        KLK6 Promotes Growth, Migration, and Invasion of Gastric Cancer Cells

        Purpose: Kallikrein (KLK) proteases are hormone-like signaling molecules with critical functions in different cancers. This study investigated the expression of KLK6 in gastric cancer and its potential role in the growth, migration, and invasion of gastric cancer cells. Materials and Methods: In this study, we compared protein levels of KLK6, vascular endothelial growth factor (VEGF), and matrix metallopeptidase (MMP) 9 in normal gastric epithelial and gastric cancer cell lines by western blot. Fluorescence-activated cell sorting was employed to sort 2 clones of SGC-7901 cells with distinct KLK6 expression, namely, KLK6-high ($KLK6^{high}$) and KLK6-low ($KLK6^{low}$), which were then expanded. Lastly, immunohistochemical analysis was performed to investigate KLK6 expression in gastric cancer patients. Results: The expression levels of KLK6, VEGF, and MMP 9, were significantly higher in the gastric cancer cell lines SGC-7901, BGC-823, MKN-28, and MGC-803 than in the normal gastric epithelial cell line GES-1. Compared to $KLK6^{low}$ cells, $KLK6^{high}$ cells showed enhanced viability, colony-forming ability, migration, and invasion potential in vitro. Importantly, immunohistochemical analysis of a human gastric cancer tissue cohort revealed that the staining for KLK6, VEGF, and MMP9 was markedly stronger in the cancerous tissues than in the adjacent normal tissues. KLK6 expression also correlated with that of VEGF and MMP9 expression, as well as several key clinicopathological parameters. Conclusions: Together, these results suggest an important role for KLK6 in human gastric cancer progression.

        Effects of Ga Content on Dynamic Recrystallization and Mechanical Properties of High Strain Rate Rolled Mg–Ga Alloys

        The Mg–xGa (x = 1, 2, 3 and 5 in mass%) alloys are subjected to high strain rate rolling (HSRR) at 275 °C with the rollingstrain rate of 9.1 s−1 to develop high performance Mg alloy sheets with high plasticity. Effects of Ga content on microstructureand mechanical properties of the Mg–Ga alloys are investigated by SEM, XRD, tensile testing and etc. The Ga additioncan reduce the critical strain of DRX in Mg alloys, which is associated with the reduced stacking fault energy, the increasedtwinning density during deformation and the more DRX nucleation sites during HSRR. With the Ga content increasing from2 to 3%, the reduced DRX degree is attributable to the hindrance of dynamic precipitates. With the Ga content increasingfrom 3 to 5%, the slightly increased DRX degree can be ascribed to the relatively coarse precipitates. The Mg–2 Ga alloysheet, featured with complete DRX, exhibits an ultra-high plasticity (with the elongation to rupture of 36.6%) and a relativelylow anisotropy of yield strength and plasticity. The Mg–5 Ga alloy sheet has the best comprehensive mechanical properties,with the ultimate tensile strength of 292 MPa, yield strength of 230 MPa and elongation to rupture of 30.3%, which can beascribed to the combination of grain refinement strengthening and precipitation strengthening.

        The Cholesky rank-one update/downdate algorithm for static reanalysis with modifications of support constraints

        Structural reanalysis is frequently utilized to reduce the computational cost so that the process of design or optimization can be accelerated. The supports can be regarded as the design variables and may be modified in various types of structural optimization problems. The location, number, and type of supports can make a great impact on the performance of the structure. This paper presents a unified method for structural static reanalysis with imposition or relaxation of some support constraints. The information from the initial analysis has been fully utilized and the computational time can be significantly reduced. Numerical examples are used to validate the effectiveness of the proposed method.

        Fault diagnosis of rolling bearing using a refined composite multiscale weighted permutation entropy

        The health status information of rolling bearings is often contained in vibration signals, but it is difficult to detect bearing defects directly through vibration signals. To effectively extract the key feature information hidden in the original signal, this paper proposes the refined composite multiscale weighted permutation entropy (RCMWPE) method to efficiently characterize the operating state of the bearing. The proposed method focuses on two aspects:the improved version reduces the dependence of entropy on the length of the original time series, and the error caused by considering the amplitude information is suppressed. The performance of the proposed method is evaluated by synthetic signals and real bearing data, and compared with other traditional methods. By analyzing bearing signals of different fault types and different degrees of damage, it is verified that the proposed method can obtain more stable and reliable results and achieve higher fault diagnosis accuracy.
