Using site-directed mutagenesis technique, I havereplaced serine 285 and serine 292 with the alanine,and assesed the binding of agonist and signalingsuch as the inhibition of adenylyl cyclase activity.I have found that serine 292 has an important rolein the signal transduction of canabinoid agonists,HU-210 and CP55940, but not in that of amino-alkylindoles derivatives WIN55,212-2. Al mutants expresusing monoclonal antibody HA 1 as compared witht h e w i l d t y p e r e c e p t o r .Interestingly, binding afinity of S285A and S292Amutants with clasical canabinoid agonist HU-243was omewhat decreased. In signaling asay, theinhibition of adenylyl cyclase by HU-210, CP55940and WIN55,212-2 is the same order in both wild typereceptor and S285A mutant receptor. However, S292Ahave ben shown that the inhibition curves of adenylylcyclase activity moved to the right by HU-210 andCP55940, but hose of adenylyl cyclase activity didthat this residue is closely related to the binding sitewith HU-210 and CP55940. In adition, serine 292m i g h t a k e m o r e i m p o r t a n t r o l e i n C B 2 r e c e p t o r a n dG-protein signaling than serine 285.

The distribution of the nerve growth factor (NGF),the glial fibrillary acidic protein (GFAP) and theciliary neurotrohic factor (CNTF) was performed incoronal sections of the mesencephalon, rhombencephalonand spinal cord in the developing Mongolian gerbils.Generaly, NGF specificaly recognizes neurons withthe NGF receptor, whereas GFAP does the glia, andwas examined separately in gerbils between embryonicdays 15 (E15) and postnatal weks 3 (PNW 3). TheNGF-IR was first observed in the spinal cord at E21,which might be related to the maturation. The GFAPreactivity was peaked at he postnatal days 2 (PND2),while the highest CNTF-reaction was expresed atPNW 2. The GFAP stains were observed in theaqueduct and the spinal cord, which appeared toproject lateraly at E19. The CNTF was observed onlyafter the birth and found in both the neurons andcerebelum and the spinal cord from PND1 to PNW3.These results uggest hat NGF, GFAP and CNTF areimportant for the development of the neurons and theneuroglia in the central nervous system at he lateprenatal and postnatal stages.

Secretion of neurotransmitters is initiated byvoltage-gated calcium influx through presynaptic, voltage-gated N-type calcium channels. However, litle isknown about their celular distribution in the mousecerebelum. In the cerebelum, α1B imunoreactivityis found mainly on the cel bodies of al Purkinjecells. In adition, the imunoreactivity was detectedon a subset of Purkinje cell dendrites, clustered tolobe vermis, imunoreactive Purkinje cel dendritesform narow stripes eparated by broad bands ofunstained endrites. Moving caudally through thevermis, these stripes become thicker as a larger fractionof the Purkinje cell dendrites become imunoreactive.This localization study of the α1B pore-formingsubunits in mouse cerebelum ay guide futureinvestigations of the role of calcium channels inneurological pathways.
