<P>AIMS: Endonuclease G (EndoG) is a mitochondrial protein that plays a role in DNA fragmentation during apoptosis. In addition, EndoG plays a role in cell proliferation and survival. It may be important to identify EndoG protein expression to predict its function in human cancers. The aim of this study was to explore whether alteration of EndoG expression might be a characteristic of colorectal or gastric carcinoma. METHODS: We investigated EndoG protein expression in 103 colorectal and 60 gastric carcinoma tissues by immunohistochemistry using a tissue microarray approach. RESULTS: Expression of EndoG was detected in 72 (70%) of the colorectal carcinomas and 41 (68%) of the gastric carcinomas in cytoplasm. By contrast, normal mucosal cells of both stomach and colon tissues showed no or very weak expression of EndoG. There was no significant association of EndoG expression with clinocopathological characteristics, including invasion, metastasis and stage. CONCLUSION: Our data indicate that EndoG inactivation by loss of expression may not occur in colorectal and gastric cancers. Rather, increased expression of EndoG in colorectal and gastric cancer cells compared to their normal mucosal epithelial counterparts suggests that neo-expression of EndoG may play a role in both colorectal and gastric tumorigenesis.</P>
Epigenetic inactivation of HOXA5 and MSH2 gene in clear cell renal cell carcinoma

<P>The high-throughput method using microarray is an easy and fast way to analyze the methylation status of hundreds of preselected genes and to screen them for signatures in methylation. The aim of our study is to detect hypermethylated genes and to analyze the association between methylation status and clinicopathological parameters of clear cell renal cell carcinoma. The genetic substrate included 62 cancer tissues and 62 matched adjacent normal kidney tissues. We adapted the GoldenGate genotyping assay to determine the methylation state of 1505 specific CpG sites in 807 genes. We identified two genes (HOXA5 and MSH2) with &bgr;-value differences of more than 0.3 between cancer and normal tissues. The high methylation group in HOXA5 had high Fuhrman's nuclear grade (<I>P</I>= 0.041). Other data in HOXA5 and MSH2 were not significant with methylation status (<I>P</I> > 0.05). Survival curve of the high methylation group in HOXA5 was slightly lower than that of the low methylation group. However, the statistical significances of overall survival in HOXA5 and MSH2 were low (<I>P</I> > 0.05). We report the hypermethylation of two genes in clear cell renal cell carcinoma. The data we obtained could provide the basis for a diagnostic test pathological assessment, or prognosis in clear cell renal cell carcinoma.</P>
Development of 3D statistical mandible models for cephalometric measurements

Purpose: The aim of this study was to provide sex-matched three-dimensional (3D) statistical shape models of the mandible, which would provide cephalometric parameters for 3D treatment planning and cephalometric measurements in orthognathic surgery. Materials and Methods: The subjects used to create the 3D shape models of the mandible included 23 males and 23 females. The mandibles were segmented semi-automatically from 3D facial CT images. Each individual mandible shape was reconstructed as a 3D surface model, which was parameterized to establish correspondence between different individual surfaces. The principal component analysis (PCA) applied to all mandible shapes produced a mean model and characteristic models of variation. The cephalometric parameters were measured directly from the mean models to evaluate the 3D shape models. The means of the measured parameters were compared with those from other conventional studies. The male and female 3D statistical mean models were developed from 23 individual mandibles, respectively. Results: The male and female characteristic shapes of variation produced by PCA showed a large variability included in the individual mandibles. The cephalometric measurements from the developed models were very close to those from some conventional studies. Conclusion: We described the construction of 3D mandibular shape models and presented the application of the 3D mandibular template in cephalometric measurements. Optimal reference models determined from variations produced by PCA could be used for craniofacial patients with various types of skeletal shape.
Influence of Addition of Organoclays on Morphologies in Nylon 6/LLDPE Blends

<P>Summary: Polymer blend nanocomposites containing linear low density polyethylene (LLDPE), nylon 6 and organoclays were prepared by melt mixing, and their morphologies and structures were examined with a field emission scanning electron microscope (FE-SEM) and an X-ray diffractometer (XRD). The size of phase-separated domains decreased considerably with increasing content of organoclay. The d-spacing of organoclay in the nanocomposites was increased from about 18.6 to over 28 Å. This effect was highly dependent on nylon 6 contents because nylon 6 is more polar and shows higher affinity to the organoclays compared to LLDPE.</P><P> <img src='wiley_img/10221352-2005-206-8-MACP200400526-gra001.gif' alt='wiley_img/10221352-2005-206-8-MACP200400526-gra001'> Graphic </P>

Stroke occurs when local thrombosis, embolic particle or the rupture of blood vessele interrupts the blood floe to the brain. -estradiol 17-valerate has been reported to exert neuroprotective effects when administered before an ischemic insult. Recently, the pathophysiology of cerebral ischemia has been studied extensively in rat with various methods. In the present study, we investigates whether -estrodiol 17-valerate can protect against brain injury. RNA sample were extracted from the hippocampus of female rat, reverse-transcription in the presence of [32p] dATP. Differential gene express-ion profiles were revealed (Bone morphogenetic protein type 1A receptor, Protein disulphide isomerase, Leukemia inhibitor factor receptor, cytochrome bc- 1 complex-x core P, thiol-specific antioxidant protein). RT-PCR was used to validate the relative expression pattern obtained by the cDNA array. The precise relationship between the early expression of recovery genes and stroke is a matter of luther investigation. Preparation of acrylic copolymers and crosslinking agents and properties as a film

<P>Copolymers of butyl acrylate (BA)-methyl methacrylate (MMA)-acrylic acid (AA) and intraparticle crosslinking agents containing N-methylol acrylamide (NMA) and ethylene glycol dimethacrylate (EGDMA) were prepared by emulsion copolymerization. After that, films were prepared from the mixture of copolymers and the interparticle crosslinking agents. The interparticle crosslinking agents were prepared from hexamethylene diisocyanate and aziridine ethanol. Mixtures of the copolymer and the interparticle crosslinking agent were cast to films and crosslinked in a convection oven. The effects of the contents of the intra/interparticle crosslinking agents were also evaluated. By increasing the contents of EGDMA, roughness of the films was increased because of the effects of EGDMA acting as an intraparticle crosslinking agent. By increasing the contents of the interparticle crosslinking agent, roughness was also increased by the reaction between the copolymers and interparticle crosslinking agent. Tensile strength and water and chemical resistance of the film were increased, whereas elongation of film was decreased by increasing the contents of interparticle crosslinking agents. © 2009 Wiley Periodicals, Inc. We report a case of primary biliary cirrhosis (PBC) that occurred in a 24-month-old male C57BL/6 mouse infected with Helicobacter pylori (H. pylori). Microscopically, the portal tract in the liver showed nonsuppurative destructive cholangitis with variable cytologic distortion of the epithelial cells and peribiliary lymphoplasmacytic infiltration. Immunohistochemistry using α-smooth muscle actin demonstrated fibrous bands associating with the wall of vasculature. The level of serum antivacuolating toxin IgG in this mouse showed the highest value (optical density=2.1470) of the H. pylori-infected group (n=13) (optical density=1.7168±0.1759, mean±SD). Spontaneously developed PBC-like lesions in C57BL/6 mice have been reported by several authors. However, this case strikingly resembles human PBC with its characterized histological features. Therefore, we propose that the increase in vacuolating toxin caused by H. pylori infection may be related to the development of PBC by molecular mimicry.
