Short branched-chain fatty acids (SBCFAs, C4-6) are versatile platform intermediates for the production of value-added products in the chemical industry. Currently, SBCFAs are mainly synthesized chemically, which can be costly and may cause environmental pollution. In order to develop an economical and environmentally friendly route for SBCFA production, we engineered Saccharomyces cerevisiae, a model eukaryotic microorganism of industrial significance, for the overproduction of SBCFAs. In particular, we employed a combinatorial metabolic engineering approach to optimize the native Ehrlich pathway in S. cerevisiae. First, chromosome-based combinatorial gene overexpression led to a 28.7-fold increase in the titer of SBCFAs. Second, deletion of key genes in competing pathways improved the production of SBCFAs to 387.4mg/L, a 31.2-fold increase compared to the wild-type. Third, overexpression of the ATP-binding cassette (ABC) transporter PDR12 increased the secretion of SBCFAs. Taken together, we demonstrated that the combinatorial metabolic engineering approach used in this study effectively improved SBCFA biosynthesis in S. cerevisiae through the incorporation of a chromosome-based combinatorial gene overexpression strategy, elimination of genes in competitive pathways and overexpression of a native transporter. We envision that this strategy could also be applied to the production of other chemicals in S. cerevisiae and may be extended to other microbes for strain improvement.

The purpose of this paper is to introduce and study the semigroups of nonlinear contractions in probabilistic normed spaces and to establish the Crandall-Liggett's exponential formula for some kind of accretive mappings in probabilistic normed spaces. As applications, we utilize these results to study the Cauchy problem for a kind of differential inclusions with accertive mappings in probabilistic normed spaces.

This paper is based on the 36 wild Japanese quails which migrated to and settled in the Weishan Lake area. The gene frequency of 10 loci encoding the enzymes in viscera and muscle was detected. After collecting the same data about 20 quail colonies in China and other countries, it clusters the 21 quail populations by fuzzy cluster analysis. The study indicates that the wild Japanese quail in the Weishan Lake area is closer to domestic quail for phylogenetic system than wild Japanese quails in Japanese Islands. The paper supports the thesis that the quail domestication area should be further studied.
Changes in quality of Phellinus gilvus mushroom by different drying methods

This study was conducted to investigate the changes in characteristics of the Phellinus gilvus mushroom as influenced by drying methods after harvest. The lowest weight loss rate of P. gilvus mushroom was 75.8% with drying in the shade and 80% by dryer (60<SUP>o</SUP>C). The size loss rate of pileus was 19.3% of that in a hot air dryer (60<SUP>o</SUP>C). The hardness of dried material context using a hot air dryer (60<SUP>o</SUP>C) was the lowest (20kg/cm<SUP>2</SUP>), and that by a dry oven (60<SUP>o</SUP>C) was the highest (457kg/m<SUP>2</SUP>). For ΔE value, 4.9 of context and 2.6 of tubes using drying in the shade (20<SUP>o</SUP>C) were found to be the lowest. The survival rate of sarcoma 180 treated with P. gilvus dried in the sun was the lowest (51.8%), and this was considered the most effective method for antitumor activity against sarcoma 180.

Background: Hepatitis B virus (HBV) reactivation is increasing, as rituximab has become widely used for B-cell lymphoma. Thus, prevention and management of HBV reactivation are important in HBV-endemic areas. Methods: Hepatitis B virus (HBV) reactivation in HBV surface antigen (HBsAg)-positive patients and HBsAg-negative/HBV core antibody (HBcAb)-positive patients who received rituximab-containing chemotherapy was investigated by the Asia Lymphoma Study Group via retrospective (n=340), and the results were compared to cross-sectional analysis with patients who were prospectively monitored in a single institute (n=127). The goal of the study was to define the frequency of HBV reactivation and the efficacy of antiviral prophylaxis. Results: HBV reactivation was found in 27.8% of HBsAg-positive patients (45/162) in the retrospective analysis, being significantly less frequent in patients receiving antiviral prophylaxis than those not (22.9%, 32/140 versus 59.1%, 13/22; p<0.001). Lamivudine was most commonly used (96/162, 59.3%), but more than 20% of HBsAg-positive patients showed breakthrough HBV reactivation. In the cross-sectional analysis, a reduced rate of HBV reactivation occurred for entecavir as compared with lamivudine prophylaxis (6.3% versus 39.3%; p<0.05). HBV DNA monitoring of HBsAg-negative/HBcAb-positive patients in the cross-sectional analysis showed HBV reactivation in only 2.4% of cases. Conclusions: This is the largest study of HBV reactivation in patients receiving rituximab-containing chemotherapy to date, and we defined the probability of HBV reactivation in an HBV-endemic region.

