        Differential Expression Profiles of Alternaria alternate Genes in Response to Carbonyl Sulfide Fumigation

        Carbonyl sulfide (COS) is a new fumigant used in phytosanitary treatments. It was developed as a potential alternative to methyl bromide, which is being phased out because of its ozone-depletion properties. To understand the molecular and cellular mechanisms occurring in fungal pathogens in response to COS fumigation, we cloned 510 cDNA fragments of Alternaria alternata (Fr.) Keissler genes that are differentially expressed; these genes were cloned using suppression subtractive hybridization. Changes in the levels of transcripts of 79 fragments were confirmed by microarray analysis and qRT-PCR. Further homology search revealed that they are highly homologous to 41 genes of other fungi, which were related to general metabolism, growth and division, defense, cellular transport, and signal transduction. These results provide an overview of differential expression profiles of A. alternata genes following COS treatment and some new clues about the mechanism of COS fungitoxicity.

        A Random Deflected Subgradient Algorithm for Energy-Efficient Real-time Multicast in Wireless Networks

        In this work, we consider the optimization problem of minimizing energy consumption for real-time multicast over wireless multi-hop networks. Previously, a distributed primal-dual subgradient algorithm was used for finding a solution to the optimization problem. However, the traditional subgradient algorithms have drawbacks in terms of i) sensitivity to iteration parameters; ii) need for saving previous iteration results for computing the optimization results at the current iteration. To overcome these drawbacks, using a joint network coding and scheduling optimization framework, we propose a novel distributed primal-dual Random Deflected Subgradient (RDS) algorithm for solving the optimization problem. Furthermore, we derive the corresponding recursive formulas for the proposed RDS algorithm, which are useful for practical applications. In comparison with the traditional subgradient algorithms, the illustrated performance results show that the proposed RDS algorithm can achieve an improved optimal solution. Moreover, the proposed algorithm is stable and robust against the choice of parameter values used in the algorithm.

        Dynamic Magnetic Field Measurement in the Air Gap of Magnetic Bearings Based on FBG-GMM Sensor

        Magnetic field in magnetic bearings is the physical medium to realize magnetic levitation, the distributionof the magnetic field determines the operating performance of magnetic bearings. In this paper, a thin-sliceFiber Bragg Grating-Giant Magnetostrictive Material magnetic sensor used for the air gap of magneticbearings was proposed and tested in the condition of dynamic magnetic field. The static property of thesensor was calibrated and a polynomial curve was fitted to describe the performance of the sensor. Measurement of dynamic magnetic field with different frequencies in magnetic bearings was implemented. Comparing with the finite element simulations, the results showed the DC component of the magneticfield was detected by the sensor and error was less than 5.87%

      • Research on Business Operating Model of New Energy Battery Electric Vehicles Used as Urban Logistics Cars

        Nowadays, because of air pollution, the lack of energy and the rising demand of motor vehicles, china’s government has published much policy to encourage the development of the new energy car industry. However, it doesn’t work as expected for the lack of effective business model of the market promotion of the new energy car industry. The development of urban public environmental protection and logistics industry call a stable demand of urban logistics cars; and the environment pollution and traffic problem provide a wide space for the market of battery electric vehicles used as urban logistics cars. This paper analyzes current circumstances and problems of the market of new energy cars, provides a business operating model of battery electric vehicles, which is setting up an individual operating company, and states different operating models with different subject of operation. Based on all these conclusions, the paper discusses the implementation condition, process, economic benefit and relative policy advice of business promotion model of the individual operating company with a topic of battery electric vehicles used as urban logistics cars, in order to commercialize them from the demand of the market.

        Experimental Study of Adaptive Sliding Mode Control for Vibration of a Flexible Rectangular Plate

        This paper aims to address the intelligent active vibration control problem of a flexible rectangular plate vibration involving parameter variation and external disturbance. An adaptive sliding mode (ASM) MIMO control strategy and smart piezoelectric materials are proposed as a solution, where the controller design can deal with problems of an external disturbance and parametric uncertainty in system. Compared with the current 'classical' control design, the proposed ASM MIMO control strategy design has two advantages. First, unlike existing classical control algorithms, where only low intelligence of the vibration control system is achieved, this paper shows that high intelligent of the vibration control system can be realized by the ASM MIMO control strategy and smart piezoelectric materials. Second, the system performance is improved due to two additional terms obtained in the active vibration control system. Detailed design principle and rigorous stability analysis are provided. Finally, experiments and simulations were used to verify the effectiveness of the proposed strategy using a hardware prototype based on NI instruments, a MATLAB/SIMULINK platform, and smart piezoelectric materials.

