      • Interaction of <i>Veratrum nigrum</i> with <i>Panax ginseng</i> against Obesity: A Sang-ban Relationship

        <P>Obesity has become a major health threat in developed countries. However, current medications for obesity are limited because of their adverse effects. Interest in natural products for the treatment of obesity is thus rapidly growing. Korean Medicine (KM) is characterized by the wide use of herbal formulas. However, the combination rule of herbal formulas in KM lacks experimental evidence. According to <I>Shennong's Classic of Materia Medica</I>, the earliest book of herbal medicine, <I>Veratrum nigrum</I> (VN) has antagonistic features against <I>Panax ginseng</I> (PG), and the PG-VN pair is strictly forbidden. In this study, we have shown the effects of PG, VN, and their combination on obesity in high-fat (HF) diet-induced obese mice and in 3T3-L1 cells. PG, VN, and PG-VN combination significantly reduced weight gain and the fat pad weight in HF diet-induced obese mice. They also significantly decreased lipid accumulation and the expressions of two major adipogenesis factors, PPAR<I><I>γ</I></I> and C/EBP<I><I>α</I></I>, in 3T3-L1 cells. In addition, the PG-VN combination had synergistic effects compared with the mixture of extracts of PG and VN on inhibition of PPAR<I><I>γ</I></I> and C/EBP<I><I>α</I></I> expressions at lower doses. These results indicate a new potential anti-obese pharmacotherapy and also provide scientific evidence supporting the usage of herbal combinations instead of mixtures in KM.</P>

        공동주택 주차장의 축적먼지 중 미량원소성분 분석과 오염원 평가

        공동주택 주차장의 먼지오염실태를 연구하기 위하여 2010년 3월말에서 6월초까지 대구지역 공동주택 36곳의 지상주차장(36지점)과 지하주차장(36지점)을 대상으로 채취한 총 72개의 시료를 100 μm 이하로 체거름하고 산추출한 후 ICP로 14개원소를 분석하였다. 농축계수를 이용한 미량원소성분의 발생원 평가결과는 지상주차장과 지하주차장 모두 Fe, K, Mg, Mn, Na, V의 성분은 자연적인 발생원에 의한 영향을 받고 있는 반면에 Cd, Cr, Cu, Ni, Pb, Zn의 성분은 인위적인 발생원에 의한 영향을 받고 있는 것으로 나타났다. 한편 Ca은 지상주차장의 경우에는 자연적인 발생원의 영향을 받았고, 지하주차장의 경우에는 인위적인 발생원의 영향을 받은 것으로 나타났다. 미량원소성분의 농도분석 결과는 자연적인 발생원과 관련된 성분이 인위적인 발생원과 관련된 성분보다 매우 높은 농도를 보였다. 인위적인 발생원성분의 기여도는 지하주차장이 지상주차장보다 높았으며, 미량원소성분의 평균농도 또한 지하주차장이 지상주차장보다 높았다. 오염지수를 이용한 중금속성분의 오염도를 평가한결과는 지하주차장이 지상주차장보다 5.5배정도 높게 오염된 것으로 나타났다. 미량원소성분간의 상관성 분석결과는 지상주차장이 지하주차장에 비해 유의한 상관성을 나타내는 항목이 많았으며, 특히 자연적 발생원 성분간 유의한 상관성이 더욱 크게 나타났다. 또한 유해 중금속성분과 지하주차장의 영향인자간의 상관성을 분석한 결과는 주차대수(공간)가 적은 지점이 큰 지점보다, 환기장치를 가동하지 않은 지점이 가동한 지점보다, 그리고 최근 도색년도와 청소일이 오래된 지점이 상대적으로 유해중금속을 많이 함유하고 있었다. In order to investigate the degree of apartment parking lot dust contamination, total 72 samples of parking lot dust (36 from ground parking lots and 36 from the underground parking lots) were collected in Daegu city from the end of March to the early June 2010. The dust samples were sieved below 100 μm, and analysed by ICP for 14 elements after an acid extraction. Results obtained from the source assessment of trace element using enrichment factor showed that Fe, K, Mg, Mn, Na and V were influenced by natural sources, while Cd, Cr, Cu, Ni, Pb and Zn were influenced by anthropogenic sources in both the ground parking lot and the underground parking lot. And results showed that Ca were influenced by natural sources in the ground parking lot, but influenced by anthropogenic sources in the underground parking lot. The measured values were remarkably higher in components from natural sources than in components from anthropogenic sources. Underground parking lot dust was more affected by anthropogenic sources and contaminated compared with the ground parking lot dust. Pollution index of heavy metals revealed that underground parking lot dust was 5.5 times more contaminated with heavy metal components than the ground parking lot dust. The results of correlation analysis among trace elements indicated that components in the ground parking lot were more correlated than those in the underground parking lot, and especially more correlated with natural sources-natural sources. Analysis for correlations between components and influencing factors in the underground parking lot showed that concentrations of heavy metals were higher with smaller number of parking spaces and no ventilation system, and older apartments in last paint and cleaning had relatively higher contents of heavy metals than those of recently painted and cleaned.