<P>We present edge gyrokinetic simulations of tokamak plasmas using the fully non-linear (full-<I>f</I>) continuum code TEMPEST. A non-linear Boltzmann model is used for the electrons. The electric field is obtained by solving the 2D gyrokinetic Poisson equation. We demonstrate the following. (1) High harmonic resonances (<I>n</I> > 2) significantly enhance geodesic-acoustic mode (GAM) damping at high <I>q</I> (tokamak safety factor), and are necessary to explain the damping observed in our TEMPEST <I>q</I>-scans and consistent with the experimental measurements of the scaling of the GAM amplitude with edge <I>q</I><SUB>95</SUB> in the absence of obvious evidence that there is a strong <I>q</I>-dependence of the turbulent drive and damping of the GAM. (2) The kinetic GAM exists in the edge for steep density and temperature gradients in the form of outgoing waves, its radial scale is set by the ion temperature profile, and ion temperature inhomogeneity is necessary for GAM radial propagation. (3) The development of the neoclassical electric field evolves through different phases of relaxation, including GAMs, their radial propagation and their long-time collisional decay. (4) Natural consequences of orbits in the pedestal and scrape-off layer region in divertor geometry are substantial non-Maxwellian ion distributions and parallel flow characteristics qualitatively like those observed in experiments.</P>

A new generalized contraction mapping principle in probabilistic metric spaces is obtained. As an application, we utilize this principle to prove the existence theorems of solutions to differential equations in probabilistic metric spaces. All the results presented in this paper are new.
MiR-145 functions as a tumor suppressor by directly targeting histone deacetylase 2 in liver cancer

Aberrant regulation of histone deacetylase 2 (HDAC2) plays a pivotal role in the development of hepatocellular carcinoma (HCC), but, the underlying mechanism leading to HDAC2 overexpression is not well understood. We performed microRNA (miRNA) profiling analysis in a subset of HCCs, and identified four down-regulated miRNAs that may target HDAC2 in HCC. Ectopic expression of miRNA mimics evidenced that miR-145 suppresses HDAC2 expression in HCC cells. This treatment repressed cancer cell growth and recapitulated HDAC2 knockdown effects on HCC cells. In conclusion, we suggest that loss or suppression of miR-145 may cause aberrant overexpression of HDAC2 and promote HCC tumorigenesis.

This paper is based on the 9 goat colonies along the middle and lower Yellow River valley and 7 local goat colonies in the Northeast, Tibet and the Yangtze valley. After collecting the same data about the 22 goat colonies in China and other countries, it establishes and composes the matrix of fuzzy similarity relation describing the genetic similarities of different colonies. It also clusters 38 colonies according to their phylogenetic relationship. The establishment of the matrix and the cluster are effected in terms of the frequency of 18 loci and 43 allelomorphs in blood enzyme and other protein variations. The study proves that the middle Yellow River valley is one of the taming and disseminating centers of domestic goats in the South and East of Central Asia. Compared with other goat populations in this vast area, the native goat populations in the west of Mongolian Plateau, the Qinghai-Tibet Plateau and the middle Yellow River valley share the same origin. The colonies in the lower Yellow River valley and those in the middle valley, however, are relatively remote in their phylogenetic relationship. The native goat colonies in the southeast of Central Asia can be classified into two genetic groups: "East Asia" and "South Asia" and the colonies in Southeast Asia belong to either group.
