        Contents of Polyphenols and Limonoids in Citron (Citrus junos Sieb. ex Tanaka) Seed Extracts and Their Antioxidant Properties

        Contents of phytochemicals in citron (Citrus junos Sieb. ex Tanaka) seeds and their effects on antioxidant activities were investigated. Methanol extract of defatted citron seeds contained the highest amounts of total polyphenols followed by 70% ethanol and water extracts. Neohesperidin was the most predominant citrus flavanones in these extracts. The highest amounts of limonoids were found in methanol extract, and this extract was the most efficient in scavenging both DPPH and ABTS radicals. All 3 extracts also exhibited good antioxidant activities against attack of linoleate free radicals on β-carotene. When methanol extract was sequentially fractionated into ether, ethyl acetate, n-butanol, and water fractions, butanol fraction contained the highest amounts of polyphenols otherwise most of limonoids were concentrated in ethyl acetate fraction. A positive relationship between radical scavenging activities and total polyphenol contents in fractions was observed while antioxidant activity on β-carotene seemed more related with contents of limonoids and other hydrophobic polyphenols.

        Flavonoids and Functional Properties of Germinated Citron (Citrus junos Sieb. ex TANAKA) Shoots

        The main objective of this study was to investigate possible application of citron (Citrus junos Sieb. ex TANAKA) seeds, which are massively produced as by-products during citron tea process, into functional food materials. First of all, citron seeds were germinated and produced citron shoots were examined for their functional properties. When contents of flavonoids in citron seeds and their germinated shoots were compared, naringenin, neohesperitin, and hesperitin were remarkably increased in shoots after germination while naringin and didymin were decreased. Concentrations of limonin and nomilin were decreased by germination otherwise their unidentified derivatives were newly formed. A methanol extract of citron shoot had lower IC?? values [0.13 and 0.07 ㎎/㎖ for 2,2-diphenyl-1-picryl-hydrazyl (DPPH) and ABTS, respectively] than citron seed extract in radical scavenging activities. Addition of 500 mg/mL of citron shoot extract suppressed fat accumulation in 3T3-L1 adipocytes by 36.9%. Oral administration of olive oil along with citron shoot extract (33 ㎎/㎏ body weight) to Sprague Dawley rats effectively inhibited absorption of lipid into a body by decreasing blood triglyceride levels from 105.1 to 74.9 ㎎/㎗ 2 hr after olive oil administration. According to these results, citron shoot extract as a rich source of flavonoids can be utilized for functional food ingredients with effective antioxidant and anti-adipogenic properties.

        가상현실 금연클리닉을 위한 시나리오 개발 및 정보구조 디자인에 대한 연구

        최근 국내 교육기관과 지자체 하물며 기업에서도 유래에 없는 흡연에 대한 강력하고 엄격한 규제를 실시하고 있으며 정부 정책과 맞추어 더욱 확대될 전망이다. 중독성 흡연행위는 질환으로 인식 되어져서 금연을 위한 여러 치료방법이 소개되어져 왔으나 시간 및 장소의 제약 그리고 운영 상 많은 한계를 가지고 있어 사회적인 수요에 대비하지 못하고 있다. 이러한 필요에 의해서 여러 중독성 치료에 효과가 있다고 검증된 가상현실 기술을 이용한 새롭고 효과적인 금연클리닉이 제안되었으며 본 연구는 가상현실 금연클리닉에 필요한 시나리오와 정보구조를 개발하는 것에 중점을 두었다. 치료효과를 높이기 위해 가상현실 상에서 피험자의 몰입감을 증대시킬 수 있는 방안들을 찾아내는 것이 주요 논점이며 이를 위해 개인의 기호에 맞는 흡연자극 시나리오를 단계별로 구현하고 각 단계 별 필요한 자극의 요소는 무엇인 지를 규명하는 과정이 필요하였다. 주로 심리학적인 근거와 흡연자들을 대상으로한 설문내용 및 임상자료들을 기초로 하여 연구과정을 전개하였다. 연구결과는 시작품으로 제작되어 실제 금연클리닉에 적용되었으며 초기 임상단계에서 가상현실 치료효과가 약물치료의 효과에 준할 수 있다는 결과를 얻었다. Recently Schools and local governments much more companies have been enforcing very strong and strict regulations on smoking. which have never been implemented in the past and it will be expanded adjusting with the government's policy. Since addictive smoking behavior has been recognized as a disease so various treatments for smokers have been introduced. However time and place constraint and since there are many limits in operating social demand is unable to prepare. For this situation to be improved a novel and effective smoking cessation clinic using virtual environments which is proven to be an effective treatment for diverse addictions have been proposed. In this research I focused on the scenarios for virtual reality smoking cessation clinic and development of the information architecture. The main point was to find the ways to increase immersions in virtual environment in order to improve the curative effect. For this I developed the scenarios according to personal preference step by step and also I had to undergo the process of identifying what kinds of stimulating-elements were in each step. The research process was mainly developed on the basis of psychological grounds surveys targeting smokers and clinical materials. The first prototype was made as a result of this research and applied to the actual smoking cessation clinic. And there were the result of virtual reality smoking cessation clinic can be equivalent to the effects of drug therapy in early clinical stage.

