        Mite and Booklouse Fauna From Vacuumed Dust Samples From Beijing

        A significant-source of allergens come from house dust that contain particles derived from arthropods, molds, and pet dander. This study evaluatedmite and booklouse fauna from vacuumed dust samples in Beijing China (a temperate zone). Our survey was carried out in Beijing in the homes ofmite allergic patients who visited our Allergy Department. In total, 38 homes were selected for the collection of dust samples by vacuuming, fromDecember 2008 to January 2010. The flotation method was used to isolate mites from house dust. Permanent slides were prepared for mite specimensand mites were identified and counted under a microscope. In total, 1,798 separate mite and insect specimens were found in 345 dust samplestaken from 38 homes. A total of 95 individual Dermatophagoides (D) siboney were detected in 35 dust samples from 19 homes (representing5.3% of all mite and insect species found in house dust); in addition, this mite was found to co-exist with D. farinae (Hughes, 1961) in 33 dust samples. Our results demonstrated the presence D. siboney that co-existed with D. farinae in house dust in Beijing China (a temperate zone).

      • Privacy-preserving Multi-keyword Ranked Search over Encrypted Cloud Data Supporting Dynamic Update

        With the development of cloud computing, the sensitive information of outsourced data is at risk of unauthorized accesses. To protect data privacy, the sensitive data should be encrypted by the data owner before outsourcing, which makes the traditional and efficient plaintext keyword search technique useless. Hence, it is an especially important thing to explore secure encrypted cloud data search service. Considering the huge number of outsourced data, there are three problems we are focused on to enable efficient search service: multi-keyword search, result relevance ranking and dynamic update. In this paper, we propose a practically efficient and flexible searchable encrypted scheme which supports both multi-keyword ranked search and dynamic update. To support multi-keyword search and result relevance ranking, we adopt Vector Space Model (VSM) to build the searchable index to achieve accurate search result. To improve search efficiency, we design a tree-based index structure which supports insertion and deletion update well without privacy leakage. We propose a secure search scheme to meet the privacy requirements in the threat model. Finally, experiments on real-world dataset are implemented to demonstrate the overall performance of the proposed scheme, which show our scheme is efficient.

      • Efficient Keyword Search Scheme in Encrypted Cloud Computing Environment

        With the increasing popularity of cloud computing, more and more sensitive or private information has been outsourced onto the cloud server. For protecting data privacy, sensitive data usually has to be encrypted before outsourcing, which makes traditional search techniques based on plaintext useless. In response to the search over encrypted data, searchable encryption is a good solution in Information Security. However, most of existing searchable encryption schemes only support exact keyword search. That means they don’t support searching for different variants of the query word, which is a significant drawback and greatly affects data usability and user experience. Recently, a fuzzy keyword search scheme proposed by some researchers aims at addressing the problems of minor typos and format inconsistence but couldn’t solve the problem above. In this paper, we formalize the problem of semantic keyword-based search over encrypted cloud data while preserving privacy. Semantic keyword-based search will greatly improves the user experience by returning all the documents containing semantically close keywords related to the query word. In our solution, we use the stemming algorithm to construct stem set, which reduces the dimension of index. And the symbol-based trie is also adopted in index construction to improve the search efficiency. Through rigorous privacy analysis and experiment on real dataset, our scheme is secure and efficient.

        Functional Connections of the Vestibulo-spino-adrenal Axis in the Control of Blood Pressure Via the Vestibulosympathetic Reflex in Conscious Rats

        Significant evidence supports the role of the vestibular system in the regulation of blood pressure during postural movements. In the present study, the role of the vestibulo-spino-adrenal (VSA) axis in the modulation of blood pressure via the vestibulosympathetic reflex was clarified by immunohistochemical and enzyme immunoassay methods in conscious rats with sinoaortic denervation. Expression of c-Fos protein in the intermediolateral cell column of the middle thoracic spinal regions and blood epinephrine levels were investigated, following microinjection of glutamate receptor agonists or antagonists into the medial vestibular nucleus (MVN) and/or sodium nitroprusside (SNP)-induced hypotension. Both microinjection of glutamate receptor agonists (NMDA and AMPA) into the MVN or rostral ventrolateral medullary nucleus (RVLM) and SNP-induced hypotension led to increased number of c-Fos positive neurons in the intermediolateral cell column of the middle thoracic spinal regions and increased blood epinephrine levels. Pretreatment with microinjection of glutamate receptor antagonists (MK-801 and CNQX) into the MVN or RVLM prevented the increased number of c-Fos positive neurons resulting from SNP-induced hypotension, and reversed the increased blood epinephrine levels. These results indicate that the VSA axis may be a key component of the pathway used by the vestibulosympathetic reflex to maintain blood pressure during postural movements.

      • Rational combination therapy with PARP and MEK inhibitors capitalizes on therapeutic liabilities in <i>RAS</i> mutant cancers

