To construct transfurmed Bm5 cells, Autographa californica nuclear polyhedrosis virus(AcNPV) IE1 gene, an immediate early viral gene was firstly used in this study. AcNPV IE1 gene, which shares on 95.3% nucleotide sequence homology with Bombyx mori nuclear polyhedrosis virus (BmNPV) IE1 gene, was isolated and cloned into pBluescript. Neomycin gene from pKO-neo was inserted under the control of the IE1 promoter to yield pAcIE1-neo. The plasmid pAcIE1-neo was transfected into Bm5 or Sf9 cells, and neomycin-resistant cells were selected in TC100 medium containing 10% fetal bovine serum (FBS) and 1 mg/ml G4l8 for two weeks. Individual clones were picked and each was amplified for further characterization. The genomic DNA from neomycin-resistant cells was isolated and characterized by PCR using AcNPV IEI gene-specific primers and by Southern blot analysis using neomycin gene probe. We concluded that AcNPV IE1 gene was functional in B. mori-derived Bm5 cells as well as Spodoptera fugjprrda-derived Sf9 cells to produce stably-transformed insect cells

<B>Summary</B><P>Interspecies intracytoplasmic sperm injection has been carried out to understand species-specific differences in oocyte environments and sperm components during fertilization. While sperm aster organization during cat fertilization requires a paternally derived centriole, mouse and hamster fertilization occur within the maternal centrosomal components. To address the questions of where sperm aster assembly occurs and whether complete fertilization is achieved in cat oocytes by interspecies sperm, we studied the fertilization processes of cat oocytes following the injection of cat, mouse, or hamster sperm. Male and female pronuclear formations were not different in the cat oocytes at 6 h following cat, mouse or hamster sperm injection. Microtubule asters were seen in all oocytes following intracytoplasmic injection of cat, mouse or hamster sperm. Immunocytochemical staining with a histone H3-m2K9 antibody revealed that mouse sperm chromatin is incorporated normally with cat egg chromatin, and that the cat eggs fertilized with mouse sperm enter metaphase and become normal 2-cell stage embryos. These results suggest that sperm aster formation is maternally dependent, and that fertilization processes and cleavage occur in a non-species specific manner in cat oocytes.</P>

<P>Hidden nodes may severely degrade the performance of wireless LANs. Focusing on the hidden node effect, we analyze goodput in a single cell WLAN environment with hidden nodes under a non-saturated condition. Uplink and downlink goodputs are analytically derived using an approximation technique and are compared with simulation results.</P>

The cobalt catalyzed carbonylation of bromobenzene having protected aldehyde group gives the corresponding ester in good yields, but 2-bromobenzaldehyde gives 3-alkoxyphthalide in the noticeable yield instead of alkyl 2-formylbenzoates.
A Novel Evolutionary Approach to Image Enhancement Filter Design: Method and Applications

<P>Image enhancement is an important issue in digital image processing. Various approaches have been developed to solve image enhancement problems, but most of them require deep expert knowledge to design appropriate image filters. To automatically design a filter, we propose a novel approach based on the genetic algorithm that optimizes a set of standard filters by determining their types and order. Moreover, the proposed method is able to manage various types of noise factors. We applied the proposed method to local and global image enhancement problems such as impulsive noise reduction, interpolation, and orientation enhancement. In terms of subjective and objective evaluations, the results show the superiority of the proposed method.</P>

Disease management of oak wilt, mainly occurred in Quercus mongolicae by Raffaelea quercus-mongolicae in Korea, have been mainly focused on the control of insect vector(Platypus koryoensis) by treatments of insecticide spray, pheromone trap, sticky roll trap, and fumigation of dead or dying trees. For the control the pathogen colonized in trees, culture suspension of antifungal Streptomyces blastmyceticus was injected into trees in the forest by trunk (ChemJet®) or root flare(Macro-infusion) injection methods before or after pathogen inoculation, and the ratios of non-conductive area, discolored area, and re-isolation of the pathogen from the treated trees were calculated for evaluating preventive or curative efficacy. Injection of S. blastmyceticus or the fungicide(Alamo®) showed 41.7% and 45.8% in non-conductive area(%), respectively. In addition, S. blastmyceticus did not show much differences between injection methods and timings in control effect, but fungicide treatment was better in Macro-infusion at root flare or curative treatments. Re-isolation rates of the pathogen in the pathogen only, preventive and curative treatments were 56.9%, 20.6%, and 36.1%, respectively. These results indicate that S. blastmyceticus has better control efficacy than the fungicide commercially available for the control of oak wilt caused by Ceratocystis fagacearum in the United States.

<P> Mepivacaine is an aminoamide-linked local anesthetic with an intermediate duration that intrinsically produces vasoconstriction both in vivo and in vitro. The aims of this in-vitro study were to examine the direct effect of mepivacaine in isolated rat aortic rings and to determine the associated cellular mechanism with a particular focus on endothelium-derived vasodilators, which modulate vascular tone. In the aortic rings with or without endothelium, cumulative mepivacaine concentration-response curves were generated in the presence or absence of the following antagonists: N<SUP>ω</SUP>-nitro-l-arginine methyl ester [l-NAME], indomethacin, fluconazole, methylene blue, 1H-[1,2,4]oxadiazolo[4,3-a]quinoxalin-1-one [ODQ], verapamil, and calcium-free Krebs solution. Mepivacaine produced vasoconstriction at low concentrations (1 × 10<SUP>−3</SUP> and 3 × 10<SUP>−3</SUP> mol/L) followed by vasodilation at a high concentration (1 × 10<SUP>−2</SUP> mol/L). The mepivacaine-induced contraction was higher in endothelium-denuded aortae than in endothelium-intact aortae. Pretreatment with l-NAME, ODQ, and methylene blue enhanced mepivacaine-induced contraction in the endothelium-intact rings, whereas fluconazole had no effect. Indomethacin slightly attenuated mepivacaine-induced contraction, whereas verapamil and calcium-free Krebs solution more strongly attenuated this contraction. The vasoconstriction induced by mepivacaine is attenuated mainly by the endothelial nitric oxide - cyclic guanosine monophosphate pathway. In addition, mepivacaine-induced contraction involves cyclooxygenase pathway activation and extracellular calcium influx via voltage-operated calcium channels. </P>
