Effects of Retinoic Acid on Regulation of Cell Cycle in Human Gingival Fibroblasts

Retinoic acid가 새포증식과 분화 그리고 노화에 매우 중요한 역할을 수행하는 것으로 알려져 있다. 그러나 retinoic acid가 인간치은섬유모세포의 세포주기조절에 미치는 영향에 관해서는 아직 연구된바 없다. 따라서 본 연구는 인간치은섬유모세포에 all-rrenr retinoic acid (RA)를 투여하여 세포주기조절에 미치는 영향을 평가하였다. 0.1nM-1μM의 RA는 인간치은섬유모세포의 세포중식을 촉진시켰으며, 그 중 1nM에서 최대증식촉진 효과를 나태냄을 알 수 있었다. 1nM RA를 인간치은섬유모세포에 투여하여 세포주기변화를 관찰한 결과, 세포주기 진행이 대조군에 비하여' 1 nM RA를 투여한 군에서 현저히 증가됨을 관찰할 수 있었다. 따라서 RA가 인간치은섬유모세포의 세포주기조절에 분자기전의 일부를 규명하고자 세포주기조절에 관여하는 유전자의 발현을 관찰하였다. 1nM RA는 cyclin E와 CDK 2의 발현을 증가시켰으며, p21과 p16^INK4A의 발현은 억제하였다. 그러나 1nM RA는 cyclin D, CDK 4, CDK 6 및 p53의 발현에 큰 영향을 주지 않았다. 이러한 결과는 1nM RA가 인간 치은섬유모세포의 세포주기진행을 촉진시키며, 그 분자기전은 cyclin E와 CDK 2의 발현을 증가시키고, 921과 P16^1NK4A의 발현은 억제하여 이루어지는 것으로 추정된다.

<P>A compact model of the current-voltage (I-V) characteristics for the Si nanowire field effect transistor (FET) taking into account dependence of the analytical electrical properties on the diameter and the concentration of the Si nanowire of the FETs with a Schottky metal-semiconductor contact has been proposed. I-V characteristics of the nanowire FETs were analytically calculated by using a quantum drift-diffusion current transport model taking into account an equivalent circuit together with the quantum effect of the Si nanowires and a Schottky model at Schottky barriers. The material parameters dependent on different diameters and concentrations of the Si nanowire were numerically estimated from the physical properties of the Si nanowire. The threshold voltage, the mobility, and the doping density of the Si nanowire and the Schottky barrier height at a metal-Si nanowire heterointerface in the nanowire FET were estimated by using the theoretical model.</P>

Background: The quality of a vaccine depends strongly on the effects of the adjuvants applied simultaneously with the antigen in the vaccine. The adjuvants enhance the protective effect of the vaccine against a viral challenge. Conversely, oil-type adjuvants leave oil residue inside the bodies of the injected animals that can produce a local reaction in the muscle. The long-term immunogenicity of mice after vaccination was examined. ISA206 or ISA15 oil adjuvants maintained the best immunity, protective capability, and safety among the oil adjuvants in the experimental group. Objectives: This study screened the adjuvant composites aimed at enhancing foot-and-mouth disease (FMD) immunity. The C-type lectin or toll-like receptor (TLR) agonist showed the most improved protection rate. Methods: Experimental vaccines were fabricated by mixing various known oil adjuvants and composites that can act as immunogenic adjuvants (gel, saponin, and other components) and examined the enhancement effect on the vaccine. Results: The water in oil (W/O) and water in oil in water (W/O/W) adjuvants showed better immune effects than the oil in water (O/W) adjuvants, which have a small volume of oil component. The W/O type left the largest amount of oil residue, followed by W/O/W and O/W types. In the mouse model, intramuscular inoculation showed a better protection rate than subcutaneous inoculation. Moreover, the protective effect was particularly weak in the case of inoculation in fatty tissue. The initial immune reaction and persistence of long-term immunity were also confirmed in an immune reaction on pigs. Conclusions: The new experimental vaccine with immunostimulants produces improved immune responses and safety in pigs than general oil-adjuvanted vaccines.

Objective : The extensive vasa vasorum network functions as a conduit for the entry of inflammatory cells or factors that promote the progression of angiogenesis and plaque formation. Therefore, we investigated the correlation between the carotid vasa vasorum activities and carotid plaque vulnerability using indocyanine green video angiography (ICG-VA) during carotid endarterectomy (CEA). Methods : Sixty-nine patients who underwent CEA were enrolled prospectively from September 2015 to December 2017. During CEA, a bolus of ICG was injected intravenously before and after resecting the atheroma. Additionally, we performed immunohistochemistry using CD68 (a surface marker of macrophages), CD117 (a surface marker of mast cells), and CD4 and CD8 (surface markers of T-cells) antibodies to analyze the resected plaque specimens. Results : The density of active vasa vasorum was observed in all patients using ICG-VA. The vasa vasorum externa (VVE) and interna (VVI) were seen in 11 (16%) and 57 patients (82.6%), respectively. Macroscopically, the VVE-type patterns were strongly associated with preoperative angiographic instability (81.8%, p=0.005) and carotid plaque vulnerability (90.9%, p=0.017). In contrast, the VVI-type patterns were weakly associated with angiographic instability (31.6%) and plaque vulnerability (49.1%). CD68-stained macrophages and CD117-stained mast cells were observed more frequently in unstable plaques than in stable plaques (p<0.0001, p=0.002, respectively). Conclusion : The early appearance of VVE, along with the presence of many microvessel channels that provided nutrients to the developing and expanding atheroma during ICG-VA, was strongly associated with unstable carotid plaques. The degree of infiltration of macrophages and mast cells is possibly related to the formation of unstable plaques.
