      • Evaluation of Preparation Methods for Scanning Electron Microscopic Observation of Plant Protoplasts

        佐藤忠彦·權 五昌**·三宅 博·谷口 武·前田英三 (名古屋大學農學部·**韓國東亞大學校農科大學) 要 旨:ペチュニアとイネのプロトプラストを用いて, 走査電子顯微鏡の試料調製法の檢討を行つた. アルコ-ル脫水處理中に生じるプロトプラストの形狀變化を??減するための固定條件を調査した. その結果, 固定條件の違いにより, 脫水處理により生じゐプロトプラストの體積の減少にかなりの差が認められ, グルタルアルデヒド固定, グルタルアルデヒド·オスミウム固定, グルタルアルデヒド·タンニン酸·オスミウム固定の順に, 體積の減少が少なくなつた, グルタルアルデヒド單獨固定の場合に, ペチュニア葉由來プロトプラストの葉綠體の綠色は, アルコ-ル脫水により脫色した. グルタルアルデヒド·タンニン酸·オスミウムで固定すゐと, ペチュニアとイネのプロトプラストの良く保存された走査電子顯微鏡像が得られた. Preparation methods for scanning electron microscopy (SEM) were studied with petunia and rice protoplasts. Fixation schedules to sustain protoplast size during alcohol dehydration were examined. When the different fixations were compared, the decrease in protoplast volume was observed to a variable extent during the dehydration process. The extent of volume decrease was reduced in order of glutaraldehyde, glutaraldehyde-osmium tetroxide, and glutaraldehyde-tannic acid-osmium tetroxide schedules. When fixed only with glutaraldehyde, the green color of the chloroplast in petunia leaf protoplasts was lost during alcohol dehydration. Well-defined scanning electron micrographs of petunia and rice protoplasts were obtained using a glutaraldehyde-tannic acid-osmium tetroxide schedule.

      • The Development of the Totalized Hydrogen Energy Utilization System for Commercial Buildings

        We have proposed the Totalized Hydrogen Energy Utilization System(THEUS) for applying to commercial buildings. THEUS consists of fuel cells, water electrolyzers, metal hydride tanks and their auxiliaries. We evaluated the energy saving of the basic THEUS in view of an actual operation using thermal demand data of actual buildings. The THEUS have ability for achieving energy saving, in comparison with an ice storage system for cooling demand. We have developed the hydrogen storage system using metal hydride and unitized reversible cell (URC) combine the functionality of a fuel cell and an electrolyzer in a single device, these are important components of THEUS. The metal hydride tank designed and manufactured to evaluate that we assumed actual operation in the experiments. We developed the numerical simulation code, and simulation results are in good agreement with experimental data. The simulation about two tanks system was done by using this code. The heat utilization can be improved greatly. To achieve URC commercial use, improvement of durability was examined. It confirmed that our URC had durability for several thousand hours.

      • Improvement in the Super Low Core-loss Soft Magnetic Materials

        We reported a P/M soft magnetic material with core loss value of , which is lower than that of 0.35mm-thick laminated material, by using high purity gas-atomized iron powder. Lack of mechanical strength and high cost of powder production are significant issues for industrial use. In order to achieve both low core loss and high strength by using inexpencive powder, the improvement of powder shape and surface morphology and binder strength was conducted. As the result, the material based on water-atomized powder with 80 MPa of TRS and 108 W/kg of core loss (W10/1k) was achieved.

      • Large Porphyrin Squares from the Self-Assembly of meso-Triazole-Appended L-Shaped meso–meso-Linked Zn<sup>II</sup>–Triporphyrins: Synthesis and Efficient Energy Transfer

        <P>meso-Triazolyl-appended Zn<SUP>II</SUP>–porphyrins were readily prepared by Cu<SUP>I</SUP>-catalyzed 1,3-dipolar cycloaddition of benzyl azide to meso-ethynylated Zn<SUP>II</SUP>–porphyrin (click chemistry). In noncoordinating CHCl<SUB>3</SUB> solvent, spontaneous assembly occurred to form tetrameric array (3)<SUB>2</SUB> from meso–meso-linked diporphyrins 3, and dodecameric porphyrin squares (4)<SUB>4</SUB> and (5)<SUB>4</SUB> from the L-shaped meso–meso-linked triporphyrins 4 and 5. The structures of these assemblies were examined by <SUP>1</SUP>H NMR spectra, absorption spectra, and their gel permeation chromatography (GPC) retention time. Furthermore, the structures of the dodecameric porphyrin squares (4)<SUB>4</SUB> and (5)<SUB>4</SUB> were probed by small- and wide-angle X-ray scattering (SAXS/WAXS) measurements in solution using a synchrotron source. Excitation-energy migration processes in these assemblies were also investigated in detail by using both steady-state and time-resolved spectroscopic methods, which revealed efficient excited-energy transfer (EET) between the meso–meso-linked Zn<SUP>II</SUP>–porphyrin units that occurred with time constants of 1.5 ps<SUP>−1</SUP> for (3)<SUB>2</SUB> and 8.8 ps<SUP>−1</SUP> for (5)<SUB>4</SUB>.</P> <B>Graphic Abstract</B> <P>Getting it together: In a noncoordinating solvent, spontaneous assembly occurs to form a tetrameric array from meso–meso-linked diporphyrins, and a dodecameric porphyrin square from L-shaped meso–meso-linked triporphyrins (see graphic).         Study of Flow Field and Pressure Distribution on a Rotor Blade of HAWT in Yawed Flow Conditions

        This paper describes the flow field and the blade pressure distribution of a horizontal axis wind turbine in various yawed flow conditions. These measurements were carried out with 2.4m-diameter rotor with pressure sensors and a 2-dimensional laser Doppler velocimeter for each azimuth angle in a wind tunnel. The results show that aerodynamic forces of the blade based on the pressure measurements change according to the local angle of attack during rotation. Therefore the wake of the yawed rotor becomes asymmetric for the rotor axis. Furthermore, the relations between aerodynamic forces and azimuth angles change according to tip speed ratio. By the experimental analysis, the flow field and the aerodynamic forces for each azimuth angle in yawed flow condition were clarified.

