a reference for the practical engineering application of this damper.

        Gene cloning and functional analysis of yellow green leaf 3 (ygl3) gene during the whole-plant growth stage in rice

        Chlorophyll is an important photosynthetic pigment in the process of photosynthesis in plants and photosynthetic bacteria. Genes involved in chlorophyll biosynthesis in Arabidopsis and photosynthetic bacteria have been well documented. In rice, however, these genes have not been fully annotated. In this paper, a yellow-green leaf gene, yellow green leaf3 (ygl3) was cloned and analyzed. ygl3 encodes magnesium chelation ChlD (D) subunit,a key enzyme for chlorophyll synthesis, resulting in a yellow-green leaf phenotype in all growth stages in rice. Expression content of ygl3 is highest in the leaf blades,followed by the leaf sheaths, while there is virtually no expression of the gene in the stems and seeds. The subcellular structure and protein content of the photosynthetic system of the ygl3 mutant were revealed by transmission electron microscopy, BN-PAGE, and western blotting. The results show that the mutation of the ygl3 gene indirectly leads to a decrease in the protein content of the photosynthetic system and severely obstructs the formation of granum thylakoids.
