Interaction of glycopolymers with human hematopoietic cells from cord blood and peripheral blood

<P>Polystyrene derivatives, poly[N-pvinylbenzyl-O-D-glucopyranosyl-(1-4)-D-glucoamide] (PV Maltose) and poly[N-p-vinylbenzyl-O-mannopyranosyl-(1-4)-D-glucoamide] (PV Mannose), which contain glucose and mannose moieties, respectively, have the specific binding ability with murine hematopoietic cells. In this study, we confirm the ability of these glycopolymers to interact specifically with human hematopoietic stem cells (HSCs) and mature cells derived from human cord blood (CB) and peripheral blood (PB). Using fluorescence isothiocyanate (FITC)-labeled glycopolymers, we observed that 98% to 93% of hematopoietic cells interacted very strongly with PV Mannose, and 63% of CB and 29% PB interacted with PV Maltose. Both glycopolymers bound better to cells from CB than from PB. Cytotoxic studies revealed that a 0.1 mM dose of PV Mannose induced apoptosis in 20% CB cells, in contrast to 3–5% PB cells. Furthermore, we demonstrated that all of CD34<SUP>+</SUP> HSCs of both origins bound specifically to PV Mannose, whereas 33–47% bound to PV Maltose. In addition, the majority of B cells (CD19<SUP>+</SUP>), T cells (CD3<SUP>+</SUP>), monocytes (CD14<SUP>+</SUP>), and erythrocytes (CD235a<SUP>+</SUP>) bound to PV Mannose, but a lower percentage interacted with PV Maltose. In vivo study, bone marrow, spleen, and liver tissues in NOD-SCID mice injected with PV Mannose conjugated CB, were detected PV Mannose positive hematopoietic cells. These data suggest that the use of PV Mannose and PV Maltose might be used for gene and drug delivery for hematopoietic cells and thus, may be useful in therapeutic settings. © 2007 Wiley Periodicals, Inc. 대형 산불이 발생했을 경우 인접 마을 등에 대한 피난 연구는 국내에서 아직까지 이루어지지 않고 있으며 대부분의 대피 대상자들이 재해 약자들로 구성되어있어 산불 대피에 관한 연구가 필요한 실정이다. 이에 본 연구에서는 산불 대피지도 알고리즘을 개발하여 산불의 진행경로 및 확산에 따른 적정한 대피경로를 제시함으로서 인적 피해를 저감하는데 그 목적이 있다.

Welfare centers for disabled person which are typical facilities in Korea are classified into two catagories One is general welfare center for all type of disabled persons and the other is special welfare centers for special disabilities such as visually handicapped persons, hearing-impaired persons and the physically disabled. In this study the movements of disabled persons in daily lifes were measured and many evacuation drills in welfare centers were conducted for the achievement of basic data such as velocities and evacuation patterns of disabled persons The evacuation patterns are characterized by different kinds of disabilities.

Although the scale and number of university facilities have increased, the maintenance of these facilities has not been efficient as maintenance costs incurred during facility operation periods, as well as management time, have not been sufficiently considered. In this study, the maintenance record data of national universities was compared and analyzed to analyze factors that incur maintenance costs according to the uses and costs of the facilities. According to the results, the number of cases of maintenance and repair work for basic educational facilities was shown to be the largest, accounting for 79%. The number of maintenance and repair work occurred according to work type was examined. According to the results, the highest ratio of maintenance and repair work occurred in the field of construction, specifically, the maintenance costs required for facility supplementation accounted for the highest ratio. Maintenance costs per unit area decreased over time in the newer university facilities, while showing a tendency to increase in older university facilities. However, there were cases where maintenance costs did not show any trends due to factors such as the limited maintenance budgets owing to the characteristics of public service facilities and facilities in the stage of deterioration or destruction after the stage of stabilization. This study is considered to be helpful for efficient university facility maintenance plans and budget calculations.

We separately cultured follicular oocytes collected from individual ovaries of slaughtered Korean native cows and examined both the embryonic development rate and pregnancy rate after embryo transplantation according to the meat yield and quality grades of the source beef carcass. Oocytes from meat yield grade B cows exhibited a higher fertilization rate and embryonic developmental rate to the eight-cell stage than oocytes from grade A or C animals (p<0.05), but there was no significant difference in rate of development to the blastocyst stage among meat yield grades A, Band C. The oocyte cleavage rate and development rate to the eight-cell stage from meat quality grade 3 cattle was higher than grades 1++, 1+, 1 and 2 (p<0.05). Embryos derived from grade animals displayed a development rate to the blastocyst stage of 19.4%, which was also higher than all other meat quality grades (p<0.05). Transplantation of in vitro-cultured oocytes from meat yield grade A ovaries led to a higher pregnancy rate (64.2%) than in vitro-cultured oocytes from meat yield grade B ovaries (56.5%), but there was no significant difference between the two groups in pregnancy or abortion rates. In conclusion, embryonic development rate and pregnancy rate has a close relation to meat quality grades of the source beef carcass, this results is to give information for the Korean native cows improvement of breed.
