        Effect of pH on Successive Foam and Sonic Droplet Fractionation of a Bromelain-invertase Mixture

        A droplet fractionation method was previously developed to concentrate a dilute nonfoaming protein solution. In that earlier study with invertase, it was demonstrated that droplets created by ultrasonic energy waves could be enriched up to 8 times that of the initial dilute invertase solution. In this study, a mixture of bromelain (a foaming protein) and invertase (a nonfoaming protein) is investigated as a preliminary step to determine if droplet fractionation can also be used to separate a non-foaming protein from foaming proteins. The foaming mixture containing bromelain is first removed by bubbling the binary mixture with air. After the foam is removed, the protein rich air-water interfacial layer is skimmed off (prior to droplet fractionation) so as not to interfere with the subsequent droplet production from the remaining bulk liquid, rich in non-foaming protein. Finally, sonic energy waves are then applied to this residual bulk liquid to recover droplets containing the non-foaming protein, presumed to be invertase. The primary control variable used in this droplet fractionation process is the pH, which ranged for separate experiments between 2 and 9. It was observed that the maximum overall protein partition coefficients of 5 and 4 were achieved at pH 2 and 4, respectively, for the initial foaming experiment followed by the post foaming droplet fractionation experiment.

        직장인의 직무 스트레스, 스트레스 대처방식 및 심리적 안녕감과의 관계

        본 연구는 직장인의 직무스트레스, 스트레스 대처방식 및 심리적 안녕감과의 관계를 알아보고자 하였으며, 연구 문제는 다음과 같다. 첫째, 직장인의 직무 스트레스, 스트레스 대처방식 및 심리적 안녕감의 관계는 어떠한가? 둘째, 직장인의 직무 스트레스, 스트레스 대처방식은 심리적 안녕감에 어떠한 영향을 미치는가? 본 연구의 결과는 다음과 같다. 첫째, 직무 스트레스와 심리적 안녕감은, 부(-)적 상관관계를 나타냈고, 직무스트레스와 스트레스 대처방식 하위요인들과의 상관관계는 모두 부(-)적 관계로 나타났다. 또한 스트레스 대처방식과 심리적 안녕감 간의 상관관계를 살펴 본 결과, 심리적 안녕감과 스트레스 대처방식 하위요인 중 문제중심적 대처방식, 사회적지지추구 대처방식, 소망적사고 대처방식은 정적인 상관관계를 나타냈으나, 정서중심적 대처 방식은 심리적 안녕감과 유의미한 상관관계를 나타내지 않았다. 둘째, 직무스트레스는 심리적 안녕감에 부적인 영향을 주는 것으로 나타났다. 직무스트레스가 안녕감에 미치는 영향에 추가적으로 스트레스 대처 방식이 영향을 미친다는 것은 스트레스 대처방식이 스트레스가 심리적 안녕감에 미치는 부정적 영향을 상쇄시킬 수 있음을 의미한다. 또한 직무스트레스와 정서중심적 대처는 심리적 안녕감에 부적으로 영향을 주었고, 문제중심적 대처는 정적으로 영향을 주었으며, 문제중심적 대처, 직무스트레스, 정서중심적 대처 순으로 심리적 안녕감을 설명하였다. The purpose of this study was to examine the relationship between job stress, stress coping strategies and psychological well-being of office workers. For this research, a survey was collected from office workers of Seoul, Icheon, and Gyeonggi-do province, and the total of 253 data was used for the analysis. The results of this study are as followed. First, correlation between job stress, stress coping strategies and psychological well-being was analyzed. job stress and psychological well-being showed a negative correlation. The sub-scales of stress coping strategies and psychological well-being showed positive correlations, except emotion focused coping strategies, which did not show a significant correlation with psychological well-being. Second, hierarchical regression was conducted to examine the effect of job stress and stress coping strategies on psychological well-being. job stress had a negative influence on psychological well-being, and sub factor of stress coping method that additionally explains psychological well-being had the variable of 19%. Also job stress and emotion-focused coping style had a negative influence on psychological well-being, but problem focused coping had a positive influence, and explained psychological well-being in order of problem focused coping, job stress, and emotion-focused coping.

