      • Inhibition of Breast Cancer Metastasis Via PITPNM3 by Pachymic Acid

        Breast cancer metastasis is the most common cause of cancer-related death in women. Thus, seeking targets of breast tumor cells is an attractive goal towards improving clinical treatment. The present study showed that CCL18 from tumor-associated macrophages could promote breast cancer metastasis via PITPNM3. In addition, we found that pachymic acid (PA) could dose-dependently inhibit migration and invasion of MDA-MB-231cells, with or without rCCL18 stimulation. Furthermore, evidence was obtained that PA could suppress the phosphorylation of PITPNM3 and the combination of CCL18 and PITPNM3. Therefore, we speculate that PA could inhibit breast cancer metastasis via PITPNM3.

        The Process Planning in Laser Thermal Adjustment of Three Cut-Outs Actuators

        Laser thermal adjustment as an application of laser forming in micro systems attracts much attention. Previous work on laser thermal adjustment of the two-bridge actuator shows that the deformations are always limited. In this paper, an actuator with three cut-outs is designed to enlarge the deformation range. A simple deformation formula is also established for such actuator based on the deformation behavior of two-bridge actuators. Four cases of the location relation between the target position and the calculated set point are considered in planning the process parameters to reach the designed target position. The planning process is validated by the numerical simulation and experiments.

        mTOR- and SGK-Mediated Connexin 43 Expression Participates in Lipopolysaccharide-Stimulated Macrophage Migration through the iNOS/Src/FAK Axis

        <P>Connexin 43 (Cx43) deficiency was found to increase mortality in a mouse model of bacterial peritonitis, and Cx43 is upregulated in macrophages by LPS treatment. In this study, we characterized a novel signaling pathway for LPS-induced Cx43 expression in RAW264.7 cells and thioglycolate-elicited peritoneal macrophages (TGEMs). LPS alone or LPS-containing conditioned medium (CM) upregulated Cx43. Overexpression or silencing of Cx43 led to the enhancement or inhibition, respectively, of CM-induced TGEM migration. This response involved the inducible NO synthase (iNOS)/focal adhesion kinase (FAK)/Src pathways. Moreover, CM-induced migration was compromised in TGEMs from Cx43<SUP>+/−</SUP> mice compared with TGEMs from Cx43<SUP>+/+</SUP> littermates. Cx43 was upregulated by a serum/glucocorticoid-regulated kinase 1 (SGK) activator and downregulated, along with inhibition of CM-induced TGEM migration, by knockdown of the <I>SGK</I> gene or blockade of the SGK pathway. LPS-induced SGK activation was abrogated by Torin2, whereas LPS-induced Cx43 was downregulated by both Torin2 and rapamycin. Analysis of the effects of FK506 and methylprednisolone, common immunosuppressive agents following organ transplantation, suggested a link between these immunosuppressive drugs and impaired macrophage migration via the Cx43/iNOS/Src/FAK pathway. In a model of <I>Escherichia coli</I> infectious peritonitis, GSK650349-, an SGK inhibitor, or Torin2-treated mice showed less accumulation of F4/80<SUP>+</SUP>CD11b<SUP>+</SUP> macrophages in the peritoneal cavity, with a delay in the elimination of bacteria. Furthermore, following pretreatment with Gap19, a selective Cx43 hemichannel blocker, the survival of model mice was significantly reduced. Taken together, our study suggested that Cx43 in macrophages was associated with macrophage migration, an important immune process in host defense to infection.</P>

        On the direct strength and effective yield strength method design of medium and high strength steel welded square section columns with slender plate elements

        The ultimate carrying capacity of axially loaded welded square box section members made of medium and high strength steels (nominal yield stresses varying from 345 MPa to 460 MPa), with large width-to-thickness ratios ranging from 35 to 70, is analyzed by finite element method (FEM). At the same time, the numerical results are compared with the predicted results using Direct Strength Method (DSM), modified DSM and Effective Yield Strength Method (EYSM). It shows that curve a, rather than curve b recommended in Code for design of steel structures GB50017-2003, should be used to check the local-overall interaction buckling strength of welded square section columns fabricated from medium and high strength steels when using DSM, modified DSM and EYSM. Despite all this, EYSM is conservative. Compared to EYSM and modified DSM, DSM provides a better prediction of the ultimate capacities of welded square box compression members with large width-thickness ratios over a wide range of width-thickness ratios, slenderness ratios and steel grades. However, for high strength steels (nominal yield strength greater than 460 MPa), the numerical and existent experimental results indicate that DSM overestimates the load-carrying capacities of the columns with width-thickness ratio smaller than 45 and slenderness ratio less than 80. Further, for the purpose of making it suitable for a wider scope, DSM has been modified (called proposed modified DSM). The proposed modified DSM is in excellent agreement with the numerical and existing experimental results.

        Analysis on Robust Passivity of Uncertain Neural Networks with Time-Varying Delays via Free-Matrix-Based Integral Inequality

        This paper investigates the robust delay-dependent passivity problem of neural networks (NNs) with time-varying delays and parameter uncertainties. A suitable augmented Lyapunov-Krasovskii functional (LKF) with triple integral term, which takes full use of the neuron activation function conditions and the information of time-delay in integral term, is constructed. Furthermore, by utilizing integral inequality proposed recently and the combining reciprocally convex method to estimate the derivative of the LKF, some less conservative robust passivity conditions are derived in terms of LMI. The superiority of presented approaches are demonstrated via two classic numerical examples.

