        Transcriptome analysis of differential gene expression in Dichomitus squalens during interspecific mycelial interactions and the potential link with laccase induction

        Interspecific mycelial interactions between white rot fungi are always accompanied by an increased production of laccase. In this study, the potential of the white rot fungus Dichomitus squalens to enhance laccase production during interactions with two other white rot fungi, Trametes versicolor or Pleurotus ostreatus, was assessed. To probe the mechanism of laccase induction and the role that laccase plays during combative interaction, we analyzed the differential gene expression profile of the laccase induction response to stressful conditions during fungal interaction. We further confirmed the expression patterns of 16 selected genes by qRT-PCR analysis. We noted that many differentially expressed genes (DEGs) encoded proteins that were involved in xenobiotic detoxification and reactive oxygen species (ROS) generation or reduction, including aldo/keto reductase, glutathione S-transferases, cytochrome P450 enzymes, alcohol oxidases and dehydrogenase, manganese peroxidase and laccase. Furthermore, many DEG-encoded proteins were involved in antagonistic mechanisms of nutrient acquisition and antifungal properties, including glycoside hydrolase, glucanase, chitinase and terpenoid synthases. DEG analyses effectively revealed that laccase induction was likely caused by protective responses to oxidative stress and nutrient competition during interspecific fungal interactions.

        Heat treatments of peptides from oyster (Crassostrea gigas) and the impact on their digestibility and angiotensin I converting enzyme inhibitory activity

        The changes of protein digestibility, the peptidesin the digestive juice and angiotensin I converting enzyme(ACE) inhibitory activity after heating of oysters wereinvestigated. The digestibility of raw oysters was 71.1%,and that of oysters heated at 100 C was 67.9%. A total of169 and 370 peptides were identified from the digestion ofraw oysters and heated oysters, respectively. According toUPLC-Q-TOF-MS spectra, the peptides with a molecularweight below 2000 Da accounted for 87.6% of the totalpeptides of raw oysters and 94% of heated oysters. Testingthe ACE inhibitory activity in vitro, the IC50 values of rawoyster and cooked oyster were 6.77 lg/mL and 3.34 lg/mL, respectively. Taken together, the results showed thatheated oysters could produce more active peptides andprovide ACE inhibitory activity.

        Flyback AC-DC Converter with Low THD Based on Primary-Side Control

        A single-stage flyback LED AC-DC converter based on primary-side control under constant current mode is proposed in this study. The proposed converter features low total harmonic distortion (THD) and high power factor (PF). It also consists of a zero-crossing distortion compensation circuit and a variable duty ratio control compensation circuit to deal with the line current distortions caused by fixed duty ratio control. The system model and layout are built in Simplis and Cadence, respectively. The feasibility and performance of the proposed circuit is verified by designing and fabricating an IC controller in the HHNEC $0.35{\mu}m$ 5 V/40 V HVCMOS process. Experimental results show that the PF can reach a level in the range of 0.985-0.9965. Moreover, the average THD of the entire system is approximately 10%, with the minimum being 6.305%, as the input line voltage changes from 85 VAC to 265 VAC.

        Mycobacterium tuberculosis PE_PGRS45 (Rv2615c) Promotes Recombinant Mycobacteria Intracellular Survival via Regulation of Innate Immunity, and Inhibition of Cell Apoptosis

        Tuberculosis (TB), a bacterial infectious disease caused by Mycobacterium tuberculosis (M. tuberculosis), is a significant global public health problem. Mycobacterium tuberculosis expresses a unique family of PE_PGRS proteins that have been implicated in pathogenesis. Despite numerous studies, the functions of most PE_PGRS proteins in the pathogenesis of mycobacterium infections remain unclear. PE_PGRS45 (Rv2615c) is only found in pathogenic mycobacteria. In this study, we successfully constructed a recombinant Mycobacterium smegmatis (M. smegmatis) strain which heterologously expresses the PE_PGRS45 protein. We found that overexpression of this cell wall-associated protein enhanced bacterial viability under stress in vitro and cell survival in macrophages. MS_PE_PGRS45 decreased the secretion of pro-inflammatory cytokines such as IL-1β, IL-6, IL-12p40, and TNF-α. We also found that MS_PE_PGRS45 increased the expression of the anti-inflammatory cytokine IL-10 and altered macrophage-mediated immune responses. Furthermore, PE_PGRS45 enhanced the survival rate of M. smegmatis in macrophages by inhibiting cell apoptosis. Collectively, our findings show that PE_PGRS45 is a virulent factor actively involved in the interaction with the host macrophage.
