Molecular mechanism of cofilin dephosphorylation by ouabain

We previously reported that phosphorylated cofilin-triosephosphate isomerase(TPI) complex interacts with Na,K-ATPase and enhances the pump activity through the phosphorylation of cofilin via Rho-mediated signaling pathway, In this study, we tested the hypothesis that the dephosphorylation of cofilin may be induced through Na,K-ATPase inhibition by ouabain. The phosphorylation level of cofilin by ouabain which decreases in a time- and dose-dependent manner in various human cell lines, remains unchanged by pretreatment with Src inhibitor, PP2; epidermal growth factor receptor (EGFR) inhibitor, AG1478; Raf-1 kinase (Raf) inhibitor, GW5074; and ERK. kinase (MEK) inhibitor, PD98059, and by transfection of Ras dominant negative mutant (RasN17). This suggests that ouabain dephosphorylates cofilin through the Src/EGFR/Ras/Raf/MEK pathway. Ouabain activates Ras/Raf/MEK pathway, but down-regulates Rho kinase (ROCK)/LIM kinase (LIMK)/cofilin pathway, implying that there may be a cross-talk by ouabain between the Ras/Raf/MEK. and the ROCK/LIMK/cofilin pathways. Immunofluorescence and flow cytometry suggest that ouabain-induced active form of cofilin may be involved in cytoskeletal reorganization and cell volume regulation. Thus, these findings demonstrate a new molecular mechanism for the dephosphorylation of cofilin through the inhibition of Na,K-ATPase by ouabain. ⓒ 2006 Elsevier Inc. All rights reserved.

Generally, concrete temperature is risen by hydration at the time of mixing right after, then that falling down gradually until the atmospheric temperature. By this temperature falling concrete is contracted. If contraction is restricted externally or internally, tensile stress is occurred at concrete. If this tensile stress exceeds tensile strength of concrete, crack is occurred at concrete. The rahmen bridge among many bridges's is typical type that is occurred tensile crack at slab. In this paper, the authors investigate how temperature of a rahmen concrete bridge is changed by slab position and the number of days. The results of field tests will be applied important crack control data by hydration of concrete. Considering results of field test, the authors analyze thermal stress to the number of days in slab concrete.

<P>Background: Attitudes toward menopause vary across cultures and influence women's experiences of menopausal symptoms, possibly leading to reduced posttreatment quality of life in breast cancer survivors. Objective: The aim of this study is to examine the effects of menopausal symptoms and attitudes on health-related quality of life in breast cancer survivors who were premenopausal at the time of diagnosis. Methods: A total of 139 women receiving chemotherapy with/without endocrine therapy were assessed with self-report questionnaires of established reliability and validity. Hierarchical regression was conducted to assess the impact of menopausal symptoms and attitudes on quality of life, while controlling for demographic characteristics. Results: Overall, participants endorsed more than half of 46 symptoms, most at the level of mild symptoms, and most reported a less positive attitude toward menopause. Lower quality of life was significantly predicted by more menopausal symptoms endorsed and more negative attitudes when controlling for demographic factors associated with quality of life (R-2= 26.1%). Most participants experienced change from premenopause to postmenopause after the completion of adjuvant chemotherapy with or without tamoxifen. Conclusions: The results suggest that more menopausal symptoms and negative attitudes toward menopause may affect health-related quality of life considerably in chemotherapy-treated Asian breast cancer survivors. Implications for Practice: Healthcare professionals should develop a better understanding of the effects of menopausal symptoms and attitudes on quality of life by using a culturally relevant perspective based on patients' sociocultural backgrounds. Furthermore, these findings help healthcare professionals communicate with their Asian clients in a more informed way and provide culturally appropriate and individualized care.</P>
Identification of the Calcium Binding Sites in Translationally Controlled Tumor Protein

Translationally controlled tumor protein (TCTP), also known as IgE-dependent histamine-releasing factor, is a growth-related tumor protein. Although the primary sequence of rat TCTP does not reveal any recognizable Ca^2+-binding motif, previous studies have demonstrated that rat TCTP consisting of 172 amino acids is a Ca^2+-binding protein. However, the region of TCTP required for Ca^2+ interaction has not been mapped to the molecule. Here, we reported that the Ca^2+ binding region of TCTP, which was mapped by using a combination of deletion constructs of rat TCTP and ^45Ca^2+-overlay assay, was confined to amino acid residues 81-112. This binding domain did not show any peculiar loop of calcium-binding motif such as CaLB domain and EF hand motif and it seems to be constituted of random coil regions neighboring the a helix. Thus, our data confirm that TCTP is a novel family of Ca^2+-binding protein.

