The Future of Holiness and Pentecostal Churches: Bringing Belief in Line with Practice

This article argues that the Holiness and Pentecostal traditions could offer the theological hermeneutics for interpreting Wesleyan traditions and even the theological heritages from Lutheran and Reformed Churches, based on the study of Dr. Donald Dayton. Churches ought to champion their distinctive contributions to Christianity in church history by bringing their beliefs to be in line with their practices. The theological perspectives of the Holiness-Pentecostal traditions are the most varied kinds--the Christocentric Fourfold Gospel (A.B Simpson), the Weselyan Fourfold Gospel, the Four-Square Gospel, the Fivefold Gospel, and the Full Gospel. To theologically analyze the Holiness and Pentecostal traditions, the fourfold gospel could be used as a representation of a better paradigm than those used for the description of the Continental Reformation.

Background/Aims Wireless motility capsule (WMC) detects the ileocolic junction (ICJ) in most non-operated patients. We find no data concerning this examination in patients where the ileocolic valve is replaced by a per definition incompetent, surgically created ICJ. We wanted to see if WMC could detect the ICJ after a right colectomy and assess the competency. Methods Prospective cohort study using a within-subject design to eliminate subject-subject variability. Selected patients operated with right colectomy underwent 3 WMC examinations (pre-operatively, 3 weeks, and 6 months after surgery). Results Twenty patients (8 men) included, 7 (4 men) excluded due to poor recordings (4) and unforeseen events (3). Thirteen patients (4 men), median age 63 years completed 3 tests. Median bowel lengths removed were 11 cm for ileum and 21 cm for colon. Thirty-nine examinations analyzed by 2 physicians who found all 13 ICJs at 3 examinations with high inter-rater reliability (intra-class correlation coefficient: 0.99, 0.91, and 0.99 respectively), whereas the computer found 9, 8, and 10 out of the 13 ICJs, respectively. Computed values significantly more often deviated from the 2 raters. Mean magnitude and duration of pH-drop at the ICJ (3 examinations) was 1.16-1.02-1.13 pH units and 3.15-4.78-3.75 minutes, respectively. pH-drop was smaller and duration longer at 3 weeks. We found no differences between the pre-operative (competent ICJ) and post-operative 6-month examinations (incompetent ICJ). Highest pressure immediately prior to ICJ was equal before and after surgery. Conclusion WMC can identify the non-physiological ICJ after right colectomy. Ileocolic competence cannot be assessed. The Direct Instruction Effects on the Improvement of Reading Literacy within a RtI Framework

The purpose of this study was to investigate the implementation effects of directinstruction reading intervention within a RtI framework. Using DIBELS Next as theuniversal screener for CBM, eight first grade students identified as below or well belowbenchmark and confirmed to be at risk for reading difficulties by their classroom teachers. Supplemental direct instruction began early in the semester(October), following the directmethod for beginning Tier 2 support. Intervention sessions occurred within the classroomsfor a minimum of 40 minutes, two times a week. EPACC and RM-FC were the Tier 2interventions implemented in addition to the core curriculum. Intervention sessions weretaught by a faculty researcher as the principal investigator (PI). Outcomes from the end ofthe year first grade DIBELS Next were compared to the grade-level means for the sample. Individual student scores indicated that students progressed and some improved during theintervention periods. Implications for pre-service teachers and teacher educators werediscussed.

<P>G protein-coupled receptors (GPCRs) have important roles in physiology and pathology, and 40% of drugs currently on the market target GPCRs for the treatment of various diseases. Because of their therapeutic importance, the structural mechanism of GPCR signaling is of great interest in the field of drug discovery. Hydrogen/deuterium exchange mass spectrometry (HDX-MS) is a useful tool for analyzing ligand binding sites, the protein-protein interaction interface, and conformational changes of proteins. However, its application to GPCRs has been limited for various reasons, including the hydrophobic nature of GPCRs and the use of detergents in their preparation. In the present study, we tested the application of bicelles as a means of solubilizing GPCRs for HDX-MS studies. GPCRs (e.g., β<SUB>2</SUB>-adrenergic receptor [β<SUB>2</SUB>AR], μ-opioid receptor, and protease-activated receptor 1) solubilized in bicelles produced better sequence coverage (greater than 90%) than GPCRs solubilized in n-dodecyl-β-D-maltopyranoside (DDM), suggesting that bicelles are a more effective method of solubilization for HDX-MS studies. The HDX-MS profile of β<SUB>2</SUB>AR in bicelles showed that transmembrane domains (TMs) undergo lower deuterium uptake than intracellular or extracellular regions, which is consistent with the fact that the TMs are highly ordered and embedded in bicelles. The overall HDX-MS profiles of β<SUB>2</SUB>AR solubilized in bicelles and in DDM were similar except for intracellular loop 3. Interestingly, we detected EX1 kinetics, an important phenomenon in protein dynamics, at the C-terminus of TM6 in β<SUB>2</SUB>AR. In conclusion, we suggest the application of bicelles as a useful method for solubilizing GPCRs for conformational analysis by HDX-MS.</P> [FIG OMISSION]</BR>

Objective: Coenzyme (CoQ10) is an enzymatic co factor used in normal cellular metabolism. Recent evidence shows that in people with heart disease it can reverse endothelial cell damage in the blood vessels. It is also a potent antioxidant. Design: One group pretest-posttest design. Methods: In the present study, endothelial function was evaluated using the response to occlusion and heat before and 2 weeks after administration of CoQ10, 300 mg/day. Thirty Eight subjects, who are physical therapy students, participated in a series of experiments to see if taking 300 mg of CoQ10 daily for 2 weeks would impact resting blood flow in the forearm skin and the blood flow response to 4 minutes of vascular occlusion and the response to local heat (42˚C) for 6 minutes. Results: The results showed that, for this population, there was no difference in the response to heat. However, the response to occlusion was improved after administration of CoQ10. Conclusions: It would appear that in a young population CoQ10 has no effect on the nitric oxide vasodilator pathway in skin but does influence other vasodilator pathways.