      • A Single Nucleotide Polymorphism within the Acetyl-Coenzyme A Carboxylase Beta Gene Is Associated with Proteinuria in Patients with Type 2 Diabetes

        <▼1><P>It has been suggested that genetic susceptibility plays an important role in the pathogenesis of diabetic nephropathy. A large-scale genotyping analysis of gene-based single nucleotide polymorphisms (SNPs) in Japanese patients with type 2 diabetes identified the gene encoding acetyl-coenzyme A carboxylase beta (<I>ACACB</I>) as a candidate for a susceptibility to diabetic nephropathy; the landmark SNP was found in the intron 18 of <I>ACACB</I> (rs2268388: intron 18 +4139 C > T, p = 1.4×10<SUP>−6</SUP>, odds ratio = 1.61, 95% confidence interval [CI]: 1.33–1.96). The association of this SNP with diabetic nephropathy was examined in 9 independent studies (4 from Japan including the original study, one Singaporean, one Korean, and two European) with type 2 diabetes. One case-control study involving European patients with type 1 diabetes was included. The frequency of the T allele for SNP rs2268388 was consistently higher among patients with type 2 diabetes and proteinuria. A meta-analysis revealed that rs2268388 was significantly associated with proteinuria in Japanese patients with type 2 diabetes (p = 5.35×10<SUP>−8</SUP>, odds ratio = 1.61, 95% Cl: 1.35–1.91). Rs2268388 was also associated with type 2 diabetes–associated end-stage renal disease (ESRD) in European Americans (p = 6×10<SUP>−4</SUP>, odds ratio = 1.61, 95% Cl: 1.22–2.13). Significant association was not detected between this SNP and nephropathy in those with type 1 diabetes. A subsequent <I>in vitro</I> functional analysis revealed that a 29-bp DNA fragment, including rs2268388, had significant enhancer activity in cultured human renal proximal tubular epithelial cells. Fragments corresponding to the disease susceptibility allele (T) had higher enhancer activity than those of the major allele. These results suggest that <I>ACACB</I> is a strong candidate for conferring susceptibility for proteinuria in patients with type 2 diabetes.</P></▼1><▼2><P><B>Author Summary</B></P><P>Although cumulative epidemiological findings have suggested that genetic susceptibility plays an important role in the pathogenesis of diabetic nephropathy, no gene conferring susceptibility to diabetic nephropathy has been definitively identified. In a large-scale association study of 1,312 Japanese subjects with type 2 diabetes using SNPs from a Japanese SNP database, we show that the T-allele of <I>ACACB</I> rs2268388 is associated with diabetic nephropathy. We also show that the association is consistently observed in patients with type 2 diabetes and proteinuria across different ethnic groups, including populations of European descent. Because a DNA fragment corresponding to the disease susceptibility allele is shown to have higher enhancer activity, we hypothesize that the increase in the expression and/or activity of the encoded acetyl-coenzyme A carboxylase beta contributes to the development and progression of diabetic nephropathy. Our present analysis provides novel insight into the pathogenesis of diabetic nephropathy. This finding is important because diabetic nephropathy is a leading cause of end-stage renal disease and affects life expectancy in subjects with type 2 diabetes.</P></▼2>

      • Improvement of Numerical Simulation Model of the Metal Hydride Tank for the Totalized Hydrogen Energy Utilization System

        Authors have proposed the Totalized Hydrogen Energy Utilization System(THEUS) for load leveling and stabilizing the grid system. THEUS consists of fuel cells, water electrolyzers, hydrogen storage and their auxiliary machinery. The basic operation for load leveling is as follows: In the nighttime, it produces hydrogen through water electrolysis by using low-cost electric power and stores the hydrogen. In the daytime, it conducts fuel cell generation using the stored hydrogen to meet the demand. Hydrogen storage equipment is the core of THEUS. In this paper, the metal hydride tank was chosen as hydrogen storage. The hydrogen desorption process is endothermic reaction. It is possible to use the cool heat for air conditioning. It is necessary to develop a useful numerical simulation code for not only the operation method but also the optimization of the structure of the tank. We developed the simulation code. The validity of the code has been improved by comparing with experimental results. Concretely, the following points were considered. (1)The thermal capacity of the tank. (2)Heat loss to the outside. (3)The copper plate effect. The simulation results become in good agreement with experimental results. And to achieve hydrogen high flow rate, new design tank performance was investigated.

      • Logic Operations of Chemically Assembled Single-Electron Transistor

        <P>Double-gate single-electron transistors (SETs) were fabricated by chemical assembling using electroless gold-plated nanogap electrodes and chemisorbed chemically synthesized gold nanoparticles. The fabricated SET showed periodic and stable Coulomb oscillations under application of voltages of both gates. The sole SET also exhibited all two-input logic operations?XOR, XNOR, NAND, OR, NOR, and AND?with an on/off ratio of 10<SUP>2</SUP>.
