        Biochemical Properties and Physiological Roles of NADP-Dependent Malic Enzyme in Escherichia coli

        Malic enzymes catalyze the reversible oxidative decarboxylation of L-malate using NAD(P)^+ as a cofactor. NADP-dependent malic enzyme (MaeB) from Escherichia coli MG1655 was expressed and purified as a fusion protein. The molecular weight of MaeB was about 83 kDa, as determined by SDS-PAGE. The recombinant MaeB showed a maximum activity at pH 7.8 and 46°C. MaeB activity was dependent on the presence of Mn^(2+) but was strongly inhibited by Zn^(2+). In order to understand the physiological roles, recombinant E. coli strains (icdNADP/ΔmaeB and icdNAD/ΔmaeB) containing NADP-dependent isocitrate dehydrogenase (IDH), or engineered NAD-dependent IDH with the deletion of the maeB gene, were constructed using homologous recombination. During growth on acetate, icd^(NAD)/ΔmaeB grew poorly, having a growth rate only 60% that of the wild-type strain (icd^(NADP)). Furthermore, icd^(NAD)P/ΔmaeB exhibited a 2-fold greater adaptability to acetate than icdNAD/ΔmaeB, which may be explained by more NADPH production for biosynthesis in icd^(NAD)P/ΔmaeB due to its NADP-dependent IDH. These results indicated that MaeB was important for NADPH production for bacterial growth on acetate. We also observed that MaeB activity was significantly enhanced (7.83-fold) in icd^(NAD), which was about 3-fold higher than that in icd^(NADP), when switching from glucose to acetate. The marked increase of MaeB activity was probably induced by the shortage of NADPH in icd^(NAD). Evidently, MaeB contributed to the NADPH generation needed for bacterial growth on two carbon compounds.

        Glasgow prognostic score and combined positive score for locally advanced rectal cancer

        Purpose: This study was performed to investigate the association of Glasgow prognostic score (GPS), combined positive score (CPS), and clinicopathological characteristics of locally advanced rectal cancer. Methods: Between February 2012 and February 2018, 103 patients with locally advanced rectal cancer treated by neoadjuvant chemoradiotherapy and total mesorectal excision (TME) were retrospectively evaluated. Results: According to the classification of the GPS, 85 (82.5%), 13 (12.6%), and 5 patients (4.9%) were classified as a score of 0, 1, and 2, respectively. Patients were classified into the GPS-low group (GPS of 0, n = 85) and GPS-high group (GPS of 1 or 2, n = 18) with an area under the curve of 0.582 for overall survival (OS). The mean programmed death-ligand 1 (PD-L1) CPS of the whole group was 2.24 (range, 0–70). The PD-L1 CPS of the GPS-high group was higher than the GPS-low group (P < 0.001). Multivariate analysis by Cox proportional hazards model indicated that GPS was associated with OS and disease- free survival (DFS). Furthermore, PD-L1 CPS was associated with DFS (hazard ratio, 1.050; 95% confidence interval, 1.017– 1.083; P = 0.003). Conclusion: Elevated GPS was related to the PD-L1 CPS. GPS and PD-L1 CPS were associated with the prognosis of locally advanced rectal cancer treated with neoadjuvant chemoradiotherapy followed by TME.

        Alterations of oral microbiota in Chinese children with viral encephalitis and/or viral meningitis

        The role of oral microbiota in viral encephalitis and/or viral meningitis (VEVM) remains unclear. In this hospital-based, frequency-matched study, children with clinically diagnosed VEVM (n = 68) and those with other diseases (controls, n = 68) were recruited. Their oral swab samples were collected and the oral microbiota was profiled using 16S rRNA gene sequencing. The oral microbiota of children with VEVM exhibited different beta diversity metrics (unweighted UniFrac distance: P < 0.001, R2 = 0.025, Bray-curtis dissimilarity: P = 0.045, R2 = 0.011, and Jaccard dissimilarity: P < 0.001, R2 = 0.017) and higher relative abundances of taxa identified by Linear discriminant analysis (LDA) with effect size (Enterococcus, Pedobacter, Massilia, Prevotella_9, Psychrobacter, Butyricimonas, Bradyrhizobium, etc., LDA scores > 2.0) when compared with the control group. The higher pathway abundance of steroid hormone biosynthesis predicted by oral microbiota was suggested to be linked to VEVM (q = 0.020). Further, a model based on oral microbial traits showed good predictive performance for VEVM with an area under the receiver operating characteristic curve of 0.920 (95% confidence interval: 0.834–1.000). Similar results were also obtained between children with etiologically diagnosed VEVM (n = 43) and controls (n = 68). Our preliminary study identified VEVM-specific oral microbial traits among children, which can be effective in the diagnosis of VEVM.

        Optimization of ultrasonic assisted membrane strategy for saponins from Gynostemma Pentaphyllum with response surface methodology

        Gynostemma pentaphyllum saponin has a variety of biological properties. Classic separation methods of saponin, such as resin absorption and preparative chromatography are limited by environmental pollution and high cost. In the study, ultrasonic assisted membrane separation was firstly used to purify saponin from Gynostemma pentaphyllum. Total proteins, polysaccharides, saponin, gypenoside A and rutin were selected as indexes to optimize the pretreatment and purification parameters by response surface methodology. The fitted models were significant (p < 0.05) and the optimal conditions were: (1) removing protein and polysaccharides by MWCO 10,000 Da, ultrasonic power 400 W and pH 7.8; (2) separation flavonoids from saponin by MWCO 1000 Da, ultrasonic power 300 W and pH 7.9. The difficulty in separating saponin from flavonoids was solved by releasing flavonoids from micelles with ultrasonic assisted membrane method. The saponin content in Gynostemma pentaphyllum extracts reached 82.81%, which was more than four times of that obtained with resin adsorption method. The protective effect of saponins on SH-SY5Y cells injury induced by H2O2 was better than that of Gynostemma pentaphyllum extracts. The study suggested that ultrasonic assisted membrane method would be widely applied in the preparation of food materials.
