        Design, Synthesis and Biological Evaluation of Novel Analogs of Bortezomib

        Novel analogs of bortezomib were designed, synthesized and in vitro biological evaluation was carried out using human tumor cell lines A549 and PC3. Docking studies of these analogs of bortezomib was discussed. According to biological investigations, the inhibitors 4, 6, and 8 were found to be more potent than reference drug candidate bortezomib. A549 cell line showed significant sensitivity towards 4, 6, and 8 with $IC_{50}$ values 14.03, 18.5, and 12.4 nM, respectively, and PC3 cell line showed IC50 values 26.1, 37.0, and 21.2 nM, respectively. The $IC_{50}$ values of bortezomib in these cell lines are 27.3 nM and 42.0 nM.

        Requirement of Non-phytate Phosphorus in Synthetic Broiler Breeder Diet

        An experiment was conducted to study the laying performance, shell quality, bone mineralization, hatchability of eggs and performance of progeny (weight at day one and 14 d of age, P content in day old chick, leg score and survivability of chicks) of synthetic broiler breeders fed different levels of non-phytate phosphorus (NPP). Six levels of NPP (1.2, 1.8, 2.4, 3.0, 3.6 and 4.2 g/kg diet) at a constant calcium (Ca) level (30 g/kg) in a maize-soya-deoiled rice bran based diet were tested. Levels of dicalcium phosphate, shell grit and deoiled rice bran were adjusted to achieve the desired levels of NPP and Ca. Each level of NPP was fed with a weighed quantity of feed (160 g/b/d) to 40 female broiler breeders from 25 to 40 weeks of age housed in individual cages. Each bird was considered as a replicate. Egg production, feed/egg mass, body weight, egg weight, shell weight, shell thickness, egg specific gravity, serum Ca content and tibia breaking strength were not influenced (p>0.05) by the variation in dietary NPP levels tested. Increasing the dietary levels of NPP did not influence the hatchability of eggs, phosphorus (P) contents both in egg yolk and day old chick, chick body weight at day one and 14 d of age, leg score and survivability of chicks up to 14 d of age. Maximum response ($p{\leq}0.01$) in shell breaking strength, tibia ash and serum inorganic P contents were observed at NPP levels of 2.09, 2.25 and 3.50 g per kg diet, respectively. The retention of Ca increased, while the P retention decreased ($p{\leq}0.01$) with increasing dietary levels of NPP. Though maximum responses in shell breaking strength, bone ash and serum inorganic P were observed at NPP higher than 1.2 g/kg diet, the broiler breeder performance in terms of egg production, shell quality, hatchability of eggs and progeny performance and their survivability was not influenced by dietary NPP concentrations. It is concluded that synthetic broiler breeders maintained in cages do not require more than 1.2 g NPP/kg diet with a daily intake of 192 mg NPP/b/d during 24 to 40 weeks of age.

        Effect of Supplementing Microbial Phytase on Performance of Broiler Breeders Fed Low Non-phytate Phosphorus Diet

        An experiment was conducted to study the production performance of broiler breeder females (25 to 40 weeks of age) fed either reference diet or low non-phytate phosphorus (NPP) diet with or without microbial phytase (500 FYT/kg) supplementation. A weighed (160 g/b/d) quantity of feed from each diet was offered daily to 40 replicates of one bird each housed in California type cage having individual feeders. Each cage was considered as a replicate. A continuous 16-h light per day was provided using incandescent bulbs. Body weight, egg production, egg weight, feed per egg mass, egg specific gravity, egg breaking strength, shell thickness, tibia ash and serum Ca and protein concentrations were not affected by reducing the NPP level from 0.30 to 0.18% in the broiler breeder diet. Supplementation of phytase (500 FYT/kg) enzyme to the diet containing 0.18% NPP had no added advantage on any of the above production parameters. The serum inorganic P was increased significantly (p<0.05) by either enhancing the NPP content from 0.18 to 0.30% or supplementing phytase @500 FYT/kg to the diet containing low P which were found comparable. Retention of Ca and P was positive on all the diets. P retention decreased significantly (p<0.05) with either increase in NPP content or phytase supplementation in the diet. Neither NPP nor phytase supplementation influenced bone mineralization in terms of tibia ash and strength. The hatchability was not influenced by either increasing the NPP content or supplementing the enzyme phytase. Similarly, the P concentration in the egg yolk and day old chick, day old and 14th day body weight and leg score was not altered by increasing the level of NPP or supplementing phytase enzyme. The mortality was within the normal limits in all the three dietary groups. Thus, it can be concluded that 0.18% NPP (288 mg NPP intake/b/d) in the broiler breeder' diet is adequate in sustaining the optimum performance from 25 to 40 wks of age. Enhancing the NPP content or supplementation of phytase (500 FYT/kg diet) to diet containing 0.18% NPP had no added advantage on performance.

