        Maternal betaine supplementation ameliorates fatty liver disease in offspring mice by inhibiting hepatic NLRP3 inflammasome activation

        BACKGROUND/OBJECTIVES: Previous research has shown maternal betaine supplementation alleviates fetal-derived hepatic steatosis. Therefore, this study examined the anti-inflammatory effect of maternal betaine intake in offspring mice and its mechanism. MATERIALS/METHODS: Female C57BL/6J mice and their offspring were randomly divided into 3 groups according to the treatment received during gestation and lactation: control diet (CD), fatty liver disease (FLD), and fatty liver disease + 1% betaine (FLD-BET). The FLD group was given a high-fat diet and streptozotocin (HFD + STZ), and the FLD-BET group was treated with HFD + STZ + 1% betaine. After weaning, the offspring mice were given a normal diet for 5 weeks and then dissected to measure the relevant indexes. RESULTS: Compared to the CD group, the offspring mice in the FLD group revealed obvious hepatic steatosis and increased serum levels of alanine aminotransferase, interleukin (IL)-6, and tumor necrosis factor (TNF)-α; maternal betaine supplementation reversed these changes. The hepatic mRNA expression levels of IL-6, IL-18, and Caspase-1 were significantly higher in the FLD group than in the CD group. Maternal betaine supplementation reduced the expression of IL-1β, IL-6, IL-18, and apoptosis-associated speck-like protein containing C-terminal caspase recruitment domain (ASC). Maternal betaine supplementation also reversed the increasing protein expressions of nitric oxide dioxygenase-like receptor family pyrin domain containing 3 (NLRP3), ASC, Caspase-1, IL-1β, and IL-18 in offspring mice exposed to HFD + STZ. Maternal betaine supplementation decreased the homocysteine (Hcy) and s-adenosine homocysteine (SAH) levels significantly in the livers. Furthermore, the hepatic Hcy concentrations showed significant inverse relationships with the mRNA expression of TNF-α, NLRP3, ASC, and IL-18. The hepatic SAH concentration was inversely associated with the IL-1β mRNA expression. CONCLUSIONS: The lipotropic and anti-inflammatory effect of maternal betaine supplementation may be associated with the inhibition of NLRP3 inflammasome in the livers of the offspring mice.

        Fluidelastic instability of a tube array in two-phase cross-flow considering the effect of tube material

        Fluidelastic instability of a tube array is a key factor of the security of a nuclear power plant. An unsteady model of the fluidelastic instability of a tube array subjected to two-phase flow was developed to analyze the fluidelastic instability of tube bundles in two-phase flow. Based on this model, a computational program was written to calculate the eigenvalue and the critical velocity of the fluidelastic instability. The unsteady model and the program were verified by comparing with the experimental results reported previously. The influences of void fraction and the tube's material properties on the critical velocity were investigated. Numerical results showed that, with increasing the void fraction of the two-phase flow, the tube array becomes more stable. The results indicate that the critical velocities of the tube array made of stainless are much higher than those of the other two tube arrays within void fraction ranging from 20%to 80%.

        Genome-wide analyses reveal the extent of opportunistic STAT5 binding that does not yield transcriptional activation of neighboring genes

        <P>Signal Transducers and Activators of Transcription (STAT) 5A/B regulate cytokine-inducible genes upon binding to GAS motifs. It is not known what percentage of genes with GAS motifs bind to and are regulated by STAT5. Moreover, it is not clear whether genome-wide STAT5 binding is modulated by its concentration. To clarify these issues we established genome-wide STAT5 binding upon growth hormone (GH) stimulation of wild-type (WT) mouse embryonic fibroblasts (MEFs) and MEFs overexpressing STAT5A more than 20-fold. Upon GH stimulation, 23 827 and 111 939 STAT5A binding sites were detected in WT and STAT5A overexpressing MEFs, respectively. 13 278 and 71 561 peaks contained at least one GAS motif. 1586 and 8613 binding sites were located within 2.5 kb of promoter sequences, respectively. Stringent filtering revealed 78 genes in which the promoter/upstream region (−10 kb to +0.5 kb) was recognized by STAT5 both in WT and STAT5 overexpressing MEFs and 347 genes that bound STAT5 only in overexpressing cells. Genome-wide expression analyses identified that the majority of STAT5-bound genes was not under GH control. Up to 40% of STAT5-bound genes were not expressed. For the first time we demonstrate the magnitude of opportunistic genomic STAT5 binding that does not translate into transcriptional activation of neighboring genes.</P>

        Preparation and Preliminary Evaluation of Macroporous Magnetic Agarose Particles for Bioseparation

        Macroporous magnetic agarose particles (MMAPs) were prepared with calcium carbonate as the porogent by the water-in-oil suspension thermal regeneration method. MMAPs with good sphericity and appropriate particle size were obtained. The physical properties of the beads were determined and it was found that the water content (92.1%), porosity (94.4%) and mean pore diameter (120.1 nm) of the MMAPs were higher than those for the normal magnetic particles, indicating successful generation of macropores after calcium carbonate addition. Compared with normal magnetic particles, the mass transfer of biomolecules in MMAPs was remarkably enhanced. Finally, MMAPs were modified with 5-amino-benzimidazol (ABI) ligand and the adsorption capacity of IgG reached 153 mg/mL, higher than that of the normal magnetic particles (126 mg/mL). Moreover, adsorption behavior of MMAPs to IgG was little changed after twenty-five recycled use. Hence, MMAPs prepared herein showed great potential for bioseparation.
