      • The Traditional Culture of Northeast Heated Kang and Development of New Type of Heated Kang

        Heated Kang as one of the most important elements of the northeast rural residence, formed a unique mode of heated brick bed and culture of heated brick bed in the long history of more than two thousand years. Kang bed-stove has many functions, for example, heating residential, communication, emotion and so on. Not only it is the carrier of the northeast family’s material culture, also it plays a major role to spatial organization and building of the rural housing. Kang bed-stove is a way of the repeated skillful use of energy. The traditional heated brick bed built with mud and brick. It connects heat able adobe sleeping platform in its outside and links chimney in its inside. The quantity of heat of Kang bed-stove comes from the waste heat of cooking .In recent years, the new rural construction of northeast is in the stage of development of high-speed. With the progress of technology and the renewal of the concept, based on the traditional heated brick bed, people have made some beneficial attempts to change and have introduced some new heated brick bed: Suspended Kang, energy saving kang and so on. All new heated brick beds both retain the traditional heated brick bed’s functions, and conform to the requirements of sustainable development planning. new heated brick beds are the combination of traditional culture and the new technology. This paper bases on rural housing of northeast area as the research object, discusses the connotation、forms and types、 heating means of traditional heated brick bed and the evolution of new type of Heated Kang.

      • A study on sustainable development of Beijing Siheyuan in the state of modern life

        Siheyuan, is the traditional Chinese courtyard style building, has a long history, and is an extremely important historical and cultural heritage. It carries the folk culture of old Beijing and the history of the old Beijing. Therefore, its cultural connotation should be maintained. With the growing number of skyscrapers, construction industrialization replacing traditional construction technology today, hutongs and Siheyuan have lost their luster, most of the traditional Siheyuan have become shabby, inner vitality and old form that were the social mechanism are rotting away. Just relying on protection and repair Siheyun is far from a healthy survival. Instead development and innovation is needed for a vibrant sustainable preservation of Siheyuan. In order to make Siheyuan more comfortable, more vibrant and to achieve its sustainable development, it should be combined with modern materials and construction technology.

      • Immune and Anti-oxidant Functions of Ethanol Extracts of Scutellaria baicalensis Georgi in Mice Bearing U14 Cervical Cancers

        Background: The objective was to study the effect of Scutellaria baicalensis Georgi ethanol extracts (SBGE) on immune and anti-oxidant function in U14 tumor-bearing mice. Materials and Methods: U14 tumor-bearing mice were randomly divided into eight groups: a control group, a cyclophosphamide (CTX) group, three dose groups of SBGEI (high, medium, low), and three dose groups of SBGEII (high, medium, low). After two weeks, the thymus and spleen weight indices of mice bearing U14 cervical cancer were calculated. Enzyme linked immunosorbent assays (ELISA) was used to determine the levels of serum IL-2, TNF-${\alpha}$, IL-8, and PCNA. MDA activity and SOD activity in plasma were measured with detection kits. Results: In the SBGE groups, thymus weight and spleen weight indices of U14 tumor-bearing mice were significantly higher than in the control group or CTX group (p<0.05). Compared to control group, the levels of serum IL-2 and TNF-${\alpha}$ in U14 tumor-bearing mice increased significantly, whereas the contents of serum IL-8 and PCNA decreased (p<0.05). The activity of SOD increased with the growing dose of SBGE, while the activity of MDA decreased significantly in the highe-rdose groups of SBGE. Conclusions: These findings suggested that SBGE, especially at high dose, 1000 mg/kg, showed significant immune and anti-oxidant effects infU14 tumor-bearing mice, which might be the mechanisms of SBGE inhibition of tumor growth.

      • Comparison of Ligasure Versus Conventional Surgery for Curative Gastric Cancer Resection: a Meta-Analysis

        Background: The LigaSure vessel sealing system has been proposed to save operation time and reduce intraoperative blood loss for various surgeries. However, its usage for gastric cancer is still controversial. The aim of the present meta-analysis was to compare the effectiveness of LigaSure with conventional surgery in gastrectomy. Materials and Methods: Sources were retrieved from the Cochrane Library, MEDLINE, EMBASE, SCOPUS and Google Scholar until February, 2015. All randomized controlled trials comparing LigaSure with conventional surgery in curative gastric cancer resection were selected. After data extraction, statistics were performed by Review Manager 5.1 software. Results: Three eligible randomized controlled trials were evaluated, with a total of 335 patients. The quality of the included trials was good, yet some methodological and clinical heterogeneity existed. There were no significant differences between the LigaSure and conventional groups in operative time (weighted mean difference [WMD], -22.95 minutes; 95% confidence interval [CI], [-59.75, 13.85]; P = 0.22), blood loss (WMD, -45.8 ml; 95% CI, [-134.5, 42.90]; P = 0.31), nor the incidence of surgical complications (odds ratio, 1.18; 95% CI, [0.68, 2.05]; P = 0.54). But there was a longer duration of hospital stay in LigaSure group (WMD, 1.41 days; 95% CI, [0.14, 2.68]; P = 0.03). Conclusions: All available randomized evidence has been summarized. LigaSure does not confer significant advantage over conventional surgery for curative gastric cancer resection. The usefulness of the device may be limited in gastrectomy. But, more trials are needed for further assessment of the LigaSure system for gastric cancer.

