단보 : Chinese Hamster Lung Cell의 소핵시험을 이용한 식물추출물 유기농업자재의 유전독성평가

님추출물과 고삼추출물은 식물추출물 유기농업자재로 우리나라에 등록되어 친환경농산물 재배에 널리 사용되고 있다. 본 연구에서는 님추출물 2종과 고삼추출물 2종에 대한 안전성을 확인하고자 Chinese hamster lung cells을 이용한 in vitro 소핵시험(vitMN)을 통해 유전독성을 평가하였다. 시험은 cytochalasin B 적용군과 비적용군으로 구분하여 시험하였으며, 양성대조물질로 mitomycin C와 colchicine을 사용하였다. 시험 결과, 님추출물 시료 2종과 고삼추출물 시료 2종은 처리 농도에 따라 소핵발생 비율이 증가하였으나 모든 시료 처리군에서 전체 발생률이 2.2% 미만으로, 음성대조군대비 2배 미만의 소핵이 관찰되었다. 따라서 님추출물 및 고삼추출물 시료는 본 시험의 vitMN 결과 최종적으로 음성으로 확인되었다. 또한 본 연구와 동일한 시료를 이용한 선행연구에서 복귀돌연변이 시험에서 음성을 보인 결과를 근거로 battery system에 의거해 종합적으로 평가하였을 때 님추출물과 고삼추출물 시료는 유전독성을 일으키지 않는 것으로 최종 판정하였다. BACKGROUND: Azadirachta Indica extract(AIE) and Sophorae radix extract(SRE) are widely used as environment-friendly organic materials of plant origin in South Korea. METHODS AND RESULTS: In this study, the in vitro micronucleus(vitMN) tests of two samples of AIE and SRE were conducted to evaluate their genotoxicity using the Chinese hamster lung(CHL) cell. This study was composed of two parts; cytochalasin B(cyto B) test and non-cyto B test. Mitomycin C and colchicine were used as positive controls. As a result, the incidence of micronucleus(MN) in all AIE and SRE treated groups increased in dose- dependent manner, but were less than 2.2% in 1,000 binucleated cells. In addition, there were no significant increases of MN incidence in all AIE and SRE treated groups, compared with the negative control group. CONCLUSION: Therefore, we suggest that AIE samples and SRE samples used in this study may have no genotoxicity in the in vitro micronucleus test using the CHL cells. In our previous study, we reported that AIE and SRE did not cause genotoxicity in Ames test. According to the genotoxicity battery system, we concluded that AIE and SRE used in this study have no genotoxic effects to humans.

시화호내ㆍ외해수와 유입되는 하천수 중 PFCs의 농도분포를 2005년 11월에 채취한 시료를 이용하여 분석하였다. 시화호의 표층수, 저층수 및 유입하천수 중의 농도는 perfluorohexanesulfonate (PFHS)가 ND~0.8, 0.3~0.5, 1.8~35.9 ng/L, perfluorooctanesulfonate (PFOS)는3 3.6~11.6, 3.6~8.1, 5.8~253.2 ng/L, perfluorooctanesulfonamide (PFOSA)는 ND~0.5, ND~0.4, 0.3~2.7 ng/L, perfluorononanoic acid (PFNA)는 ND~3.5, ND~0.6, ND-125.0 ng/L, pefluorooctanoic acid (PFOA)는 ND~3.3, ND~0.7, 0.7~405.5 ng/L의 범위로 각각 검출되었다. 전반적으로 PFOS와 PFOA가 전체 PFCs에서 상대적으로 높았으며, 지역별로는 시화호로 유입되는 하천수 중 산업단지부근에 위치한 L1~L5 지점에서 다른 조사지점의 농도에 비해 상대적으로 매우 높은 농도를 보였다. Water samples collected from the surface and bottom layer of Sihwa lake, and the inflow stream water around Sihwa lake were analyzed to determine the concentration of PFCs on November, 2005. Ranges of concentrations of perfluorohexanesulfonate (PFHS) in seawater of the surface and bottom layer of Sihwa lake and the inflow stream water around Sihwa lake were ND-0.8, 0.3~0.5 and 1.8~35.9 ng/L, respectively. Those of perfluorooctanesulfonate (PFOS) were 3.6~11.6, 3.6~8.1, 5.8~253.2 ng/L, perfluorooctanesulfonamide (PFOSA) were ND~0.5, ND~0.4, 0.3~2.7 ng/L, perfluorononanoic acid (PFNA) were ND~3.5, ND~0.6, ND~125.0 ng/L, perfluorooctanoic acid (PFOA) were ND~3.3, ND~0.7, 0.7~405.5 ng/L, respectively. Dominant compounds of PFCs were PFOS and PFOA. Concentrations in site(L1~L4) of inflow stream located on the industrial complex were higher than the values of seawater and the others stream.
다발성 외상 환자에서 Emergency Room Transfusion Score (ETS)를 이용한 응급수혈의 예측인자