        Alterations of oral microbiota in Chinese children with viral encephalitis and/or viral meningitis

        The role of oral microbiota in viral encephalitis and/or viral meningitis (VEVM) remains unclear. In this hospital-based, frequency-matched study, children with clinically diagnosed VEVM (n = 68) and those with other diseases (controls, n = 68) were recruited. Their oral swab samples were collected and the oral microbiota was profiled using 16S rRNA gene sequencing. The oral microbiota of children with VEVM exhibited different beta diversity metrics (unweighted UniFrac distance: P < 0.001, R2 = 0.025, Bray-curtis dissimilarity: P = 0.045, R2 = 0.011, and Jaccard dissimilarity: P < 0.001, R2 = 0.017) and higher relative abundances of taxa identified by Linear discriminant analysis (LDA) with effect size (Enterococcus, Pedobacter, Massilia, Prevotella_9, Psychrobacter, Butyricimonas, Bradyrhizobium, etc., LDA scores > 2.0) when compared with the control group. The higher pathway abundance of steroid hormone biosynthesis predicted by oral microbiota was suggested to be linked to VEVM (q = 0.020). Further, a model based on oral microbial traits showed good predictive performance for VEVM with an area under the receiver operating characteristic curve of 0.920 (95% confidence interval: 0.834–1.000). Similar results were also obtained between children with etiologically diagnosed VEVM (n = 43) and controls (n = 68). Our preliminary study identified VEVM-specific oral microbial traits among children, which can be effective in the diagnosis of VEVM.

        A hybrid method for dynamic stiffness identification of bearing joint of high speed spindles

        Bearing joint dynamic parameter identification is crucial in modeling the high speed spindles for machining centers used to predict the stability and natural frequencies of high speed spindles. In this paper, a hybrid method is proposed to identify the dynamic stiffness of bearing joint for the high speed spindles. The hybrid method refers to the analytical approach and experimental method. The support stiffness of spindle shaft can be obtained by adopting receptance coupling substructure analysis method, which consists of series connected bearing and joint stiffness. The bearing stiffness is calculated based on the Hertz contact theory. According to the proposed series stiffness equation, the stiffness of bearing joint can be separated from the composite stiffness. Then, one can obtain the bearing joint stiffness fitting formulas and its variation law under different preload. An experimental set-up with variable preload spindle is developed and the experiment is provided for the validation of presented bearing joint stiffness identification method. The results show that the bearing joint significantly cuts down the support stiffness of the spindles, which can seriously affects the dynamic characteristic of the high speed spindles.

        Dynamics and Control of a Flexible-Link Flexible-Joint Space Robot with Joint Friction

        Multi-DOF flexible space robots play a significant role in the on-orbit service. The flexibility of the robots is mainly caused by the flexible links and some flexible drive elements in joints. The vibration generated by the component flexibility can affect the accuracy of control. In this paper, the dynamics and control issues of flexible-link flexible-joint (FLFJ) space robots considering joint friction are studied. Firstly, the Spong’s model is employed to depict flexible joints, and the Coulomb friction model is adopted to describe joint friction. Then based on the single direction recursive construction method and velocity variation principle, dynamic equations of the system are derived. Secondly, a trajectory of joint motion is given and a trajectory tracking controller with friction compensation is designed with the computed torque method. Finally, several numerical simulations are carried out to verify the accuracy of the dynamic model and illustrate the effect of joint friction and joint flexibility on the dynamic characteristics of space robots. Besides, simulation results suggest that the controller designed in this paper could effectively achieve the trajectory tracking control for the 6-DOF FLFJ space robot with joint friction.

        Identification and Characterization of Ectoine Biosynthesis Genes and Heterologous Expression of the ectABC Gene Cluster from Halomonas sp. QHL1, a Moderately Halophilic Bacterium Isolated from Qinghai Lake

        The moderately halophilic bacterium Halomonas sp. QHL1was identified as a member of the genus Halomonas by 16SrRNA gene sequencing. HPLC analysis showed that strainQHL1 synthesizes ectoine in its cytoplasm. The genes involvedin the ectoine biosynthesis pathway were identifiedon the chromosome in the order ectABC. Subsequently, theectB gene from this strain was amplified by PCR, and theentire ectABC gene cluster (3,580 bp) was cloned using genomewalking. Analysis showed that the ectA (579 bp), ectB(1269 bp), and ectC (390 bp) genes were organized in a singletranscriptional unit and were predicted to encode threepeptides of 21.2 kDa, 46.4 kDa, and 14.7 kDa, respectively. Two putative promoters, a δ70-dependent promoter and aδ38-controlled promoter, as well as several conserved motifswith unknown function were identified. Individual ectA, ectB,and ectC genes, and the entire ectABC gene cluster were insertedinto the expression plasmid pET-28a(+) to generatethe recombinant plasmids pET-28a(+)-ectA, pET-28a(+)-ectB, pET-28a(+)-ectC and pET-28a(+)-ectABC, respectively. Heterologous expression of these proteins in Escherichiacoli BL21 (DE3) was confirmed by SDS-PAGE. The recombinantE. coli strain BL21 (pET-28a (+)-ectABC) displayeda higher salt tolerance than native E. coli cells but producedfar less ectoine than the wild-type QHL1 strain.
