        Comprehensive Echocardiographic Assessment of the Right Ventricle in Murine Models

        Background: Non-invasive high-resolution echocardiography to evaluate cardiovascular function of small animals is increasingly being used due to availability of genetically engineered murine models. Even though guidelines and standard values for humans were revised by the American Society of Echocardiography, evaluations on murine models are not performed according to any standard protocols. These limitations are preventing translation of preclinical evaluations to clinical meaningful conclusions. We have assessed the right heart of two commonly used murine models according to standard clinical guidelines, and provided the practical guide and sample values for cardiac assessments. Methods: Right heart echocardiography evaluations of CD1 and C57BL/6 mice were performed under 1–3% isoflurane anesthesia using Vevo® 2100 Imaging System with a high-frequency (18–38 MHz) probe (VisualSonics MS400). We have provided a practical guide on how to image and assess the right heart of a mouse which is frequently used to evaluate development of right heart failure due to pulmonary hypertension. results: Our results show significant differences between CD1 and C57BL/6 mice. Right ventricle structural assessment showed significantly larger (p < 0.05) size, and pulmonary artery diameter in CD1 mice (n = 11) compared to C57BL/6 mice (n = 15). Right heart systolic and diastolic functions were similar for both strains. Conclusion: Our practical guide on how to image and assess the right heart of murine models provides the first comprehensive values which can be used for preclinical research studies using echocardiography. Additionally, our results indicate that there is a high variability between mouse species and experimental models should be carefully selected for cardiac evaluations.

      • The Features of Ukrainian Media Art in a Global Context

        The article seeks to explore Ukrainian media art and its features in a global context. In particular, it performs an in-depth analysis of the stages of its development from video art of the 1990s, media installations of the 2000s, and to various digital and VR technologies today. Due to historical circumstances, the development of media art was quite rapid, as young artists sought to gain new experience in media art. Most often, their experience was broadened through international cooperation and studying abroad. The paper analyzes the presentation of Ukrainian media art outside the country during 1993-2020 and distinguishes the main thematic areas of the artists' work. It examines how artists present and reveal the peculiarities of the Ukrainian worldview, aesthetics, and culture through the problematization of certain aspects and themes in their work. Specifically, among the principal topics which are problematized in the work of media artists are a sensory experience, limited sensory capabilities, gender issues, the Chernobyl tragedy, the development of utopian models of the city, and global communication. The leading themes of Ukrainian artists also include revolution and war. Notably, political and social sentiments determine the unique energy and vitality of contemporary Ukrainian art, create a revolutionary creative environment, and unite media artists in group art projects. It is concluded that by showing, exhibiting, and presenting them outside the country, artists shape the image of Ukrainian art in a global context.

        Crawler robot kinematics modeling by using a two-wheeled approach

        The aim of this paper is to analyze the kinematics of a small crawler robot. A mathematical model of kinematics based on a twowheeled approach is proposed. This model is experimentally verified using vision-based motion measurements of a crawler vehicle equipped with encoders and a remote control system. It is assumed that the vehicle moves along a few curves with different angular speeds of its wheels. Based on the model of motion and the values of these speeds, numerical simulations are investigated. The results obtained from numerical and experimental validation are presented and discussed. The comparison delivered some important conclusions.

        A case study of large phytoplankton blooms off the New Jersey coast with multi-sensor observations

        Taking the 2011 summer bloom as a study case, we investigated the transport and nutrient delivery mechanisms for large phytoplankton blooms of the New Jersey coast. The horizontal advection of the chlorophyll is approximately one order larger than the horizontal diffusion averaging the whole study area. And the sea surface currents are found to correlate with the wind stress but only for part of the middle shelf. Based on the horizontal advection simulation driven by HF radar current, the bloom is divided into three sub-regions. The northern coastal section of the bloom shows high-low-high pattern in chlorophyll time series, which is related to coastal upwelling and a clockwise eddy. Coastal currents play an important role in the eddy formation and the southward transport of the bloom. The southern coastal section of the bloom is supported by the nutrients from coastal upwelling and driven offshore by wind. The northeast section of the bloom is fueled by nutrient source upwelled through Ekman pumping. The magnitude of Ekman pumping is of the same order as the magnitude of the upwelling along the coast. These identified mechanisms could also applied to more than half of bloom events during 2002 to 2013. In the summer of 2011 off the coast of New Jersey, processes in different sub-regions worked together, leading to this large phytoplankton bloom.