Purpose: The purposes of this study are finding the elements for a fast determination of the need for a transfusion to a multiple trauma patient arriving at this clinic in the initial stage establishing objective bases for a doctor in an emergency department to determine the need for a transfusion immediately after a patient has arrived at the emergency department, and providing treatment by considering various factors based on the nine criteria suggested in the emergency room transfusion score (ETS). Methods: This study was conducted on 375 multiple-trauma patients who visited the Chosun University Hospital Emergency Medical Center and activated the Emergency Trauma Team from January 2010 to December 2013. The patients were divided into the transfused group and the non-transfused group by retrospectively analyzing their medical records. Subsequently, the medical records were examined using the nine items suggested by the ETS and the results were analyzed. Results: Three hundred seventy-five patients with multiple traumas visited the Chosun University Hospital Emergency Medical Center and activated the Emergency Trauma Team. Among them, 258 died and 117 recovered and left the hospital. The deceased patients consisted of 182 males and 76 females with an average age 45. Of the 375, 165 were transferred from other hospitals, and 245 were blunt trauma patients. One hundred sixty-nine patients were injured in traffic accidents, and 119 of those 169 who had systolic blood pressure less than 90 mm Hg died. Two hundred twenty-six (60.3%) out of the 375 patients with multiple traumas receivedan emergency blood transfusion and their average age was 48. The 375 patients consisted of 156 males, 151 who had been transferred from other hospitals, 218 who presented with blunt trauma, 134 who had been injured in traffic accidents, 156 who had a systolic blood pressure less than 90 mm Hg, 134 who scored higher than 9 points on the GCS, and 162 who had a stable pelvic fracture of these 143 died. Conclusion: During this study, 226 (60.3%) out of the patients with multiple traumas received an emergency blood transfusion. After analyzing the results related to emergency blood transfusion by using ETS, we found that an emergency blood transfusion had to be prepared quickly when patients were transferred from other hospitals when the systolic blood pressure was less than 90 mmHg. when abnormalities had been detected by ultrasonography and when the patient presented with a stable pelvic fracture. 광양만 유역의 수질 및 표층퇴적물에서의 PFOS 및 PFCs의 오염에 관한 연구

Per Fluorinated Compounds(PFCs) are fully fluorinated organic molecules produced synthetically in an electrochemical fluorination process PFOS, PFOA, PFHS, PFOSA are including among PFCs these compounds have high molecular weight and created are environmentally stable these compounds used in a variety of applications, such as in lubricants, paints, cosmetics, and fire-fighting foams. Produced and used widely over the last half-century, PFCs are being detected in wildlife and humans around the globe. Kwangyang Bay is a semi-closed coastal sea on the south coast of Korea. The total population within the bay watershed is approximately 240,000 people. Several rivers and ditches, including the Seomjin River, one of Korea's five biggest river systems, enter the bay, and seawater is exchanged with the South Sea at the southern part of the bay. The water depth is less than 5m in the western part of the bay, and is more than 20m near Myodo and in the eastern part of the bay. The area is characterised by a semidiurnal tide. Land-based pollution sources(river water, sediment) were investigated in January 2005 and seawater and sediment samples were collected in November 2004 and January 2005. Ranges of concentrations of PFOS, PFOA, PFHS and PFOSA in stream waters were ND-1411.5ng/L, ND-376.0ng/L, ND-488.0ng/L and ND-3.6ng/L, respectively. Stream sediments were ND-5.43ng/g dry-wt, ND-0.47ng/g dry-wt, ND-0.14ng/g dry-wt and ND-0.47ng/g dry-wt, respectively. Ranges of concentrations of PFOA in sea water at November 2004 were ND-5.0ng/L.. PFOS and PFOA in sea water at January 2005 were ND-14.3ng/L and ND-4.9ng/L, respectively.