        NONLINEAR OPTICAL TRANSMISSION OF SURFACE-MODIFIED NICKEL SULPHIDE NANOPARTICLES: SATURATION OF ABSORPTION AND OPTICAL LIMITING

        Saturable absorbers and optical limiters have contrary optical transmission properties. We report observations of simultaneous occurrence of both these effects in a nickel sulphide nanoparticle (average dia. ~5 nm) solution and a simultaneous quantitative measurement of both. Intensity dependent nonlinear transmission studies carried out using a 7 ns Nd:YAG laser at 532 nm by the Z-scan method, revealed efficient optical limiting in nickel sulphide nanoparticle suspensions. Induced nonlinear optical scattering was identified to be the mechanism of optical limiting, and absorption at 532 nm was found to saturate. A modification of the conventional Z-scan implementation led to the retrieval of the saturation intensity which is otherwise overshadowed by very strong nonlinear scattering.

        Utilization of Graded Levels of Finger Millet (Eleusine coracana) in Place of Yellow Maize in Commercial Broiler Chicken Diets

        An experiment was conducted to study the performance, carcass traits, serum lipid profile and immune competence in commercial broilers (2 to 42 d of age) fed graded levels (25, 50, 75 and 100%) of finger millet (FM) (Elusine coracana) in place (w/w) of yellow maize (YM). Each diet was fed to eight replicates (five female Vencobb broilers/replicate) housed in stainless steel battery brooders. The estimated metabolizable energy content of FM was about 540 kcal less than the YM. FM contained more protein (10.42 vs. 9.05%) and fibre (9.52 vs. 2.24%) compared to YM. Body weight gain, ready to cook yield, relative weights of giblet, liver, intestine and length of intestine at 42 d of age was not affected due to replacing YM with FM. But, the feed efficiency decreased in broilers fed diets containing 75 and 100% FM in place of YM at both 21 and 42 d of age. The amount of fat deposited in abdominal area decreased and the relative weight of gizzard increased with increase in level of FM in the diet. The serum HDL cholesterol at 21 and 42 d of age and serum triglycerides at 42 d of age decreased with increase in level of FM in diet. The relative weight of spleen and antibody titers against sheep red blood cells (SRBC) at 5 d post inoculation (PI) decreased in broilers fed FM at 100% of YM. However, the relative weight of bursa, SRBC titers at 10 d PI, antibody titers against ND virus and mortality were not affected due to incorporation of FM in place of YM in diet. The fat content in thigh muscle and liver decreased, while the protein content in these tissues increased with increase in the level of FM in broiler diet. Based on the results, it may be concluded that YM can be replaced with FM up to 25% on weight basis without affecting weight gain, carcass yields and immunity in commercial broiler diet (up to 42 d of age). Further, inclusion of finger millet reduced the fat deposition in thigh muscle, liver and in abdominal area compared to those fed maize as the principal source of energy.

        Utilization of Low Glucosinalate and Conventional Mustard Oilseed Cakes in Commercial Broiler Chicken Diets

        An experiment was conducted to study the effect of replacing soyabean meal (SBM) at 50 and 100% with conventional (CMC) and low glucosinalate mustard cakes (LGMC) in iso-caloric and iso-nitrogenous diets in broiler chickens. All these diets contained 0.1% choline chloride with a purity of 50% (w/w). Another diet was prepared by replacing SBM in toto with CMC with no supplemental choline to find out the possible role of supplemental choline in mustard cake (MC) based diets. Two hundred and seventy day-old broiler chicks were distributed randomly in 54 stainless steel battery brooder pens of five chicks in each pen. Each experimental diet was allotted at random to nine battery brooders and offered ad-libitum from day 2 through 42 days of age. Body weight gain was significantly depressed by total replacement of SBM with either LGMC or CMC at 21 days of age. Non-supplementation of choline significantly depressed the growth compared to those fed CMC 100% with supplemental choline. However, at 42 days of age, such an effect was seen only with CMC. Replacement of SBM with CMC 100% with or without choline supplementation depressed the body weight gain. The concentrations of cholestorol and tryglicerides in serum and the relative weights of ready to cook yield, giblet and gizzard decreased by incorporation of mustard cakes in broiler diets. The trend in fat and protein contents in breast and thigh muscles and liver was not clearly attributable to the treatment effect. Based on the results, it is concluded that SBM can be replaced in toto with LGMC (535.0 and 466.5 g/kg starter and finisher diets, respectively) or up to 50% (215.0 and 186.7 g/kg starter and finisher diets, respectively) with CMC in commercial broiler chicken diets. Choline supplementation at 0.1% level in broiler diets containing CMC was found to be beneficial during starter phase.

