Hepatitis C virus (HCV) infection is a global health concern with chronic liver damage threatening 3% of the world's population. To date, the standard of care is a combination of pegylated interferon-alpha with ribavirin, and recently two direct acting antivirals have entered the clinics. However, because of side effects, drug resistance and viral genotype-specific differences in efficacy current and potentially also future therapies have their limitations. Here, we describe the development of a phenotypic high-throughput assay to identify new cross-genotype inhibitors with novel mechanism of action, by combining a genotype (gt) 1 replicon with the infectious HCV gt2 cell culture system. To develop this phenotypic multiplex assay, HCV reporter cells expressing RFP-NLS-IPS and gt1b replicon cells expressing NS5A-GFP were co-plated and treated with compounds followed by inoculation with gt2a HCV. At 72h post treatment, RFP translocation as a marker for HCV infection and GFP fluorescence intensity as a marker for gt1 RNA replication were measured. Additionally, the total cell number, which serves as an indicator of cytotoxicity, was determined. This phenotypic strategy supports multi-parameter data acquisition from a single well to access cross-genotypic activity, provides an indication of the stage of the viral life cycle targeted, and also assesses compound cytotoxicity. Taken together, this multiplex phenotypic platform facilitates the identification of novel compounds for drug development and chemical probes for continuing efforts to understand the HCV life cycle.
Robotic-assisted Total Hip Arthroplasty and Spinopelvic Parameters: A Review

Total hip arthroplasty (THA) is an effective treatment for osteoarthritis, and the popularity of the direct anterior approach has increased due to more rapid recovery and increased stability. Instability, commonly caused by component malposition, remains a significant concern. The dynamic relationship between the pelvis and lumbar spine, deemed spinopelvic motion, is considered an important factor in stability. Various parameters are used in evaluating spinopelvic motion. Understanding spinopelvic motion is critical, and executing a precise plan for positioning the implant can be difficult with manual instrumentation. Robotic and/or navigation systems have been developed in the effort to enhance THA outcomes and for implementing spinopelvic parameters. These systems can be classified into three categories: X-ray/fluoroscopy-based, imageless, and computed tomography (CT)-based. Each system has advantages and limitations. When using CT-based systems, preoperative CT scans are used to assist with preoperative planning and intraoperative execution, providing feedback on implant position and restoration of hip biomechanics within a functional safe zone developed according to each patient’s specific spinopelvic parameters. Several studies have demonstrated the accuracy and reproducibility of robotic systems with regard to implant positioning and leg length discrepancy. Some studies have reported better radiographic and clinical outcomes with use of robotic-assisted THA. However, clinical outcomes comparable to those for manual THA have also been reported. Robotic systems offer advantages in terms of accuracy, precision, and potentially reduced rates of dislocation. Additional research, including conduct of randomized controlled trials, will be required in order to evaluate the long-term outcomes and cost-effectiveness of robotic-assisted THA

The Kerr gate technique is used to time-resolve the gain in an In0:02Ga0:98N/In0:16Ga0:84N multiple quantum well sample. A newwayofanalyzingthe datain suchavariable stripelength methodgainexperiment isusedto analyzeboththetime-integrated andtime-is caused by the change of the chemical potential along the excited stripe due to the interaction of the carrier and photon densities, andthegain thresholddensityisestimated. This is found to t very well with our data, even beyond the saturation region. Fur-thermore, we have extended the investigation to examine the dynamics of the emission and gain. <P>The Taiwanese-American Occultation Survey project is designed for the detection of stellar occultations by small-size Kuiper Belt Objects, and it has monitored selected fields along the ecliptic plane by using four telescopes with a 3 deg<SUP>2</SUP> field of view on the sky since 2005. We have analyzed data accumulated during 2005-2012 to detect variable stars. Sixteen fields with observations of more than 100 epochs were examined. We recovered 85 variables among a total of 158 known variable stars in these 16 fields. Most of the unrecovered variables are located in the fields observed less frequently. We also detected 58 variable stars which are not listed in the International Variable Star Index of the American Association of Variable Star Observers. These variable stars are classified as 3 RR Lyrae, 4 Cepheid, 1 δ Scuti, 5 Mira, 15 semi-regular, and 27 eclipsing binaries based on the periodicity and the profile of the light curves.</P>

<P>The progress made in understanding spontaneous toroidal rotation reversals in tokamaks is reviewed and current ideas to solve this ten-year-old puzzle are explored. The paper includes a summarial synthesis of the experimental observations in AUG, C-Mod, KSTAR, MAST and TCV tokamaks, reasons why turbulent momentum transport is thought to be responsible for the reversals, a review of the theory of turbulent momentum transport and suggestions for future investigations.</P>
