This study was carried out to explore the effect of DNA hypomethylation on chondrocytes phenotype, in particular the effect on chondrocyte hypertrophy, maturation, and apoptosis. Chondrocytes derived from caudal region of day 17 embryonic chick sterna were pretreated with hypomethylating drug 5-aza-2'-deoxycytidine for 48 hours and then maintained in the normal culture medium for up to 14 days. Histological studies showed distinct morphological changes occurred in the pretreated cultures when compared to the control cultures. The pretreated chondrocytes after 7 days in culture became bigger in size and acquired more flattened fibroblastic phenotype as well as a loss of cartilage specific extracellular matrix. Scanning electron microscopy at day 7 showed chondrocytes to have increased in cell volume and at day 14 in culture the extracellular matrix of the pretreated cultures showed regular fibrillar structure heavily embedded with matrix vesicles, which is the characteristic feature of chondrocyte hypertrophy. Transmission electron microscopic studies indicated the terminal fate of the hypertrophic cells in culture. The pretreated chondrocytes grown for 14 days in culture showed two types of cells: dark cells which had condense chromatin in dark patches and dark cytoplasm. The other light chondrocytes appeared to be heavily loaded with endoplasmic reticulum indicative of very active protein and secretory activity; their cytoplasm had large vacuoles and disintegrating cytoplasm. The biosynthetic profile showed that the pretreated cultures were actively synthesizing and secreting type X collagen and alkaline phosphatase as a major biosynthetic product.

Web technology is evolving with the passage of time, from a single node server to high availability and then in the form of Kubernetes. In recent years, the research community have been trying to provide high availability in the form of multi master cluster with a solid election algorithm. This is helpful in increasing the resources in the form of pods inside the worker node. There are new impact of known DDoS attack, which is utilizing the resources at its peak, known as Yoyo attack. It is kind of burst attack that can utilize CPU and memory to its limit and provide legit visitors with a bad experience. In this research, we tried to mitigate the Yoyo attack by introducing a firewall at load-balancer level to prevent the attack from going to the cluster network.
Kashmir: Self-Determination versus State Terrorism

The unprecedented and condemnable terrorist strikes of September 11 have united the world community to counter this menace. In combating terrorism, the causes of terrorism must also be addressed. The international community has already found the political will, expressed through the UN Security Council, to condemn and actively intervene in grave breaches of human rights, in Rwanda, Bosnia and Kosovo. International tribunals were set up. No state and no state official can now plead sovereign immunity for grave human rights violations within any country, let alone within an occupied territory. Therefore the world community can no longer close its eyes to grave violations, however important and powerful the offending state. The campaign against terrorism must not be used to perpetuate oppression and human rights abuses against oppressed and occupied peoples who retain the right to armed struggle to further their right of self-determination in the face of such oppression. Occupied Kashmir, where 13 million Kashmiris have waited for 53 years to gain their UN Security Council-sanctioned right of self-determination, and have lost 80,000 precious lives since 1990, and where self determination is pitted against state terrorism, constitutes the most important global test case for the sincerity of the world community to actively and effectively intervene to end this humanitarian crisis, which constitutes a threat to international peace and security, and to bring about a dialogue between India and Pakistan leading to a peaceful resolution of this dispute on the basis of the aspirations of the Kashmiri people and the United Nations resolutions.

Enhanced recovery after hepatopancreaticobiliary surgery: A single-center case control study

Backgrounds/Aims: The aim of this study was to find the safety and effectiveness of enhanced recovery after surgery (ERAS) in patients who undergo hepatopancreaticobiliary (HPB) surgeries and its association with the postoperative complications and survival rate of the patients. Methods: This study was conducted on patients who underwent HPB surgeries in Imam Khomeini Hospital Complex, Iran from 2018 to 2020. Patients who underwent surgery after from 2019 to February 2020 considered as the ERAS group (n=47) in which ERAS was implemented postoperatively including removing nasogastric tube and initiating surgical diet at 6 hours post operation, and removing intraabdominal drains and Foley catheter at postoperative day one. Other patients (n=43) were considered as the control group in which conventional postoperative care was implemented. Results: Ninety patients with the mean age of 47.3±13.3 yrs/old (range=17-76) including 39 females were enrolled into the study. There were no significant differences between the demographic and preoperative comorbidities between the two groups. Pain severity of the patients in the ERAS group was significantly lower than the control group (visual analogue scales of 3.4±0.77 vs. 4.47±0.88, p<0.001). However, there were no significant differences between the other postoperative data between the two groups. One patient in each group died during hospitalization period due to myocardial infarction. Conclusions: ERAS may be safe and effective in patients who undergo HPB surgery and may be associated with less severe postoperative pain.
High-intensity Fitness Training Among a National Sample of Male Career Firefighters

Obesity and fitness have been identified as key health concerns among USA firefighters yet little is known about the current habits related to exercise and diet. In particular, high-intensity training (HIT) has gained increasing popularity among this population but limited quantitative data are available about how often it is used and the relationship between HIT and other outcomes. Using survey methodology, the current study evaluated self-reported HIT and diet practice among 625 male firefighters. Almost one-third (32.3%) of participants reported engaging in HIT. Body composition, as measured by waist circumference and percentage body fat, was significantly related to HIT training, with HIT participants being approximately half as likely to be classified as obese using body fat [odds ratio (OR) = 0.52, 95% confidence interval (CI) = 0.34-0.78] or waist circumference (OR = 0.61, 95% CI = 0.37-0.98). Those who engaged in HIT were more than twice as likely as those who did not (OR = 2.24, 95% CI = 1.42-3.55) to meet fitness recommendations. Findings highlight directions for future prevention and intervention efforts.

In this research, a novel method based on Canonical Correlation Analysis (CCA) and Artificial Neural Network (ANN) to detect epileptic seizures from EEG signals is proposed. CCA was applied on EEG signals and feature vectors corresponding to Eigen values were extracted. These Eigen values were fed as input to Artificial Neural Network (ANN)’s widely explored model Multilayer Perceptron Neural Networks (MLPNNs) for classification between occurrence of non-epileptic seizures and epileptic seizures. The extracted Eigen values using CCA proved to be a better epileptic seizures detector and provide average classification accuracy, sensitivity and specificity as 92.583%, 93.25% and 91% respectively.

Teaching and learning are one of the major issues during this pandemic (COVID-19). Since the pandemic started, there are many changes in teaching and learning styles as everything related to studies started online. Game-Based Learning has got remarkable importance in the educational system and pedagogy as an effective way of increasing student inspiration and engagement. In this field, most of the work has been carried out in digital games. This research uses an Animated Game-Based Learning design in enhancing student engagement and perception of learning. In teaching Computer Science (CS) concepts in higher education, to enhance the pedagogy activities in CS concepts, more specifically the concepts of "Data Structures (DS)" i.e., Array, Stack, and Queue concepts are focused. This study aims to observe the difference in students' learning with the use of different learning methods i.e., the traditional learning (TL) method and the Animated Game-Based Learning (AGBL) Method. The experimental results show that learning DS concepts has been improved by the AGBL method as compared to the TL method.