Purpose: This study aimed to investigate the effect of the professional quality of life on depression in clinical nurses from a tertiary hospital. Methods: A descriptive study design was used. The participants were 139 nurses who worked at a tertiary hospital in C city. The collected data were analyzed using t-test, ANOVA, Pearson correlation coefficient, and multiple regressions analysis with SPSS 26.0 program. Results: Depression was positively correlated with compassion fa-tigue (r=.28, p<.001) and burnout (r=.49, p<.001), whereas compassion satisfaction was negatively associated with de-pression (r=-.29, p<.001). The factor influencing depression was burnout (β=.36, p=.002), which explained 24.2% of the variance in depression. Conclusion: It is necessary to develop a program that can effectively manage burnout to reduce depression in clinical nurses.

<P>Autologous adipose stromal vascular fractions (SVFs) containing adipose tissue-derived stem cells (ASCs) are currently being used in clinical settings for various orthopedic applications for human patients. Due to its potential capability of regenerating cartilage, bone, and tendons, autologous adipose SVFs are being tried in treating patients with osteoarthritis (OA), chondromalacia, meniscus tear, osteonecrosis of the femoral head, and tendon injuries. Here, we have reviewed available human clinical studies with regard to patient applications of autologous adipose SVF containing ASCs, specifically assessing effectiveness and safety in the field of orthopedic disorders. All studies reviewed in this article presents potential benefits of autologous adipose SVF in various orthopedic applications without any serious side effects.</P>
Why cannot a β-lactamase gene be detected using an efficient molecular diagnostic method?

<P><B>Objective:</B></P><P>Fast detection of β-lactamase (<I>bla</I>) genes can minimize the spread of antibiotic resistance. Although several molecular diagnostic methods have been developed to detect limited <I>bla</I> gene types, these methods have significant limitations, such as their failure to detect almost all clinically available <I>bla</I> genes. We have evaluated a further refinement of our fast and accurate molecular method, developed to overcome these limitations, using clinical isolates.</P><P><B>Methods:</B></P><P>We have recently developed the efficient large-scale <I>bla</I> detection method (<SUB>large-scale</SUB><I>bla</I>Finder) that can detect <I>bla</I> gene types including almost all clinically available 1,352 <I>bla</I> genes with perfect specificity and sensitivity. Using this method, we have evaluated a further refinement of this method using clinical isolates provided by International Health Management Associates, Inc. (Schaumburg, Illinois, USA). Results were interpreted in a blinded manner by researchers who did not know any information on <I>bla</I> genes harbored by these isolates.</P><P><B>Results:</B></P><P>With only one exception, the <SUB>large-scale</SUB><I>bla</I>Finder detected all <I>bla</I> genes identified by the provider using microarray and multiplex PCR. In one of the <I>Escherichia coli</I> test isolates, a <I>bla</I><SUB>DHA-1</SUB> gene was detected using the multiplex PCR assay but it was not detected using the <SUB>large-scale</SUB><I>bla</I>Finder.</P><P><B>Conclusion:</B></P><P>The truncation of a <I>bla</I><SUB>DHA-1</SUB> gene is an important reason for an efficient molecular diagnostic method (<SUB>large-scale</SUB><I>bla</I>Finder) not to detect the <I>bla</I> gene.</P>

<B>ABSTRACT</B><P>The coexistence of<I>qnrB62</I>and<I>bla</I>VIM-2was detected in aCitrobacterclinical isolate. The reduced fluoroquinolone susceptibility is attributable to<I>qnrB62</I>, mutations of quinolone-resistance-determining regions, and an efflux pump or pumps. The genetic context surrounding chromosomal<I>qnrB62</I>was a novel complex class 1 integron (In1184::IS<I>CR1</I>::<I>qnrB62</I>) containing a unique gene array (<I>bla</I>VIM-2-<I>aacA4′-8</I>-<I>gucD</I>). An 18-nucleotide deletion at the 3′ end of the<I>pspA</I>gene [<I>pspA</I>(Δ<I>18</I>)], upstream of<I>qnrB62</I>, and an inverted repeat region (IRR2) were detected in In1184::IS<I>CR1</I>::<I>qnrB62</I>, indicating past transposition events.</P>