        <P>The rhodium(III)-catalyzed olefination of various carboxamides with alpha-CF3-substituted allylic carbonate is described. This reaction provides direct access to linear CF3-allyl frameworks with complete trans-selectivity. In particular, a rhodium catalyst provided Heck-type gamma-CF3-allylation products via the beta-O-elimination of rhodacycle intermediate and subsequent olefin migration process.</P>

        Establishment of an in vitro culture system for chicken preblastodermal cells

        <P>To develop an alternative source for chicken pluripotent cells, we examined (1) whether undifferentiated preblastodermal cells could be subcultured in vitro for an extended period and (2) how subculturing affected the physiological properties of preblastodermal cells. The average number of preblastodermal cells was 2,397 in stage V embryos and 36,345 in stage VII embryos; stage X embryos had an average of 53,857 blastodermal cells. The average cell size decreased significantly (70.63–18.83 µm in diameter; P < 0.0001) as the embryo grew; this was closely related to a reduction in the size and number of lipid vesicles in the cell cytoplasm. The culture conditions were optimized for the stage V preblastodermal cells and the control stage X blastodermal cells. On STO feeder cells, the preblastodermal cells achieved stable growth in vitro only in HES medium or a mixed medium of the Knockout DMEM and HES media. However, more than 10 passages of preblastodermal cells at intervals of 3–4 days was possible only by using the Knockout/HES mixed medium and BRL cell-conditioned HES medium for the primary cultures and subcultures, respectively. Colony-forming preblastodermal cells had well-delineated cytoplasm, which was positively stained for stem cell-specific markers by anti-stage-specific embryo antigen-1 antibody, periodic acid-Schiff's solution, and alkaline phosphatase. When preblastodermal cells with or without culturing were transferred into the blastodermal cavity of stage X embryos, only in vitro-cultured preblastodermal cells at stage V (4/5 = 80%) and stage VII (2/8 = 25%) induced somatic chimerism in recipient chickens. In conclusion, undifferentiated preblastodermal cells could be subcultured, and only the colony-forming preblastodermal cells that stained positively for stem cell markers could induce somatic chimerism.         <P>We have demonstrated a very compact eight-channel 200-GHz-spacing multiwavelength laser (MWL) module for the optical line terminal source of wavelength-division-multiplexing passive optical network. The wavelength shift of MWLs and external cavity lasers from the ITU grid was within +0.14 nm for eight channels, which was well matched to the target wavelength shift of less than plusmn0.2 nm. The oscillation characteristics of our MWL module are better than those of conventional MWLs with hybrid integration.</P>

      • Effects of acclimation and pH on ammonia inhibition for mesophilic methanogenic microflora