        Ultimate capacity of welded box section columns with slender plate elements

        For an axially loaded box-shaped member, the width-to-thickness ratio of the plate elements preferably should not be greater than 40 for Q235 steel grades in accordance with the Chinese code GB50017- 2003. However, in practical engineering the plate width-to-thickness ratio is up to 120, much more than the limiting value. In this paper, a 3D nonlinear finite element model is developed that accounts for both geometrical imperfections and residual stresses and the ultimate capacity of welded built-up box columns,with larger width-to-thickness ratios of 60, 70, 80, and 100, is simulated. At the same time, the interaction buckling strength of these members is determined using the effective width method recommended in the Chinese code GB50018-2002, Eurocode 3 EN1993-1 and American standard ANSI/AISC 360-10 and the direct strength method developed in recent years. The studies show that the finite element model proposed can simulate the behavior of nonlinear buckling of axially loaded box-shaped members very well. The width-tothickness ratio of the plate elements in welded box section columns can be enlarged up to 100 for Q235 steel grades. Good agreements are observed between the results obtained from the FEM and direct strength method. The modified direct strength method provides a better estimation of the column strength compared to the direct strength method over the full range of plate width-to-thickness ratio. The Chinese code and Eurocode 3 are overly conservative prediction of column capacity while the American standard provides a better prediction and is slightly conservative for b/t = 60. Therefore, it is suggested that the modified direct strength method should be adopted when revising the Chinese code.

      • Cytokeratin 15 is an Effective Indicator for Progression and Malignancy of Esophageal Squamous Cell Carcinomas

        Purpose: To compare the expression level of CK 15 in normal esophageal and esophageal squamous-cell carcinoma (ESCC) tissues and analyse possible functions of CK15 in occurrence and development. Materials and Methods: Immunohistochemistry was used to compare CK14, CK15 and proliferating cell nuclear antigen (PCNA) expression levels in ESCCs. Expression level of CK15 was also assessed by Western blotting. In addition, levels of CK15, cytokeratin 19 fragment antigen 21-1 (CYFRA21-1) and PCNA were detected in serum by enzymelinked immunosorbent assay (ELISA) and chemiluminescence methods. Relationships between clinicopathological parameters and CK14 and CK15 expression were then analyzed. Results: According to immunohistochemistry, in esophageal and intraepithelial neoplasia (SIN) tissues, the expression of CK14, CK15 and PCNA localized to basal layer of the epithelium. CK14 and CK15 levels were higher in normal esophageal squamous epithelial tissue than in SIN and ESCC, and greater in highly differentiated than poorly differentiated carcinoma tissue. By Western blotting, we found more pronounced expression of CK15 in normal esophageal tissue, compared with carcinoma tissue. The specificity of changed CK15 and CYFRA21-1 expression was respectively 90.0% and 96.7% in serum of ESCC patients. Joint detection could improve the sensitivity of esophageal carcinoma diagnosis. Relationships between CK14, CK15 expression and clinical parameters were not statistically significant (P>0.05). Postoperative survival in patients of CK14, CK15 positive expression was longer than with negative expression ($x^2=4.35$, P=0.037; $x^2=9.852$, P=0.002). Conclusions: CK15 expression decreased in esophageal squamous cell carcinoma tissue and serum of esophageal squamous carcinoma patients. We infer that CK15 may play an important role for the occurrence and development of esophageal squamous-cell carcinoma. In the future, CK15 may be used for the diagnosis, treatment and prognostic evaluation of esophageal squamous-cell carcinoma.

      • Prognostic Significance of GSTP1, XRCC1 and XRCC3 Polymorphisms in Non-small Cell Lung Cancer Patients

        Aim: Individual differences in chemosensitivity and clinical outcome in non-small cell lung cancer (NSCLC) patients treatment with platinum-based chemotherapy may be due to genetic factors. Our study aimed to investigate the prognostic role of GSTP1, XRCC1 and XRCC3 in NSCLC patients treated with chemotherapy. Methods: A total of 460 cases were consecutively selected from The Affiliated Hospital of Nantong University between Jan. 2003 to Nov. 2006, and all were followed-up until Nov. 2011. Genotyping of GSTP1 Ile105Val, XRCC1 Arg194Trp, XRCC1 Arg399Gln and XRCC3 Thr241Met was conducted by duplex polymerase-chain-reaction with confronting-two-pair primer methods. Results: Patients with GSTP Val/Val exhibited a shorter survival time, and had a 1.89 fold greater risk of death than did those with the IIe/IIe genotype. For XRCC1 Arg194Trp, the variant genotype Trp/Trp was significantly associated with a decreased risk of death from NSCLC when compared with the Arg/Arg. Individuals carrying XRCC1 399Gln/Gln genotype had a longer survival time, with a lowered risk of death from NSCLC. Conclusion: This study indicated that GSTP1 Ile105Val, XRCC1 Arg194Trp and XRCC1Arg399Gln genes have a role in modifying the effect of platinum-based chemotherapy for NSCLC patients in a Chinese population. Our findings provide information for therapeutic decisions for individualized therapy in NSCLC cases.