        Use of Speckle Tracking Echocardiography to Detect Induced Regional Strain Changes in the Murine Myocardium by Acoustic Radiation Force

        BACKGROUND: It is difficult to simulate the abnormal myocardial strain patterns caused by ischemic coronary artery disease (CAD) which are a precursor to heart failure (HF) within an animal model. Simulation of these strain changes could contribute to better understanding of the early formative stages of HF. This is especially important in investigating the poorly understood pathogenesis of heart failure with preserved ejection fraction (HFpEF). Here, we discuss delivery of high intensity focused ultrasound (HIFU) in a murine model to alter left ventricular (LV) regional longitudinal strain (RLS), and use of speckle tracking echocardiography to detect these changes. METHODS: HIFU pulses (pressure amplitude 1.7 MPa) were generated by amplifying a sinusoidal waveform from a function generator into a piezoelectric transducer. These pulses were then directed extracorporeally towards the anterior LV surface of C57BI6 mice during three time periods (early, mid, and late diastole). Speckle tracking echocardiography was then used to quantify changes in RLS within six segments of the LV. RESULTS: We observed an increase in LV RLS with acoustic augmentation during all three time periods. This augmentation was most prominent near the anterior apical region in early diastole and near the posterior basilar region during late diastole. CONCLUSIONS: Our findings demonstrate the application of HIFU to non-invasively induce changes in RLS within a murine model. Our results also reflect the capability of speckle tracking echocardiography to analyze and quantify these changes. These findings represent the first demonstration of ultrasound-induced augmentation in LV RLS within a small animal model.

        Superior Vena Cava Echocardiography as a Screening Tool to Predict Cardiovascular Implantable Electronic Device Lead Fibrosis

        Background: Currently there is no noninvasive imaging modality used to risk stratify patients requiring lead extractions. Wereport the novel use of superior vena cava (SVC) echocardiography to identify lead fibrosis and complex cardiac implantableelectronic device (CIED) lead extraction. With an aging population and expanding indications for cardiac device implantation,the ability to deal with the complications associated with chronically implanted device has also increased. Methods: This was a retrospective analysis of Doppler echocardiography recorded in our outpatient Electrophysiology/DeviceClinic office over 6 months. Images from 109 consecutive patients were reviewed. Results: 62% (68/109) did not have a CIED and 38% (41/109) had a CIED. In patients without a CIED, 6% (4/68) displayedturbulent color flow by Doppler in the SVC, while 22% (9/41) of patients with a CIED displayed turbulent flow. Fisher’s exacttest found a statistically significant difference between the two groups (p value < 0.05). The CIED group was subdivided into 2groups based on device implant duration (< 2 years vs. ≥ 2 years). Of the CIED implanted for ≥ 2 years, 27% (9/33) hadturbulent flow in the SVC by Doppler, while no patients (0/8) with implant durations < 2 years demonstrated turbulent flow. Nine patients underwent subsequent lead extraction. A turbulent color pattern successfully identified all 3 patients that hadsignificant fibrosis in the SVC found during extraction. Conclusion: Our data suggests that assessing turbulent flow using color Doppler in the SVC may be a valuable noninvasivescreening tool prior to lead extraction in predicting complex procedures.

        Enhancement of Innate and Adaptive Immune Functions by Multiple Echinacea Species

        Echinaceapreparations are commonly used as nonspecific immunomodulatory agents. Alcohol extracts fromthree widely used Echinaceaspecies, Echinacea angustifolia, Echinacea pallida, and Echinacea purpurea, were investigatedfor immunomodulating properties. The three Echinaceaspecies demonstrated a broad difference in concentrations of indi-vidual lipophilic amides and hydrophilic caffeic acid derivatives. Mice were gavaged once a day (for 7 days) with one of theEchinaceaextracts (130 mg/kg) or vehicle and immunized with sheep red blood cells (sRBC) 4 days prior to collection ofimmune cells for multiple immunological assays. The three herb extracts induced similar, but differential, changes in the per-centage of immune cell populations and their biological functions, including increased percentages of CD49. and CD19.lymphocytes in spleen and natural killer cell cytotoxicity. Antibody response to sRBC was significantly increased equally byextracts of all three Echinacea species. Concanavalin A-stimulated splenocytes from E. angustifolia- and E. pallida-treatedmice demonstrated significantly higher T cell proliferation. In addition, the Echinaceatreatment significantly altered the cy-tokine production by mitogen-stimulated splenic cells. The three herbal extracts significantly increased interferon-. produc-tion, but inhibited the release of tumor necrosis factor-. and interleukin (IL)-1.. Only E. angustifolia- and E. pallida-treatedmice demonstrated significantly higher production of IL-4 and increased IL-10 production. Taken together, these findingsdemonstrated that Echinaceais a wide-spectrum immunomodulator that modulates both innate and adaptive immune responses.In particular, E. angustifoliaor E. pallidamay have more anti-inflammatory potential.
