<P>The PKA-CREB signaling pathway is involved in many cellular processes including autophagy. Recent studies demonstrated that PKA-CREB inhibits autophagy in yeast; however, the role of PKA-CREB signaling in mammalian cell autophagy has not been fully characterized. Here, we report that the integral membrane protein ITM2A expression is positively regulated by PKA-CREB signaling and ITM2A expression interferes with autophagic flux by interacting with vacuolar ATPase (v-ATPase). The <I>ITM2A</I> promoter contains a CRE element, and mutation at the CRE consensus site decreases the promoter activity. Forskolin treatment and PKA expression activate the <I>ITM2A</I> promoter confirming that ITM2A expression is dependent on the PKA-CREB pathway. ITM2A expression results in the accumulation of autophagosomes and interferes with autolysosome formation by blocking autophagic flux. We demonstrated that ITM2A physically interacts with v-ATPase and inhibits lysosomal function. These results support the notion that PKA-CREB signaling pathway regulates ITM2A expression, which negatively regulates autophagic flux by interfering with the function of v-ATPase.</P>

<P>The extracellular signal-regulated kinases/mitogen activated protein kinase (ERK/MAPK) and nuclear factor-κB (NF-κB) pathways are critical for cell survival and proliferation. Alpinumisoflavone (AIF), isolated from the African medicinal plant <I>Erythrina lysistemon</I>, is a member of the isoflavone group. In this report, we demonstrated that AIF treatment induces cell death of human lung tumor cells. Incubation of lung tumor cells with AIF increased the sub-G1 population and caspase 3/7 activity, suggesting that the cell death is caused by apoptosis. To identify the signaling pathway involved in the tumor cell death, we examined the modulation of transcriptional activity using various reporter constructs and found that AIF significantly deregulated both the ERK/MAPK and NF-κB pathways. Western blot analysis with antibodies to MAP/ERK kinase (MEK) and ERK showed that AIF dephosphorylates both MEK and ERK. Alpinumisoflavone also repressed lipopolysaccharide (LPS)-induced nitric oxide (NO) production in RAW264.7 cells by inhibiting NF-κB-dependent transcription. Therefore, the cell death induced by AIF may be <I>via</I> repressing both the ERK/MAPK and NF-κB pathways.</P>

Autophagy is one of the major degradative mechanisms that can eliminate excessive nutrients, toxic protein aggregates, damaged organelles and invading microorganisms. In response to obesity and obesity-associated lipotoxic, proteotoxic and oxidative stresses, autophagy plays an essential role in maintaining physiological homeostasis. However, obesity and its associated stress insults can often interfere with the autophagic process through various mechanisms, which result in further aggravation of obesity-related metabolic pathologies in multiple metabolic organs. Paradoxically, inhibition of autophagy, within specific contexts, indirectly produces beneficial effects that can alleviate several detrimental consequences of obesity. In this minireview, we will provide a brief discussion about our current understanding of the impact of obesity on autophagy and the role of autophagy dysregulation in modulating obesity-associated pathological outcomes.

Dynamic flow characteristics of a solenoid valve are affected by pressure difference in inlet and outlet of orifice, gas temperature, and supply voltage of a coil. In this paper, the dynamic flow characteristics for deviations of various conditions are studied Static and dynamic flow for variation on-time of a solenoid valve open signal are measured in basic bench test. The solenoid valve is applied to a compressed natural gas(CNG) engine test for validation of flow control performance. The experimental results show that flow of high pressure gas can controlled precisely by using a solenoid valve.

In this paper, an engine clutch engagement shock is analyzed during mode change of plug-in hybrid electric vehicles. Multi-driving mode includes EV (electric vehicle) mode, HEV (hybrid electric vehicle) mode and engine operating mode. According to mode change, the engine clutch is engaged or disengaged. A magnitude of shock during clutch engagement is very important because it depends on the vehicle acceleration and clutch synchronizing speed, which affects a ride comfort dominantly. The performance simulator of plug-in hybrid electric vehicle is developed using MATLAB/Simulink. The simulation results show that it needs the mode change control algorithm to minimize a shock during engaging the clutch.

플러그인 하이브리드 전기자동차는 내연기관과 전기모터를 동력원으로 사용하며 주행 상황에 따라 다양한 주행 모드을 갖는다. 주행 모드에는 전기모터로만 주행하는 EV 모드(전기주행), 내연기관으로 주행하는 엔진 운전 모드, 두 개의 동력원을 이용하는 HEV 모드(하이브리드 주행)가 있다. 특히 병렬형 구조를 갖는 하이브리드 전기자동차는 모드변환에 따라 엔진 클러지가 접합되거나 해제되는데, 클러치 접합 시 나타나는 충격은 차량의 승차감에 영향을 주기 때문에 중요하다. 본 논문에서는 플러그인 하이브리드 전기자동차의 성능 시뮬레이터를 MATLAB/Simulink를 이용하여 개발하고, 시뮬레이션 결과를 통해 엔진 클러치 접합 시 나타나는 충격 특성을 분석하였다. The plug-in hybrid electric vehicle has a high fuel economy and can be driven long distances. Its different modes include the electric vehicle, hybrid electric vehicle, and only engine operating mode. A power management strategy is important to determine which mode should be selected. The strategy makes the vehicle more efficient using appropriate power sources for driving. However, the strategy usually needs a driving speed profile which is future driving cycle. If the profile is known, the strategy easily determines which mode is driven efficiently. However, it is difficult to estimate the speed profile for a real system. To address this problem, this paper proposes a new power distribution strategy using a neural network. The average speed and driving range are used as input parameters to train the neural network system. The strategy determines a limit for the use of the battery and the desired power is distributed between the engine and the motor simultaneously. Its fuel economy can increase by improving the basic strategy.
Stress granules dynamics: benefits in cancer

Stress granules (SGs) are stress-induced subcellular compartments,which carry out a particular function to cope with stress. Thesegranules protect cells from stress-related damage and cell deaththrough dynamic sequestration of numerous ribonucleoproteins(RNPs) and signaling proteins, thereby promoting cell survivalunder both physiological and pathological condition. Duringtumorigenesis, cancer cells are repeatedly exposed to diversestress stimuli from the tumor microenvironment, and the dynamicsof SGs is often modulated due to the alteration of geneexpression patterns in cancer cells, leading to tumor progressionas well as resistance to anticancer treatment. In this mini review,we provide a brief discussion about our current understandingof the fundamental roles of SGs during physiological stress andthe effect of dysregulated SGs on cancer cell fitness and cancertherapy.

Dry clutch has compact structure, which consists of clutch disk and electric actuator. It doesn’t need lubricant that is necessary for a wet clutch. Therefore, it shows better efficiency as well as fuel economy compared to the wet clutch. However, it is sensitive to the actuating position so it requires deliberate control. If control strategy is not accurate, the wear of the clutch increases and it results in shorter clutch life. Dry dual clutch transmission (DCT) which is composed of twin clutches is the main component in this study. DCT enables smooth shift by preselecting gear-synchronizer. When the shift occurs, engine torque transfers from the off-going clutch to the on-coming clutch. After shift process is completed, angular velocity of engine shaft is synchronized with that of the clutch. When the synchronization occurs, engine throttle is controlled. With the engine control algorithm, crank shaft of engine synchronizes faster than without control and shift quality improves.
