        D. candidum has in vitro anticancer effects in HCT-116 cancer cells and exerts in vivo anti-metastatic effects in mice

        BACKGROUND/OBJECTIVES: D. candidum is a traditional Chinese food or medicine widely used in Asia. There has been little research into the anticancer effects of D. candidum, particularly the effects in colon cancer cells. The aim of this study was to investigate the anticancer effects of D. candidum in vitro and in vivo. MATERIALS/METHODS: The in vitro anti-cancer effects on HCT-116 colon cancer cells and in vivo anti-metastatic effects of DCME(Dendrobium canidum methanolic extract) were examined using the experimental methods of MTT assay, DAPI staining, flow cytometry analysis, RT-PCR, and Western blot analysis. RESULTS: At a concentration of 1.0 mg/mL, DCME inhibited the growth of HCT-116 cells by 84%, which was higher than at concentrations of 0.5 and 0.25 mg/mL. Chromatin condensation and formation of apoptotic bodies were observed in cancer cells cultured with DCME as well. In addition, DCME induced significant apoptosis in cancer cells by upregulation of Bax, caspase 9, and caspase 3, and downregulation of Bcl-2. Expression of genes commonly associated with inflammation, NF-κB, iNOS, and COX-2, was significantly downregulated by DCME. DCME also exerted an anti-metastasis effect on cancer cells as demonstrated by decreased expression of MMP genes and increased expression of TIMPs, which was confirmed by the inhibition of induced tumor metastasis in colon 26-M3.1 cells in BALB/c mice. CONCLUSIONS: Our results demonstrated that D. candidum had a potent in vitro anti-cancer effect, induced apoptosis, exhibited anti-inflammatory activities, and exerted in vivo anti-metastatic effects.

        Positive enhancement of Lactobacillus fermentum HY01 on intestinal movements of mice having constipation

        Lactobacilli have been used to treat many gastrointestinal disorders. But the outcome of Lactobacilli are strain specific. The strain Lactobacillus Fermentum, HY01, (LF-HY01) has a good performance in the environment of gastrointestinal tract. In this study, the aim is to investigate the preventive effects of LF-HY01 against activated-carbon-induced constipation in mice. Mice are randomized into four groups. Normal group was fed a normal diet, model group also has the same with activated carbon treatment, and low and high concentration groups are treated with LF-HY01. We have determined many indexes such as body weight, water content in faeces, defecation conditions, the level of small intestinal villi damages and levels of various neurotransmitters in serum, including motilin (MTL), gastrin (GT), endothelin (ET), somatostatin (SST), acetylcholinesterase (AchE), substance P (SP), and vasoactive intestinal peptide (VIP). LF-HY01 has no significant difference in each group, but it can significantly improve water content of faeces, defecation time of first black stool and activated carbon propelling rate in small intestine as compared of model group. Furthermore, LF-HY01 can effectively prevent small intestinal villi damages, which is less than that of model group. Moreover, LF-HY01 has the consistency to increase the levels of MTL, GT, ET, AchE, SP and VIP, and LF-HY01 can also have the ability to reduce the level of SST. These results suggest that Lactobacillus Fermentum, HY01, has a great impact in enhancing intestinal peristalsis ability and has the ability to prevent from activated-carbon-induced constipation in mice.

        Effects of Three Types of Resistant Starch on Intestine and Their Gastric Ulcer Preventive Activities in vivo

        To examine the effects of three types of resistant starch (RS) on intestinal metabolites, structure and their gastric injury preventive activities, Sprague-Dawley mice were fed diet containing 15% RS for 4 weeks. Response relationships among three types of RS (RS2, RS3, and RS4) food intake, body weight gain, food efficiency, wet weight of cecum (with or not with contents), pH, ammonia production, and short-chain fatty acids (SCFAs) concentration of cecal contents as well as intestinal structure were investigated. All three types of RS had effects on food intake, weight control, lowering of pH, ammonia production in cecal contents, increasing the wet weight of cecum (with or without contents), changes in SCFAs concentration of cecal contents, and physiological structure of small intestine and cecum compared to the control group (common starch-fed rats). RS3group showed minimum weight gain and food efficiency,compared to both the control group and the other two types of RS diet. RS3 group showed more significant lowering of pH,ammonia production, and SCFAs of cecal contents. In addition, all three types of RS shortened villous height, and muscle and mucosal thickening in small intestine, resulting in significantly decreased villous height and mucosal thickness, whereas significantly increased muscle thickness in the cecum. However,they did not cause detectable pathological changes in the small intestine and cecum. Animal model was also used to check its gastric ulcer preventive effects. The gastric ulcer level was significantly down-regulated by RS3, demonstrating its antiinflammatory properties. RS3 reduced the levels of serum proinflammatory cytokines of IL-6, IL-12, TNF-α, and IFN-γcompared to RS2 and RS4. RS3 increased the somatostatin (SS)and vasoactive intestinal peptide (VIP), decreased the motillin (MOT) and substance P (SP) serum levels. These results suggest that RS3 shows the best gastric ulcer preventive effect.
