        Gum arabic-encapsulated gold nanoparticles modulate hypoxamiRs expression in tongue squamous cell carcinoma

        Background Oral tongue squamous cell carcinoma (OTSCC) is a popular aggressive malignancy of the oral cavity. Despite advances in OTSCC therapy, the overall 5-year survival rate is low. The tumor microenvironment resistance factors lead to chemotherapy failure, especially intratumoral hypoxia. HIF-1α, the main protein in hypoxia pathway, influences cell survival and angiogenesis. Hypoxia/HIF-1α system is a potential strategic target in cancer therapeutics. The expression of hypoxia-regulating miRNAs (hypoxamiRs; miR-210 and miR-21), regulators of HIF-1α, is high in OTSCC. Gum Arabic-encapsulated gold nanoparticles (GA-AuNPs) have been reported as promising modality in cancer treatment. Objective This study aimed to investigate the influence of GA-AuNPs on the hypoxia regulators in OTSCC (CAL-127 cells). GA-AuNPs cytotoxicity assessed by MTT assay; cell death mode was detected by dual DNA staining; monitoring of cellular hypoxia was followed by pimonidazole; miR-210 and miR-21 expression was assessed by qPCR; and their targets (HIF-1α and c-Myc) assayed by immunocytofluorescence and ELISA, respectively. Results GA-AuNPs (75–80 nm; λmax of ~ 540 nm) reduced cell viability with IC50 of 392.3 and 247.3 µg/ml after 24 h and 48 h, respectively. Cell death was mainly due to apoptosis. CAL-27 cells exhibited high hypoxia and the treatment with GA-AuNPs inhibited this hypoxia in a dose-dependent manner, as detected by pimonidazole. GA-AuNPs (30% IC50) significantly reduced miR-210 and miR-21 expression. HIF-1α and c-Myc were inhibited by GA-AuNPs (30% IC50, for 48 h). Conclusion The study findings may suggest GA-AuNPs as a promising carrier for chemotherapies to diminish intratumoral hypoxia-stimulated resistance.       • The Emergence of the Conserved AAA+ ATPases Pontin and Reptin on the Signaling Landscape

        <P>Pontin (also known as RUVBL1 and RVB1) and Reptin (also called RUVBL2 and RVB2) are related members of the large AAA+ (adenosine triphosphatase associated with diverse cellular activities) superfamily of conserved proteins. Various cellular functions depend on Pontin and Reptin, mostly because of their functions in the assembly of protein complexes that play a role in the regulation of cellular energetic metabolism, transcription, chromatin remodeling, and the DNA damage response. Little is known, though, about the interconnections between these multiple functions, how the relevant signaling pathways are regulated, whether the interconnections are affected in human disease, and whether components of these pathways are suitable targets for therapeutic intervention. The First International Workshop on Pontin (RUVBL1) and Reptin (RUVBL2), held between 16 and 19 October 2012, discussed the nature of the oligomeric organization of these proteins, their structures, their roles as partners in various protein complexes, and their involvement in cellular regulation, signaling, and pathophysiology, as well as their potential for therapeutic targeting. A major outcome of the meeting was a general consensus that most functions of Pontin and Reptin are related to their roles as chaperones or adaptor proteins that are important for the assembly and function of large signaling protein complexes.</P>

      • Pectic-Oligoshaccharides from Apples Induce Apoptosis and Cell Cycle Arrest in MDA-MB-231 Cells, a Model of Human Breast Cancer

        Background: The effects of plant products on cancer cells has become a field of major importance. Many substancesmay induce apoptosis in anti-cancer treatment. Pectins, a family of complex polysaccharides, and their degradation products may for exasmple exert apoptotic effects in cancer cells. Apples and citrus fruits are the main sources of pectin which can be applied for anti-cancer research. The present study concerned an intact form of pectic-oligoshaccharide named pectic acid (poly galactronic acid). Materials and Methods: Inhibition of cell proliferation assays (MTT), light microscopy, fluorescence microscopy (acridin orange/ethidium bromide), DNA fragmentation tests, cell cycle analysis, annexin PI and Western blotting methods were applied to evaluate apoptosis. Results: The results indicated that pectic acid inhibited cell growth and reduced cell attachment after 24h incubation. This did not appear to be due to necrosis, since morphological features of apoptosis were detected with AO/EB staining and cell cycling was blocked in the sub-G1 phase. Annexin/PI and DNA fragmentation findings indicated that apoptosis frequency increased after 24h incubation with pectic acid. In addition, the data showed pectic acid induced caspase-dependent apoptosis. Conclusions: These data indicate that apple pectic acid without any modification could trigger apoptosis in MDA-MB-231 human breast cancer cells and has potential to improve cancer treatment as a natural product.

        한.중.일 중학생의 정신건강에 영향을 미치는 요인

        Objectives: This study analyzed the factors that influence the mental health of adolescents in Korea, China, and Japan. Methods: The survey was conducted in each country between November 2008 and January 2009. The respondents were 1,390 Korean, Chinese, and Japanese students aged between 14 years and 16. The mental health was measured by the School Mental Health Scale of Ochanomizu University (2004), which is composed of six scales: physical symptoms, eating disorders, depression, interpersonal relationship, powerlessness, and impulsiveness. Results: The relationship with family and friends are strongly associated with mental health. As for Korean and Chinese female students experienced mental illness more than male. Regarding Korean students, the time to use TV computer game is related to relationship and impulsiveness. The mobile phone use more than three hours is related to depression and relationship for Japanese, and the number of family member less than three, is associated with powerlessness for Chinese. Conclusion: Regular and effective health education is required in order to improve students lifestyles and family and peer relationships. This study also offers the fundamental information for health promotion programs for Korean, Chinese and Japanese students.