        Replacement of Yellow Maize with Pearl Millet (Pennisetum typhoides), Foxtail Millet (Setaria italica) or Finger Millet (Eleusine coracana) in Broiler Chicken Diets Containing Supplemental Enzymes

        An experiment was conducted to study the performance of broilers chicks (2 to 42 d of age) fed diets containing pearl millet (PM, Pennisetum typhoides), foxtail millet (FOM, Setaria italica) or finger millet (FIM, Elusine coracana) totally replacing (w/w) yellow maize (YM) with and with out supplementing non-starch polysaccharide (NSP) hydrolysing enzymes at the rate of 0.5 g/kg diet. Enzyme preparation contained amylase 2,400 units, hemi-cellulase 5,400 units, cellulase 12,000 units, protease 2,400 units and beta-glucanase 106 units/g. Each diet was fed to eight replicates (five female Vencob broilers/replicate) housed in stainless steel battery brooders. The estimated metabolizable energy (ME) contents of YM, PM, FOM and FIM were FM (PM) were about 3,389, 2,736, 3,303 and 2,846 kcal/kg, respectively. Total replacement of YM with FOM did not influence the body weight gain, ready to cook yield, relative weights of giblet, liver, intestine, lymphoid organs (bursa and spleen) and length of intestine, antibody titers and livability at 42 d of age. But the food efficiency decreased significantly in FOM fed broilers compared those fed YM. Further, the fat content in thigh muscle reduced with FOM fed groups compared to those fed YM. The performance of broilers decreased significantly in PM and FIM fed broilers compared to those fed YM. The relative weights of giblet, gizzard and liver increased in FIM fed groups compared to those fed YM as the principal source of energy in broilers. Incorporation of NSP hydrolysing enzymes in commercial broiler diets improved the efficiency of feed utilization during starter phase but not at 42 d of age. The results thus indicate that yellow maize can be replaced in toto on weight basis in commercial broiler diets without affecting the performance. Supplementation of NSP hydrolysing enzymes was beneficial in enhancing feed utilization during the starter phase.

        Growth, Bone Mineralization and Mineral Excretion in Broiler Starter Chicks Fed Varied Concentrations of Cholecalciferol

        An experiment was conducted to study the growth performance, bone mineralization and mineral excretion in broiler starter chicks fed high levels of cholecalciferol (CC) at sub-optimal levels of calcium (Ca) and non-phytate phosphorus (NPP). Five hundred and sixty day-old Vencobb female broiler chicks were housed in raised wire floor stainless steel battery brooder pens ($24"{\times}30"{\times}18"$) at the rate of five chicks per pen. A maize-soyabean meal basal diet was supplemented with dicalcium phosphate, oyster shell powder and synthetic CC to arrive at two levels each of Ca (0.50 and 0.60%), and NPP (0.25 and 0.30%) and four levels of CC (200, 1,200, 2,400 and 3,600 ICU/kg) in a $2{\times}3{\times}4$ factorial design. Each diet was fed ad libitum to chicks in 7 pens from 2 to 21days of age. Body weight gain, feed intake and bone weight increased (p<0.05) with increase in level of CC at both the Ca and NPP levels tested. The CC levels required to obtain significant improvement in body weight gain and feed intake reduced (2,400 ICU/kg vs. 1,200 ICU/kg) with increase in levels of P in diet (0.25% vs. 0.3%, respectively). The feed conversion ratio was significantly improved (p<0.05) with increase in level of CC from 200 to 1,200 ICU/kg diet at 0.5% Ca, while at 0.6% Ca, the level of CC in diet did not influence the feed efficiency. Tibia mineralization (density, breaking strength and ash content) and Ca and P contents in serum increased significantly (p<0.05) with increase in levels of CC in diet. The CC effect on these parameters was more pronounced at lower levels of Ca and NPP (0.5 and 0.25%, respectively). The data on body weight gain and feed intake indicated that NPP level in diet can be reduced from 0.30 to 0.25% by increasing CC from 200 to 2,400 ICU/kg. Similarly, the bone mineralization (tibia weight, density and ash content) increased non-linearly (p<0.01) with increase in CC levels in diet. Concentrations of P and Mn in excreta decreased (p<0.01), by increasing CC level from 200 to 2,400 ICU/kg diet. It can be concluded that dietary levels of Ca and NPP could be reduced to 0.50 and 0.25%, respectively by enhancing the levels of cholecalciferol from 200 to 2,400 ICU/kg with out affecting body weight gain, feed efficiency and bone mineralization. Additionally, phosphorus and manganese excretion decreased with increase in levels of CC in broiler diet.