        포항시 통합 모바일 서비스 개발 과정 연구

        본 연구는 한 중소도시의 통합 모바일 서비스를 개발하는 과정을 모바일 서비스 구성과 모바일 UX 디자인 개발로 진행한 내용에 대한 것이다. 정보화시대를 지나면서 특별시 및 광역시 포함 전국 80 여개 크고 작은 시청 및 산하기관이 보유하고 있는 웹사이트의 수는 기하급수적으로 증가하였다. 각 시청 홈페이지를 방문하면 수많은 서비스들의 나열과 유관기관으로 연결되는 엄청난 수의 링크들은 방문자들에게 편리한 서비스를 제공한다는 취지를 무색하게 만들고 있는 상황을 자주 접하게 된다. 웹 서비스 대비 모바일 서비스의 장점이 부각되면서 각 지자체의 웹 기반 서비스들도 모바일 환경으로 속속 옮겨가고 있다. 이러한 상황에서 그간 만들어졌던 웹 기반 서비스들을 통합하고 시민들에게 최적의 모바일 서비스를 제공하기 위해서는 과학적인 분류 방법과 과정이 있어야 가능하다. 이를 위해 본 연구에서는 통합 모바일 서비스 사용자경험의 목표를 즉각성, 간결함, 그리고 사용자환경으로 정하고 이에 맞는 서비스 도출과 사용자경험을 개발하고자 하였다. 첫째, 최적의 모바일 서비스를 구성하기 위해 먼저 주 사용자층을 포항시민과 방문자로 나누었다. 가로축에 사용자층을 근거한 모바일 서비스 그룹을 배치하고 세로축에 기존 웹 서비스를 배치하여 교차점에서 필요한 서비스를 추출하였다. 둘째, 선정된 서비스를 모바일 환경에 최적화하기 위해 국내 주요 지자체 모바일 서비스를 평가하고 정부 가이드라인을 분석하여 UX 디자인 가이드라인을 제안하였다. 이를 통해 산발적으로 개발되었던 수많은 웹사이트 서비스들을 우선순위에 따라 함축시킬 수 있었고, 포항시의 차별화된 모바일핀테크 기술과 유관 기관의 회원증을 더한 최적의 통합 모바일 서비스를 개발할 수 있었다. This Study is about the process of developing integrated mobile service for Pohang City by selecting proper services and developing the user experience design. Over the information age, the number of websites held by 80 major and small government offices nationwide, including special cities and metropolitan cities, increased exponentially. Too many services and links make us extremely confused and give us stress when we visit each city’s homepages. As the advantages of mobile services over Web services are highlighted, the web-based services of each local government are also shifting to the mobile environment. In this situation, it is possible to integrate the numerous web-based services that have been created and provide optimal mobile services to citizens with scientific classification methods and processes. To this reason, the purpose of this study was to develop mobile service and mobile user experience in accordance with the goals of the integrated mobile service user experience, including its immediate, concise and user experience. First, in order to organize the proper mobile services, the main user group was divided into Pohang citizens and visitors. Users-oriented mobile service groups were placed on the horizontal axis and existing Web services were placed on the vertical axis to extract the required services at their intersections. Second, In order to optimize selected services for mobile environments, the UX design guidelines were presented by evaluating mobile services of Major cities in Korea and analyzing government UX guidelines. This enabled the development of an optimal integrated mobile service with differentiated mobile fintech technology in Pohang City and membership of each agency.