      • Dominant Role of the cbb3 Oxidase in Regulation of Photosynthesis Gene Expression through the PrrBA System in Rhodobacter sphaeroides 2.4.1

        <B>ABSTRACT</B><P>In this study, the H303A mutant form of the <I>cbb</I>3 oxidase (H303A oxidase), which has the H303A mutation in its catalytic subunit (CcoN), was purified from <I>Rhodobacter sphaeroides</I>. The H303A oxidase showed the same catalytic activity as did the wild-type form of the oxidase (WT oxidase). The heme contents of the mutant and WT forms of the <I>cbb</I>3 oxidase were also comparable. However, the <I>puf</I> and <I>puc</I> operons, which are under the control of the PrrBA two-component system, were shown to be derepressed aerobically in the <I>R. sphaeroides</I> strain expressing the H303A oxidase. Since the strain harboring the H303A oxidase exhibited the same cytochrome <I>c</I> oxidase activity as the stain harboring the WT oxidase did, the aerobic derepression of photosynthesis gene expression observed in the H303A mutant appears to be the result of a defective signaling function of the H303A oxidase rather than reflecting any redox changes in the ubiquinone/ubiquinol pool. It was also demonstrated that ubiquinone inhibits not only the autokinase activity of full-length PrrB but also that of the truncated form of PrrB lacking its transmembrane domain, including the proposed quinone binding sequence. These results imply that the suggested ubiquinone binding site within the PrrB transmembrane domain is not necessary for the inhibition of PrrB kinase activity by ubiquinone. Instead, it is probable that signaling through H303 of the CcoN subunit of the <I>cbb</I><I>3</I> oxidase is part of the pathway through which the <I>cbb</I><I>3</I> oxidase affects the relative kinase/phosphatase activity of the membrane-bound PrrB.</P>

        Original Article : Relationship of Hypertropia and Excyclotorsion in Superior Oblique Palsy

        Purpose: To evaluate the correlation between hypertropia and excyclotorsion in acquired superior oblique palsy (SOP). Methods: Thirty-one patients with acquired unilateral SOP were recruited for this study. The torsional angle of each patient was assessed via one objective method (fundus photography) and two subjective methods (double Maddox rod test and major amblyoscope). The patient population was divided into two groups (concordance group, n = 19 and discordance group, n = 12) according to the correspondence between the hypertropic eye (paralytic eye) and the more extorted eye (non-fixating eye), which was evaluated by fundus photography. Results: The mean value of objective torsion was 5.09° ± 3.84°. The subjective excyclotorsion degrees were 5.18° ± 4.11° and 3.65° ± 1.93° as measured by double Maddox rod test and major amblyoscope, respectively. Hypertropia and the excyclotorsional angle did not differ significantly between the groups ( p = 0.257). Although no correlation was found in the discordance group, the concordance group showed a significant and positive correlation between hypertropia and excyclotorsion ( p = 0.011). Conclusions: Torsional deviation was not related to hypertropia. However, in the concordance patients in whom the hypertropic eye showed excyclotorsion, a significant positive correlation was found between hypertropia and excyclotorsion.

        Efficacy of Intracytoplasmic Sperm Injection for Leukocytospermia

        White blood cells (WBCs) are present in most human ejaculates, but abnormally high concentration of seminal leukocytes may reflect an underlying pathological condition. The World Health Organization (WHO) has defined leukocytospermia as status of more than $10^6$ WBC/mL of semen. The purpose of this study was firstly, to compare the outcomes between conventional IVF and ICSI in leukocytospermia, and secondly, to investigate whether ICSI may be an alternation treatment for patients with leukocytospermia. Total 346 cycles of conventional IVF and ICSI candidates underwent IVF cycles at Eulji Hospital Infertility Clinic. Semen Parameters including concentration, motility, morphology of spermatozoa and concentration of leukocytes were assessed from the raw ejaculates. There was no difference in sperm concentration, motility and morphology. The rates of fertilization and good embryo development from ICSI were significantly higher than those from conventional IVF in leukocytospermia (60.4% & 32.5%, respectively for ICSI group and 44.4% & 28.5%, respectively for IVF group, P<0.00l). The pregnancy rate after ICSI was also higher than that from conventional IVF (34.0% vs 29.1 %, P<0.05). These results indicate that the presence of seminal leukocyes (> 1$\times10^6$ WBC/mL of semen) is adversely related with fertilization, embryo development and pregnancy rate. Therfore the measurement of seminal leukocytes in routine semen analysis appears to be of prognostic value with regard to male fertilizing potential. In conclusion, it is suggested that ICSI is an alternative choice of treatment for patients with leukocytospermia.