        <P><B>Abstract</B></P> <P>This work evaluated the toxicity of ammonia toward mesophilic methanogenic microflora, with respect to the acclimation and pH. Microflora acclimated to total ammonia nitrogen (TAN) concentrations of 1.0–2.5 g N/L and neutral to alkaline pH, 7.5–8.0, were used for anaerobic toxicity assays (ATA) with TAN up to 6.1 and pH ranged from 7 to 8.2. The effect of ammonia on the specific methanogenic activity (SMA) and methane yield (MY) was described using a noncompetitive model. Acclimation increased the tolerance of the methanogenic microflora to ammonia over the acclimation range. There was no significant difference in the values of SMA and MY at each TAN concentration in the examined pH range, although free ammonia nitrogen (FAN) concentrations increased along with pH. It implied that TAN rather than FAN would be the useful criterion for ammonia inhibition. Propionate degradation was more inhibited compared to acetoclastic methanogenesis by ammonia. The half-maximal inhibitory concentrations (<I>IC</I> <SUB>50</SUB>) of TAN for the SMA from acetate, SMA from propionate, MY from acetate, and MY from propionate were 3.42–4.26, 3.14–3.91, 3.67–4.07, and 3.34–3.88 g N/L, respectively, at pH 7.4. 454-pyrosequencing analysis of the inoculum showed that the archaeal community was dominant of <I>Methanosarcinaceae</I> and <I>Methanobacteriaceae</I>, which were known as tolerant to ammonia inhibition.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Acclimation increased the tolerance to ammonia inhibition over the acclimated range. </LI> <LI> Total ammonia rather than free ammonia would be the direct inhibition criterion. </LI> <LI> Propionate degradation was more sensitive to NH<SUB>3</SUB> than acetoclastic methanogenesis. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        CD4+ TH1 Cells Generated by Ii-PADRE DNA at Prime Phase Are Important to Induce Effectors and Memory CD8+ T Cells

        The requirement for CD4 T cells in priming and maintaining cytotoxic T-lymphocyte responses presents a long-standing paradox in cellular immunology. In this study, we used sequential coadministration of a DNA vaccine encoding an invariant (Ii) chain in which the class II-associated Ii-peptide region is replaced with CD4 T-helper epitope, PADRE [Pan human leukocyte antigen-DR reactive epitope (Ii-PADRE)] or Bcl-xL with a DNA vaccine encoding Sig/E7/LAMP-1 to verify the role of CD4 T cells for the generation of effectors and memory E7-specific CD8 T-cell immune responses. Sequential vaccination, with Ii-PADRE+Sig/E7/LAMP-1 priming followed by Bcl-xL+Sig/E7/LAMP-1 boosting led to generation of E7-specific CD8 T cells, and was nearly equivalent in effect to coadministration with Ii-PADRE+Sig/E7/LAMP-1 or Bcl-xL+Sig/E7/LAMP-1 at both prime and boost. The mice vaccinated with the Ii-PADRE+Sig/E7/LAMP-1 prime-Bcl-xL+Sig/E7/LAMP-1 boost regimen exhibited better long-term E7-specific immune responses and tumor prevention effects in vivo than the mice vaccinated with the reverse sequential coadministration. After CD4 T-cell depletion, mice primed with Ii-PADRE+Sig/E7/LAMP-1 generated low numbers of E7-specific CD8 T cells and suppressed long-term memory CD8 T-cell response regardless of the sequence or combination of DNA vaccines administered. Mice primed with Bcl-xL+Sig/E7/LAMP-1 only suppressed long-term memory CD8 T-cell response after depletion of CD4 T cells before priming. Our findings suggest that activated CD4 T cells at prime phase are important to generate the antigen-specific CD8 T-cell immune responses and CD4 T cells, which are naive or activated, play a role to maintain the long-term memory responses.

        Simultaneous determination of the metabolites of the herbicide metazachlor in agricultural crops by LC–MS/MS

        To manage the safety of the herbicide metazachlor, analytical methods are required for the determination of metazachlor metabolites in agricultural crops. Herein, a liquid chromatography–tandem mass spectrometry (LC–MS/ MS) method was developed for the simultaneous determination of metazachlor metabolites (479M04, 479M08, and 479M16) in various agricultural commodities. After extraction using acetonitrile and adjusting the pH to 3, the samples were purified using a hydrophilic–lipophilic balance cartridge. The matrix-matched calibration curves (0.002–0.2 μg/mL) were linear (r2 > 0.99). For validation, recovery tests were carried out at three fortification levels (limit of quantification (LOQ), 10 LOQ, and 50 LOQ) in various agricultural samples. The recoveries of 479M04, 479M08, and 479M16 were 79.6–113.0, 76.9–97.7, and 79.1–102.1%, respectively, with relative standard deviation values of less than 17.0%. Furthermore, inter-laboratory testing was conducted to validate the method. All the values corresponded to the criteria of both the CODEX (CAC/GL 40-1993, 2003) and Ministry of Food and Drug Safety guidelines. Therefore, the proposed LC–MS/MS method can be used as an analytical method for the determination of metazachlor.
