      • Effects of L-malic acid on alpha-glucosidase: inhibition kinetics and computational molecular dynamics simulations.

        <P>The inhibitory effect of L-malic acid (MA) on alpha-glucosidase (EC 3.2.1.20) was investigated by combination study between inhibition kinetics and computational simulations. The results from the serial kinetics demonstrated that MA could directly inactivate the enzyme activity in a dose-dependent manner and a typical non-competitive type, as well as in a fast inactivate process without detectable time course. The tertiary conformation study with an application of spectrofluorimetry showed that MA modulated the tertiary structural conformation of alpha-glucosidase both on the overall and on regional active site pocket, which monitored by red-shift intrinsic fluorescence peak with decreases intensities, and the significant intensity increasing of 1-anilinonaphthalene-8-sulfonate (ANS)-binding fluorescence, respectively. To have more insight, we also adapted the computational molecular dynamics (MD) simulations. The results showed that MA was located in the entrance of active pocket for the catalytic reaction and blocked the passage of substrate. It confirmed that MA inhibits as a non-competitive type, not direct docking to the glucose binding site. Our study provides important molecular mechanisms to figure out alpha-glucosidase inhibition that might associate to development of type 2 diabetes mellitus drug.</P>

        Physical property effects of the compression process with supercritical carbon dioxide as working fluid

        The compressor is one of the key components in the closed supercritical carbon dioxide (S-CO 2 ) Brayton cycle, but its design method is far from mature. It is naturally expected that the well-established design method of the air compressor can provide favorable guidelines, on the basis of further understanding the effects of the physical property on the compressor flow field. Considering that isentropic compression is one of the core physical processes in the compressor, the physical property effects on this process were mainly investigated in this work. Similarity criterion was considered, and the change rate discrepancy of the main variables in this process between S-CO 2 and the ideal air was fully analyzed. Results show that S-CO 2 is compressed faster than the ideal air in most cases, along with generating smaller Mach number and larger pressure rise ratio. It is noted the important parameter of the static pressure coefficient distribution with S-CO 2 in the compression process is almost the same as that with the ideal air at low Mach number, which is conductive to the extension of the air compressor research experience, but it is quite different at high Mach number. The simulation cases about compressor cascade are further applied and prove the suitability of the revealed physical property effects in the compressor passage. Understanding these effects on the compression process is helpful to improve the design method of the S-CO 2 compressor.

        There are as much as 12 departments in China social security management system, which has a serious impact on a unified decision-making and management of social security undertakings. Learning from the world`s major countries` experiences and according to the theory of government and division of labor, this paper proposes to merge and unify the functions of the various departments of decentralized management of social security into a complete social security Super-Ministry System, and construct a four-in-one system of social security management which contains legislation, administration, fund revenues and expenditures, and supervision to meet the objectives and requirements of the service-oriented government building, to achieve standardized management of social security undertakings, and to promote the healthy and sustainable development of social security undertakings.

        An Asynchronous Burst Time Plan Generation Method for Broadband Satellite Multimedia System

        In broadband satellite multimedia (BSM) system, burst time plan (BTP) is always periodically generated. We find that this method can have a great effect on the system response ability to bandwidth requests. A general analysis model of BTP generation method is given. An optimized BTP generation (O-BTPG) method is presented by deducing the optimal bandwidth allocation period (BAP) and bandwidth allocation latency (BAL) without considering the signaling overhead caused by BTP. Then a novel asynchronous BTP generation (A-BTPG) method in which the BTP is generated asynchronously according to the traffic load from users` bandwidth requests is proposed. Simulation results show that A-BTPG can flexibly realize a trade-off between the system response ability and BTP signaling overhead. What`s more, it can be widely used in various regenerative onboard switching BSM systems

        RNA Interference-Based Transgenic Maize Resistant to Maize Dwarf Mosaic Virus

        Maize dwarf mosaic virus (MDMV) is a widespread pathogenic virus that causes serious loss of yield in maize (Zea mays). RNA interference (RNAi) triggered by hairpin RNA (hpRNA) transcribed from a transgenic inverted-repeat sequence is an effective way to defend against viruses in plants. In this study, an hpRNA expression vector containing a sense arm and an antisense arm of 150 bp separated by an intron of the maize actin gene was constructed to target the P1 protein (protease)gene of MDMV and used to transform Agrobacterium tumefaciens strain EHA105. The transformed Agrobacterium strain was used to transform maize embryonic calli isolated from immature embryos by an improved culture technique. In all, 46 plants were regenerated after stringent hygromycin B selection, and 18 of them were certified to be positive by PCR amplification. Of these positive plants,13 were grown to produce offspring, and nine were identified by Southern blotting to have the transgene integrated with one or two copies. The resistance of three T2 lines was evaluated in a field trial of dual MDMV inoculation in two environments and was found to be improved compared with the non-transformed control. The disease indexes of the transgenic plant lines h2, 13, and h1were not significantly different from the highly resistant control line H9-21. The viral titers of the inoculated plants were detected by double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA), and the result was in accord with the resistance evaluated in the field trial. The addition of uniconazole S3307 (0.25 mgl−1) and ABT rootpromoting powder (0.5 mgl−1) showed a significant improvement of hardening in regenerated plantlets, which were stronger and generated a better fibrous root system than the control. This improvement could facilitate the transgenic operation of maize.

        A Single-Arm Phase II Study of Nab-Paclitaxel Plus Gemcitabine and Cisplatin for Locally Advanced or Metastatic Biliary Tract Cancer

        Purpose Patients with advanced biliary tract cancer (BTC) have a poor survival. We aim to evaluate the efficacy and safety of nab-paclitaxel plus gemcitabine and cisplatin regimen in Chinese advanced BTC patients.Materials and Methods Eligible patients with locally advanced or metastatic BTC administrated intravenous 100 mg/m<sup>2</sup> nab-paclitaxel, 800 mg/m<sup>2</sup> gemcitabine, and 25 mg/m<sup>2</sup> cisplatin every 3 weeks. The primary endpoint was progression-free survival (PFS). The secondary endpoints included overall survival (OS) and adverse events, while exploratory endpoint was the association of biomarkers with efficacy.Results After the median follow-up of 25.0 months, the median PFS and OS of 34 enrolled patients were 7.1 months (95% confidence interval [CI], 5.4 to 13.7) and 16.4 months (95% CI, 10.9 to 23.6), respectively. The most common treatment-related adverse events at ≥ 3 grade were neutropenia (26.5%) and leukopenia (26.5%). Survival analyses demonstrated that carcinoembryonic antigen (CEA) levels could monitor patients’ survival outcomes. A significant increase in the number of infiltrating CD4+ cells (p=0.008) and a decrease in programmed death-1–positive (PD-1+) cells (p=0.032) were observed in the response patients.Conclusion In advanced BTC patients, nab-paclitaxel plus gemcitabine and cisplatin regimen showed therapeutic potential. Potential prognostic factors of CEA levels, number of CD4+ cells and PD-1+ cells may help us maximize the efficacy benefit.