        Effect of Supplementing Betaine on Performance, Carcass Traits and Immune Responses in Broiler Chicken Fed Diets Containing Different Concentrations of Methionine

        An experiment was conducted with broiler (Cobb 400) male chicks (n = 480) to determine the effect of betaine (Bet) supplementation (0 and 800 mg/kg) to diets containing five concentrations (15, 18, 20, 22 and 24 g/kg crude protein, CP) of methionine (Met) in a $2{\times}5$ factorial study for performance, carcass traits, immune responses, and serum parameters. Each diet was fed ad libitum from 1 to 42 d of age to 8 replicates of 6 chicks. Birds were housed in battery brooders placed in an open-sided poultry shed. Body weight gain, feed intake, feed conversion efficiency and slaughter variables were recorded at 21 and 42 d of age. Serum biochemical profile, antibody production against Newcastle disease (ND) and lymphocyte proliferation ratio (LPR) were analysed at 42 d of age. Supplementing Bet to diets containing sub-optimal concentrations of Met (15 g/kg) improved weight gain and breast yield at 21 d of age (p<0.01), and feed conversion efficiency at 42 d of age (p<0.05). Feed efficiency at 21 d of age, body weight gain at 42 d of age, slaughter variables except breast yield at 21 d of age and ND antibody titres were not affected (p>0.05) by the interaction. LPR increased (p<0.05) with Bet supplementation at 20 g Met/kg CP equal to those broilers fed 24 g Met without Bet. Bet supplementation enhanced the concentrations of protein, globulin and cholesterol in serum of broilers fed sub-optimal concentrations of Met. Results suggested that Bet supplementation (800 mg/kg diet) enhanced growth (21 d), feed conversion efficiency (42 d), breast yield and lymphocyte proliferation in broilers fed a diet containing 15 g Met/kg CP.

        Dual Responsive Pectin Hydrogels and Their Silver Nanocomposites: Swelling Studies, Controlled Drug Delivery and Antimicrobial Applications

        Novel dual responsive pectin hydrogels composed from poly(acrylamidoglycolic acid-co-vinylcaprolactam)/ Pectin (PAV-PC) and also PAV-PC hydrogels are used as templates for the production of silver nanoparticles. 5-Fluorouracil is an anticancer drug and has been loaded in situ into PAV-PC hydrogels. Structure and morphology characterization of PAV-PC hydrogels were investigated by fourier transform infrared spectroscopy, differential scanning calorimetry, thermo gravimetric analysis, X-ray diffraction studies, scanning electron microscopy and transmission electron microscopy. The results revealed a molecular level dispersion of the drug in PAV-PC hydrogels. In vitro release of 5-fluorouracil from the PAV-PC hydrogels has been carried out in GIT fluids as well as in various temperatures. 5-Fluorouracil released from PAV-PC hydrogels was 50% at pH 1.2, and 85% at pH 7.4 within 24 h. The release profile was characterized with PAV-PC hydrogels and initial burst effect was significantly reduced in two buffer media (1.2 and 7.4), followed by a continuous and controlled release phase, the drug release mechanism from polymer was due to Fickian diffusion. In situ fabrication of silver nanoparticles inside the hydrogel network via the reduction of sodium borohydrate by PAV-PC chains led to hydrogel nanocomposites. The diameter of the nanocomposites was about 50-100 nm, suitable for uptake within the gastrointestinal tract due to their nanosize range and mucoadhesive properties. These nanocomposite PAV-PC hydrogels showed strong antimicrobial activity towards Bacillus subtilis (G+ve) and Escherichia coli (G-ve).
