Prion Protein Genotypes in Pakistani Goats

The PCR-amplified prion protein (PrP) gene was sequenced to determine the frequency of scrapie-associated as well as novel PrP genotypes in 72 healthy goats representing five breeds. A total of six genotypes were detected, resulting from the three reported 143 (H/R), 154 (R/H) and 240 (S/P) and the two novel 39 (S/R) and 185 (I/F) amino acid polymorphisms. Of the four silent mutations 42 (a$\rightarrow$g), 138 (c$\rightarrow$t), 231 (c$\rightarrow$a) and 237 (g$\rightarrow$c) detected in this study, 237 (g$\rightarrow$c) is novel. A genotype (SIP/RFP) harboring three amino acid polymorphisms 39 (S/R), 185 (I/F) and 240 (S/P) was found in few goats. Although both scrapie-associated genotypes with 143 (H/R) and 154 (R/H) polymorphisms and others with 39 (S/R), 185 (I/F) and 240 (S/P) polymorphisms were present in the studied Pakistani goats, their frequency was lower than that of the wild-type genotype SHRIS/SHRIS (34.7%). These results emphasize the need for further sequencing of the PrP gene in a large number of goats representing the five studied breeds, so that overall PrP variability can be assessed in these breeds in research addressing future concerns about scrapie.

<P>Substoichiometric tungsten oxide films of approximately 10 nm thickness deposited with pulsed laser ablation on single-crystal TiO<SUB>2</SUB> substrates with (001) and (110) orientation show defect states near the Fermi energy in the valence-band X-ray photoelectron spectroscopy (XPS) spectra. The spectral weight of the defect states is particularly strong for the film grown on the (001) surface. In situ XPS under an oxygen pressure of 100 mTorr shows that the spectral weight of the defect states decreases significantly at 500 K for the film on the (110) substrate, whereas that of the film grown on the (001) substrate remains the same at a temperature up to 673 K. Furthermore, diffusion of titanium from the substrate to the film surface is observed on the (110) substrate, as is evidenced by the sudden appearance of the Ti 2p core level signature above 623 K and below 673 K. The film grown on the (001) surface does not show such an interdiffusion effect, which suggests that the orientation of the substrate can have a significant influence on the high-temperature integrity of the tungsten oxide films. Background Subacute sclerosing panencephalitis (SSPE) is a delayed and fatal manifestationof measles infection. Fulminant SSPE is a rare presentation in which the disease progresses todeath over a period of 6 months. The clinical features are atypical and can be misleading. Case Report We report herein a teenage boy who presented with acute-onset gait ataxia followed by right hemiparesis that evolved over 1 month, with left-hemispheric, delta-range slowing on the electroencephalogram (EEG). Magnetic resonance imaging disclosed multiple whitematter hyperintensities, suggesting a diagnosis of acute disseminated encephalomyelitis. Hereceived intravenous steroids, and within 4 days of hospital admission he developed unilateralslow myoclonic jerks. Repeat EEG revealed Rademecker complexes, pathognomonic of SSPE,and an elevated titer of IgG antimeasles antibodies was detected in his cerebrospinal fluid. Thedisease progressed rapidly and the patient succumbed within 15 days of hospitalization. The diagnosis of SSPE was confirmed by autopsy. Conclusions This case illustrates the difficulty of recognizing fulminant SSPE when it manifests with asymmetric clinical and EEG abnormalities.

<P>Rh0.5Pd0.5/CeO2 bimetallic nanoparticles were subjected to reducing and oxidizing atmospheres at different temperatures and characterized by means of the Ambient Pressure X-Ray Photoelectron Spectroscopy (AP-XPS). The surface atomic population is calculated for all cases. By using a high temperature pretreatment at 480 degrees C in H-2 atmosphere, it was possible to control the surface atomic population with Pd atoms frozen at the skin layer even in an oxidizing atmosphere, which is not observed without high temperature pre-treatment (Rh atoms at the skin layer). The change of the Rh/Pd surface segregation behavior after the high temperature pre-treatment is attributed to the geometrical factor of Strong Metal Support Interaction (SMSI) effect, i.e., the occurrence of the ceria capping layer around nanoparticles.