        An Improved Integrative Transformation System for Pichia pastoris with DNA-polyethylenimine-dextran Sulfate Nanoparticles

        Pichia pastoris is a suitable host for selecting mutant enzymes, since it can secret many gene products to the medium. However, poor transformation efficiency is often encountered. This makes it difficult to obtain a large number of transformants necessary to thoroughly cover a large library. We report here that pre-coating DNA with polyethyleneimine and dextran sulfate increased gene integration significantly in Pichia electroporation. This improvement on integrative efficiency has been proved in different voltage, resistance, cell phase, and DNA concentrations. The condensed DNA nanoparticles make Pichia available as a host for screening large libraries of random mutants.

        Genomic diversity of the Avian leukosis virus subgroup J gp85 gene in different organs of an infected chicken

        The genomic diversity of Avian leukosis virus subgroup J (ALV-J) was investigated in an experimentally infected chicken. ALV-J variantsin tissues from four different organs of the same bird were re-isolated in DF-1 cells, and their gp85 gene was amplified and cloned. Ten clonesfrom each organ were sequenced and compared with the original inoculum strain, NX0101. The minimum homology of each organ rangedfrom 96.7 to 97.6%, and the lowest homology between organs was only 94.9%, which was much lower than the 99.1% homology of inoculumNX0101, indicating high diversity of ALV-J, even within the same bird. The gp85 mutations from the left kidney, which contained tumors,and the right kidney, which was tumor-free, had higher non-synonymous to synonymous mutation ratios than those in the tumor-bearing liverand lungs. Additionally, the mutational sites of gp85 gene in the kidney were similar, and they differed from those in the liver and lung, implyingthat organ- or tissue-specific selective pressure had a greater influence on the evolution of ALV-J diversity. These results suggest that moreALV-J clones from different organs and tissues should be sequenced and compared to better understand viral evolution and molecularepidemiology in the field.

      • 4-Hydroxynonenal Promotes Growth and Angiogenesis of Breast Cancer Cells through HIF-1α Stabilization

        4-Hydroxynonenal (4-HNE) is a stable end product of lipid peroxidation, which has been shown to play an important role in cell signal transduction, while increasing cell growth and differentiation. 4-HNE could inhibit phosphatase and tensin homolog (PTEN) activity in hepatocytes and increased levels have been found in human invasive breast cancer. Here we report that 4-HNE increased the cell growth of breast cancer cells as revealed by colony formation assay. Moreover, vascular endothelial growth factor (VEGF) expression was elevated, while protein levels of hypoxia inducible factor 1 alpha (HIF-$1{\alpha}$) were up-regulated. Sirtuin-3 (SIRT3), a major mitochondria NAD+-dependent deacetylase, is reported to destabilize HIF-$1{\alpha}$. Here, 4-HNE could inhibit the deacetylase activity of SIRT3 by thiol-specific modification. We further demonstrated that the regulation by 4-HNE of levels of HIF-$1{\alpha}$ and VEGF depends on SIRT3. Consistent with this, 4-HNE could not increase the cell growth in SIRT3 knockdown breast cancer cells. Additionally, 4-HNE promoted angiogenesis and invasion of breast cancer cells in a SIRT3-dependent manner. In conclusion, we propose that 4-HNE promotes growth, invasion and angiogenesis of breast cancer cells through the SIRT3-HIF-$1{\alpha}$-VEGF axis.

        Protective effect of procyanidin B2 on hydrogen peroxide (H2O2)-induced oxidative damage in MCF-7 cells

        The aim of this study is to assess the cytoprotection and potential molecular mechanisms of procyanidin B2 ( PCB2) on hydrogen peroxide ( H2O2)-induced oxidative damage in MCF-7 cells. The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was performed to determine the viability of MCF-7 cell exposure to H2O2 or PCB2. We measured the antioxidant properties of PCB2 by determining the activities of SOD, GSH-Px, LDH and MDA levels, and evaluated apoptosis and intracellular reactive oxygen species (ROS) levels. The related proteins expression levels were monitored by Western blot. MCF-7 cells induced with H2O2 had a remarkable decrease in cell viability that was suppressed when it was interfered with PCB2 (0.1–10.0 μM). PCB2 interference memorably and dose-dependently inhibited H2O2- induced LDH leakage, ROS and MDA overproduction, while PCB2 markedly increased H2O2- induced the activities of SOD and GSH-Px. Eventually, H2O2 prominently down-regulated the ratio of Bcl-2/Bax and the relative proteins expression levels of Nrf2, GCLC, NQO1 and HO-1, and up-regulated the relative proteins expression levels of cytochrome c, caspase-3 and Keap1. However, the relative expression levels of these proteins were reversed in PCB2- interfered MCF-7 cells. This study implied that protective effect of PCB2 on H2O2- induced oxidative damage in MCF-7 cells might be related to inhibition of mitochondria-dependent apoptosis, activation of Keap1/Nrf2/HO-1 signaling pathway and improvement of the antioxidant enzymes activities.

        A novel intelligent fault diagnosis method based on dual convolutional neural network with multi-level information fusion

        Due to the complicacy of mechanical instruments and the noise interference in the working environment, the equipment status information contained in a single sensor is insufficient, and multi-source information contains more complete status information. In order to effectively fuse multi-sensor information and improve the reliability of diagnosis, a multi-level fusion dual convolution neural network (MFDCNN) for fault diagnosis of rotating machinery is proposed in this paper. This approach realizes multi-level fusion of fault information by utilizing the flexibility of the structure of the convolutional neural network. During the training process, the two subnets automatically extract representative features from the multi-sensor timedomain signal and its frequency spectrum in parallel, and then fuse the extracted features for pattern recognition to achieve end-to-end fault diagnosis. Compared with the single sensor diagnosis method and single level information fusion method, this approach has better diagnosis performance.