        <P>Mutant <I>RAS</I> has remained recalcitrant to targeted therapy efforts. We demonstrate that combined treatment with poly(adenosine diphosphate–ribose) polymerase (PARP) inhibitors and mitogen-activated protein kinase (MAPK) kinase (MEK) inhibitors evokes unanticipated, synergistic cytotoxic effects in vitro and in vivo in multiple <I>RAS</I> mutant tumor models across tumor lineages where <I>RAS</I> mutations are prevalent. The effects of PARP and MEK inhibitor combinations are independent of <I>BRCA1/2</I> and <I>p53</I> mutation status, suggesting that the synergistic activity is likely to be generalizable. Synergistic activity of PARP and MEK inhibitor combinations in <I>RAS</I> mutant tumors is associated with (i) induction of BIM-mediated apoptosis, (ii) decrease in expression of components of the homologous recombination DNA repair pathway, (iii) decrease in homologous recombination DNA damage repair capacity, (iv) decrease in DNA damage checkpoint activity, (v) increase in PARP inhibitor–induced DNA damage, (vi) decrease in vascularity that could increase PARP inhibitor efficacy by inducing hypoxia, and (vii) elevated PARP1 protein, which increases trapping activity of PARP inhibitors. Mechanistically, enforced expression of FOXO3a, which is a target of the RAS/MAPK pathway, was sufficient to recapitulate the functional consequences of MEK inhibitors including synergy with PARP inhibitors. Thus, the ability of mutant <I>RAS</I> to suppress FOXO3a and its reversal by MEK inhibitors accounts, at least in part, for the synergy of PARP and MEK inhibitors in <I>RAS</I> mutant tumors. The rational combination of PARP and MEK inhibitors warrants clinical investigation in patients with <I>RAS</I> mutant tumors where there are few effective therapeutic options.</P>

        Multiple down-regulated cytochrome P450 monooxygenase genes contributed to synergistic interaction between chlorpyrifos and imidacloprid against Nilaparvata lugens

        Insecticide mixtures are an effective strategy in pest resistance management. The synergistic chlorpyrifos and imidacloprid mixture could significantly increase toxicity against rice pest, Nilaparvata lugens, despite their high levels of resistance. However, synergism mechanisms to explain this phenomenon remain unknown. Chlorpyrifos and imidacloprid at a 1:0.5 ratio showed significant synergism on N. lugens with a combination index value of 0.18 after topical exposure. We constructed a genetic database of the genes expressed in individual and synergistic chlorpyrifos and imidacloprid treatments of N. lugens using Illumina Hiseq™ X Ten, and 17 co-downregulated genes putatively involved in synergism were detected by comparative transcriptome analyses. Expression patterns of the 17 candidate synergistic genes matched with transcriptome sequencing data by quantitative real-time PCR analyses. Feeding of dsRNAs further reduced the expression levels of 10 of these candidate synergistic genes (from 1.68 to 4.13-fold). Nymphs fed with only dsRNAs of CYP4DE1, CYP6AY1v2, CYP353D1, and CYP439A1 experienced more high mortality rates (81.45–90.34%) to improve synergism between chlorpyrifos and imidacloprid. Multiple reductive expressed P450 genes were potentially associated with synergism of a mixture of chlorpyrifos and imidacloprid, as confirmed by comparative transcriptome analyses and RNAi assays. Our findings suggested that synergistic interactions between chlorpyrifos and imidacloprid might be controlled by P450s.

        BRD4 Inhibition Is Synthetic Lethal with PARP Inhibitors through the Induction of Homologous Recombination Deficiency

        <P><B>Summary</B></P> <P>Poly(ADP-ribose) polymerase inhibitors (PARPi) are selectively active in cells with homologous recombination (HR) deficiency (HRD) caused by mutations in <I>BRCA1</I>, <I>BRCA2</I>, and other pathway members. We sought small molecules that induce HRD in HR-competent cells to induce synthetic lethality with PARPi and extend the utility of PARPi. We demonstrated that inhibition of bromodomain containing 4 (BRD4) induced HRD and sensitized cells across multiple tumor lineages to PARPi regardless of <I>BRCA1/2</I>, <I>TP53</I>, <I>RAS</I>, or <I>BRAF</I> mutation status through depletion of the DNA double-stand break resection protein CtIP (C-terminal binding protein interacting protein). Importantly, BRD4 inhibitor (BRD4i) treatment reversed multiple mechanisms of resistance to PARPi.       • Highly Sensitive Diagnosis of Small Hepatocellular Carcinoma Using pH-Responsive Iron Oxide Nanocluster Assemblies

        <P>Iron oxide nanoparticle (IONP)-based magnetic resonance imaging (MRI) contrast agents have been widely used for the diagnosis of hepatic lesions. However, current IONP-based liver-specific MRI contrast agents rely on single-phase contrast enhancement of the normal liver, which is not sensitive enough to detect early stage small hepatocellular carcinomas (HCCs). We herein report i-motif DNA-assisted pH-responsive iron oxide nanocluster assemblies (termed RIAs), which provide an inverse contrast enhancemt effect to improve the distinction between normal liver and target HCC tissues. The acidic pH of the tumor microenvironment triggers the disassembly of the RIAs, which leads to a drastic decrease in their relaxivity ratio (<I>r</I><SUB>2</SUB>/<I>r</I><SUB>1</SUB>), thus converting the RIAs from a T2 to T1 contrast agent. This inverse contrast enhancement of normal liver darkening and HCC brightening under T1 imaging mode was validated on an orthotopic HCC model. Our design provides a novel strategy for the exploitation of the next-generation intelligent MRI contrast agents.</P> [FIG OMISSION]</BR>

        A study of hydroelastic behavior of hinged VLFS

        This paper introduces a new method to study the hydroelastic behavior of hinged Very Large Floating Structures (VLFSs). A hinged two-module structure is used to confirm the present approach. For each module, the hydroelasticity theory proposed by Lu et al. (2016) is adopted to consider the coupled effects of wave dynamics and structural deformation. The continuous condition at the connection position between two adjacent modules is also satisfied. Then the hydroelastic motion equation can be established and numerically solved to obtain the vertical displacement, force and bending moment of the hinged structure. The results calculated by the present new method are compared with those obtained using three-dimensional hydroelasticity theory (Fu et al., 2007), which shows rather good agreement.