      • A microfluidic device for evaluating the dynamics of the metabolism-dependent antioxidant activity of nutrients.

        <P>Various food components are known for their health-promoting effects. However, their biochemical effects are generally evaluated in vitro, and their actual in vivo effect can vary significantly, depending on their metabolic profiles. To evaluate the effect of the liver metabolism on the antioxidant activity, we have developed a two-compartment microfluidic system that integrates the dynamics of liver metabolism and the subsequent antioxidant activity of food components. In the first compartment of the device, human liver enzyme fractions were immobilized inside a poly(ethylene glycol) diacrylate (PEGDA) hydrogel to mimic the liver metabolism. The radical scavenging activity was evaluated by the change of the 2,2-diphenyl-1-picrylhydrazyl (DPPH) absorbance in the second compartment. Reaction engineering and fluid mechanics principles were used to develop a simplified analytical model and a more complex finite element model, which were used to design the chip and determine the optimal flow conditions. For real-time measurements of the reaction on a chip, we developed a custom-made photospectrometer system with an LED light source. The developed microfluidic system showed a linear and dose-dependent antioxidant activity in response to increasing concentration of flavonoid. We also compared the antioxidant activity of flavonoid after various liver metabolic reactions. This microfluidic system can serve as a novel in vitro platform for predicting the antioxidant activity of various food components in a more physiologically realistic manner, as well as for studying the mechanism of action of such food components.</P>

        뽕잎과 꾸지뽕잎의 활성 성분 함량 및 지방세포분화 억제 효과 비교

        본 연구에서는 뽕잎과 꾸지뽕잎의 성분함량을 비교하기 위해 뽕잎과 꾸지뽕잎을 추출하고 이들의 성분으로 알려진 15개 성분을 HPLC를 이용하여 분석한 결과, protocatechuic acid, catechin, chlorogenic acid, caffeic acid, epicatechin, umbelliferone, kaempferol 3-β-D-glucopyranoside, resveratrol, oxyresveratrol, isoquercitrin 성분은 뽕잎에서 함량이 높게 나타났으며, p-coumaric acid, quercitrin, quercetin, kaemperol, cryptochlorogenic acid 성분은 꾸지뽕에서 함량이 높게 나타났다. 또한 뽕잎과 꾸지뽕잎 추출물의 지방세포분화 억제 효과를 측정한 결과, 두 가지 추출물 모두 유의적인 효과를 나타내었으나 뽕잎의 지방세포분화 억제 효과가 상대적으로 높았다. 이는 뽕잎에서 높은 함량을 보인 isoquercitrin, resveratol, protocatechuic acid, chlorogenic acid 성분이 기여하는 것으로 판단되며, 항비만 활성 부분에서는 뽕잎의 활용성이 높을 것으로 기대된다. Morus alba L. and Cudrania tricuspidata leaves have been reported to show anti-oxidative, anti-diabetes, and anti-obesity effects. However, no study to date has carried out a comparison of the contents of these leaves and their anti-adipogenesis effects. In this study, 15 compounds (caffeic acid, p-coumaric acid, chlorogenic acid, cryptochlorogenic acid, protocatechuic acid, catechin, epicatechin, kaempferol, kaempferol 3-D-glucopyranoside, quercetin, quercitrin, isoquercitrin, resveratrol, oxyresveratrol, and umbelliferone) contained in the extracts of Morus alba L. and Cudrania tricuspidata leaves were analyzed, and their inhibitory effects on adipogenesis were examined using 3T3-L1 preadipocytes. The results showed that the contents of 10 compounds were higher in Morus alba L. leaves extracts compared to Cudrania tricuspidata with the exceptions being p-coumaric acid, quercitrin, quercetin, kaemperol, and cryptochlorogenic acid. Also, Morus alba L. leaves extract had a higher inhibitory activity on adipocyte differentiation compared to that of Cudrania tricuspidata leaves extract. These results suggest that Morus alba L. leaves would be useful as an anti-adipogenesis material.
